Fusarium disease is an infectious condition that occurs in the cultivation of kuruma shrimp (Penaeus japonicus), primarily presenting as lesions in the gills. These lesions contain proteolytic enzymes of likely bacterial origin. This is the first study to demonstrate fungal-bacterial mixed infection in gill lesions of Fusarium-infected kuruma shrimp. Tenacibaculum spp. showing proteolytic activity were isolated from the lesions, whereas Fusarium spp. showed no such activity. Two Tenacibaculum strains were identified as Tenacibaculum mesophilum based on whole-genome sequencing (WGS) using MiSeq…

Accurate pathogen diagnosis is fundamental for effective crop disease management. In Kenya, a pathogen causing significant damage in potato farms was initially misidentified as Phytophthora infestans. This study aimed to correctly identify this pathogen and explore initial control measures using a polyphasic approach. The methodology integrated morphological observation, pathogenicity testing, and molecular analysis using ITS and EF1-α gene sequencing. The results confirmed the pathogen's identity as Fusarium equiseti, with morphological features consistent with this species and molecular sequ…

Fungal keratitis is a severe corneal infection most commonly caused by filamentous fungi. Even with prompt treatment with current antifungals, it often results in corneal perforation and blindness. In this report, we observe that the beta-adrenergic antagonist, propranolol, displays antifungal activity against Aspergillus and Fusarium corneal isolates in vitro and strikingly blocks disease establishment in a murine model of Aspergillus fumigatus keratitis. These findings suggest that beta-blockers have potential as a novel FK treatment.CLINICAL TRIALSThis study is registered with ClinicalTrial…

Fusarium wilt is a significant problem restricting the growth of the lily industry. However, Lilium regale Wilson is a valuable germplasm resource because of its Fusarium Wilt resistance. In this study, six endo-β-1,4-glucanase genes (Cel1-Cel6) in L. regale were identified among the differentially expressed genes reveled by omics data for L. regale infected Fusarium osysporum, which causes Fusarium wilt. Interestingly, the abundance of a few proteins and the expression of endo-β-1,4-glucanase genes, including LrCel1, decreased in L. regale roots infected with F. oxysporum. Thus, the function …

The wheat head blight disease caused by Fusarium graminearum is a major concern for food security and the health of both humans and animals. As a pathogenic microorganism, F. graminearum produces virulence factors during infection to increase pathogenicity, including various macromolecular and small molecular compounds. Among these virulence factors, secreted proteins and deoxynivalenol (DON) are important weapons for the expansion and colonization of F. graminearum. Besides the presence of virulence factors, sexual reproduction is also crucial for the infection process of F. graminearum and i…

Fusarium wilt, caused by (Fusarium udum Butler), is a significant threat to pigeonpea crops worldwide, leading to substantial yield losses. Traditional approaches like fungicides and resistant cultivars are not practical due to the persistent and evolving nature of the pathogen. Therefore, native biocontrol agents are considered to be more sustainable solution, as they adapt well to local soil and climatic conditions. In this study, five isolates of F. udum infecting pigeonpea were isolated from various cultivars and characterized morphologically and molecularly. The isolate from the ICP 8858 …

Although Fusarium spp. rarely cause infections in healthy people, they can cause fusariosis, particularly in neutropenic hematological malignancies, bone marrow transplant patients, and immunocompromised patients, such as those with acquired immune deficiency syndrome (AIDS), and rarely in solid organ transplant recipients. Here, we present a case of a liver transplant recipient with F. solani species complex (FSSC) infection treated with posaconazole. A 61-year-old man presented with multiple itchy, painful, palpable, irregular, subcutaneous nodules on the right leg and total dystrophic onych…

In this study, the optimal emulsifier for prothioconazole nanoemulsions was initially screened based on appearance, microscopic observation, mean droplet size and polydispersity index (PDI). In addition, the BoxBehnken design method is adopted, and the optimal formula is screened with an emulsification time, emulsifier content, and solvent content as a single factor. On this basis, the nanoemulsion meets FAO standards for various indicators. The contact angle of droplets on wheat leaves was significantly reduced. This nanoemulsion also showed good inhibitory activity against Fusarium graminear…

