Computational design of superstable proteins through maximized hydrogen bonding.

Hydrogen bonds are fundamental chemical interactions that stabilize protein structures, particularly in β sheets, enabling resistance to mechanical stress and environmental extremes. Here, inspired by natural mechanostable proteins with shearing hydrogen bonds, such as titin and silk fibroin, we de novo designed superstable proteins by maximizing hydrogen-bond networks within force-bearing β strands. Using a computational framework combining artificial intelligence-guided structure and sequence design with all-atom molecular dynamics MD simulations, we systematically expanded protein architect

Novel Bioinspired Nerve Scaffold with High Synchrony between Biodegradation and Nerve Regeneration for Repair of Peripheral Nerve Injury.

The morphological structure reconstruction and functional recovery of long-distance peripheral nerve injury (PNI) are global medical challenges. Biodegradable nerve scaffolds that provide mechanical support for the growth and extension of neurites are a desired way to repair long-distance PNI. However, the synchrony of scaffold degradation and nerve regeneration is still challenging. Here, a novel bioinspired multichannel nerve guide conduit (MNGC) with topographical cues based on silk fibroin and ε-polylysine modification was constructed. This conduit (SF(A) + PLL MNGC) exhibited sufficient m

Tubular TPU/SF nanofibers covered with chitosan-based hydrogels as small-diameter vascular grafts with enhanced mechanical properties.

Native grafts such as internal mammary artery and saphenous vein are the main choice for coronary artery bypass graft. However, due to the limitations associated with their availability and rapid failure caused by hyperplasia, small diameter tissue-engineered vascular grafts (TEVGs) with sufficient post-implantation patency are urgently demanded as artificial alternatives. In our previous work, we innovatively fabricated a bilayer vascular graft providing appropriate structural and biological properties using electrospinning and freeze-drying methods. It was proved that the mechanical properti

A novel biomimetic strategy for mimicking amelogenesis to repair enamel.

According to the principle of minimal invasiveness in modern dentistry, biomimetic remineralization therapy constitutes a significant strategy for the prevention and treatment of early enamel caries. Based on the three "key events" of amelogenesis in vivo, silk fibroin (SF) combined with carboxymethyl chitosan (CMC) successfully formed an SF/CMC composite, and amorphous calcium phosphate (ACP) was then used to form an SF/CMC-ACP nanocomposite with remineralization properties. In our study, SF was used as a template protein for biomimetic amelogenin, ACP was stabilized with CMC and the reminera

Osteogenic surgical sutures for tendon traction and fixation: A model of achilles tendon sleeve avulsion.

Currently, the repair of Achilles tendon sleeve avulsion is a challenge due to their limited research and particularly difficult treatment. In tendon repair surgery, the construction of bone tunnels is required for the suspensory fixation of ruptured tendon by sutures. However, due to the biologically inert of commonly used tendon sutures, postoperative fixation instability, bone tunnel enlargement, and even tendon reconstruction failure can easily occur under stressful conditions. In this study, core-spun nanoyarns containing β-tricalcium phosphate (β-TCP) were prepared by electrospinning to

Silk-based multilayer coatings for anticoagulation and durability improvement in ECMO hollow fiber membranes.

In clinical extracorporeal membrane oxygenation (ECMO), prolonged blood residence time in oxygenators can lead to thrombus formation and complications. Heparin-based bioactive coatings are commonly used for anticoagulation, but prolonged shear stress during blood flow can cause heparin desorption, degradation, and inactivation, reducing durability and antithrombotic efficacy. In this study, we developed a silk fibroin (SF)-based multilayer coating to enhance both mechanical stability and anticoagulant performance. A SF foundation layer was first fabricated on hollow-fiber membranes using layer

Preparation and evaluation of antibacterial wound dressing based on vancomycin-loaded silk/dialdehyde starch nanoparticles.

One of the main reasons infected wounds go untreated is that antibiotic-resistant bacteria mainly cause infection. Vancomycin is an antibiotic used against Gram-positive bacteria, such as MRSA, but it has limited intravenous use due to its toxicity. This study describes using a local drug delivery approach at the wound site. The aim is to prepare a silk dressing containing dialdehyde starch nanoparticles loaded with vancomycin that can cure infection through the controlled release of antibiotics. First, the starch was oxidized by sodium periodate solution and converted to dialdehyde starch. Di

Short-Term Preclinical Application of Functional Human Induced Pluripotent Stem Cell-Derived Airway Epithelial Patches.

Airway pathologies including cancer, trauma, and stenosis lack effective treatments, meanwhile airway transplantation and available tissue engineering approaches fail due to epithelial dysfunction. Autologous progenitors do not meet the clinical need for regeneration due to their insufficient expansion and differentiation, for which human induced pluripotent stem cells (hiPSCs) are promising alternatives. Airway epithelial patches are engineered by differentiating hiPSC-derived airway progenitors into physiological proportions of ciliated (73.9 ± 5.5%) and goblet (2.1 ± 1.4%) cells on a silk f

Protocol for extrusion 3D bioprinting of human-scale cardiac patches using cardiac spheroid-laden bioinks.

Extrusion-based bioprinting is a promising approach for developing novel clinical therapies for heart failure patients. Here, we present a protocol for bioprinting personalized human-scale cardiac patches incorporating human cardiac spheroids in a 1% silk fibroin/4% alginate/8% gelatin hydrogel. We describe steps for extrusion 3D bioprinting using a temperature-controlled hydrogel bioink. We also detail adjusting print parameters for optimal results. Although prepared for a specific experimental setup, the techniques described can be applied to other temperature-controlled biomaterials or extr

Development of a decellularized human amniotic membrane-based electrospun vascular graft capable of rapid remodeling for small-diameter vascular applications.

The performance of small-diameter vascular grafts adapted to vascular replacement is commonly hindered by stenosis. To address this issue, a graft featuring rapid remodeling with degradation is warranted. In this work, a 1.8-mm-diameter graft was constructed by fabricating a decellularized human amniotic membrane (HAM) with polycaprolactone (PCL)/silk fibroin (SF) around it through electrospinning, namely, an HPS graft, and applied in a rat aortic grafting model for comparison to a decellularized porcine small intestinal submucosa (SIS)-integrated PCL/SF (SPS) graft and an autologous aorta. Clinically Oriented Oral Environment-Triggered Underwater Adhesives for Root Caries Treatment through Dentinal Tubule Occlusion and Remineralization.

The application of silk fibroin (SF) hydrogels is often limited by their brittleness; on the other hand, increasing ductility can lead to insufficient strength of the hydrogel. These drawbacks make it difficult to apply to treat root surface caries, which are continuously exudated by crevicular fluid and have special locations and shapes. Herein, we design an underwater adhesive hydrogel with a fluid-solid spontaneous transition triggered by water for root caries treatment. Guanidine hydrochloride (GH), amorphous calcium phosphate nanoparticles (ACP), and tannic acid (TA) are applied to coasse

Thermoplastic silk-plasticizer membranes for biodegradable artificial corneal endothelium.

Corneal endothelial dysfunction (CED) induces stromal edema through aqueous influx, disrupting collagen organization and causing corneal opacity, with chronic cases progressing to fibrosis. As a major cause of corneal blindness, CED currently requires corneal transplantation for treatment. However, the scarcity of human donors necessitates the urgent development of artificial corneal endothelial transplant materials. Previously reported non-biodegradable polymer materials suffer from inadequate adhesion and poor water barrier efficiency, with long-term implantation potentially causing foreign-

Psidium guajava L. phenolic compound-reinforced lamellar scaffold for tracheal tissue engineering.

The aim of this work was to develop a dense lamellar scaffold, as a biomimetic material with potential applications in the regeneration of tracheal tissue after surgical tumor resection. The scaffolds were produced by plastic compression technique, exploiting the use of total phenolic compounds (TPC) from Psidium guajava Linn as a potential cross-linking agent in a polymeric mixture based on collagen (COL), silk fibroin (SF), and polyethylene glycol 400 (PEG 400). Fourier transform infrared spectroscopy (FTIR) and differential scanning calorimetry (DSC) confirmed the chemical interactions betw

The effects of silk layer-by-layer surface modification on the mechanical and structural retention of extracellular matrix scaffolds.

Naturally derived extracellular matrix scaffolds can effectively promote tissue repair and regeneration due to their remarkable bioactivity. However, their rapid degradation leads to the decrease of mechanical retention and the failure of physical support in vivo which limit their applications. In this paper, we modified a classic extracellular matrix scaffold - small intestinal submucosa (SIS) - by a silk fibroin (SF) layer-by-layer (LbL) assembly to replace the existing chemical crosslinking methods for improving its mechanical and structural stability. Experimental results showed that the S

Bioengineered porous composite curcumin/silk scaffolds for cartilage regeneration.

Articular cartilage repair is a challenge due to its limited self-repair capacity. Cartilage tissue engineering supports to overcome following injuries or degenerative diseases. Herein, we fabricated the scaffold composed of curcumin and silk fibroin as an appropriate clinical replacement for defected cartilage. The scaffolds were designed to have adequate pore size and mechanical strength for cartilage repair. Cell proliferation, sulfated glycosaminoglycan (sGAG) content and mRNA expression analysis indicated that chondrocytes remained viable and showed its growth ability in the curcumin/silk

MFG-E8 glycoprotein in inflammatory tissue regeneration: molecular mechanisms, therapeutic potential and advanced polymeric biomaterial delivery strategies.

Milk fat globule epidermal growth factor 8 (MFG-E8) is a multifunctional glycoprotein that coordinates efferocytosis, inflammatory resolution, angiogenesis, and matrix remodeling during tissue repair. In ischemic and inflammatory microenvironments, endogenous MFG-E8 is functionally deficient due to proteolytic cleavage and oxidative inactivation, while free recombinant protein suffers from rapid clearance and poor targeting. Polymeric biomaterials, including nanoparticles, hydrogels, and extracellular vesicle carriers, offer protection, sustained release, and targeted delivery. This review sys

In situ fluid-to-solid converting injectable wet adhesive for comminuted fracture repair via water-activated bonding.

Comminuted fractures, characterized by multiple irregular bone fragments, present significant challenges for traditional fixation methods like plates and screws, often resulting in complex surgeries and delayed healing. Injectable hydrogel adhesives have been applied for comminuted fractures, but face challenges such as excessive fluidity before curing and curing processes that involve high temperatures or ionic release, which can result in material leakage, localized inflammation and reduced adhesion. To address these challenges, we developed a humidity-responsive hydrogel adhesive system, co