Nitrogen availability might play an essential role in plant diseases by enhancing fungal cell growth and influencing the expression of genes required for successful pathogenesis. Nitrogen availability could modulate secondary metabolic pathways as evidenced by the significant differential expression of several core genes involved in mycotoxin biosynthesis and genes encoding polyketide synthase/nonribosomal peptide synthetases, cytochrome P450 and carbohydrate-active enzymes in Fusarium sacchari, grown on different nitrogen sources. A combined analysis was carried out on the transcript and meta…

Due to an increasing number of patients at risk (i.e., those with a highly compromised immune system and/or receiving aggressive chemotherapy treatment), invasive fungal infections (IFI) are increasingly being reported and associated with high mortality rates. Aspergillus spp., particularly A. fumigatus, is the major cause of IFI caused by filamentous fungi around the world followed by Fusarium spp., however, other fungi are emerging as human pathogens. The aim of this study was to explore the epidemiology and prevalence of the non-Aspergillus and non-Fusarium filamentous fungi in human clinic…

Eumycetomas are chronic suppurative granulomas caused by fungi characterised by invasive tumefactive lesions, sinuses and discharging grains. Herein, we describe a case of pedal eumycetoma due to Fusarium solani sensu stricto in a person with diabetes mellitus. A 45-year-old gentleman presented with an insidious onset swelling over his right foot with nodules and discharging grains. He had received itraconazole and anti-tuberculous therapy elsewhere, without response. Re-evaluation included a biopsy which confirmed eumycetoma and newly diagnosed diabetes. Surgical excision followed by histopat…

Fungal denitrification is claimed to produce non-negligible amounts of N2 O in soils, but few tested species have shown significant activity. We hypothesized that denitrifying fungi would be found among those with assimilatory nitrate reductase, and tested 20 such batch cultures for their respiratory metabolism, including two positive controls, Fusarium oxysporum and Fusarium lichenicola, throughout the transition from oxic to anoxic conditions in media supplemented with NO 2 - . Enzymatic reduction of NO 2 - (NIR) and NO (NOR) was assessed by correcting measured NO- and N2 O-kinetics for ab…

Plant pathogenic fungi are a major threat to food security and impose a severe economic burden, thus there is a continuous need to develop new strategies to manage them. NAD+ is a co-factor in numerous enzymatic activities and determines the metabolic fate of the cell. Therefore, maintenance of NAD+ concentration is important for cellular viability. Consequently, the NAD+ biosynthetic pathway and redox homeostasis was suggested as a target for antifungal development. We aimed to study how Fusarium oxysporum senses and responds to nicotinaldehyde (NA), an inhibitor of Pnc1, a key enzyme in the …

Dihydroxyacid dehydratase (DHAD), encoded by ILV3, catalyses the third step in the biosynthetic pathway of branched-chain amino acids (BCAAs), which include isoleucine (Ile), leucine (Leu), and valine (Val). Enzymes involved in BCAA biosynthesis exist in bacteria, plants, and fungi but not in mammals and are therefore attractive targets for antimicrobial or herbicide development. In this study, three paralogous ILV3 genes (FgILV3A, FgILV3B, and FgILV3C) were identified in the genome of Fusarium graminearum, the causal agent of Fusarium head blight (FHB). Deletion of FgILV3A alone or combined w…

Deoxynivalenol (DON), zearalenone (ZEN) and fumonisin B1 (FB1) are among the most toxicologically important Fusarium toxins commonly found in nature that lead to nephrotoxicity in animals. The present study investigated that the individual and combined effects of subcytotoxic DON (0.25 μM), ZEN (20 μM) and FB1 (10 μM) on oxidative stress and apoptosis in porcine kidney cells (PK15). In addition, the protective effect of N-acetylcysteine (NAC) against the toxicity of Fusarium toxins was also evaluated. Our results showed that the activities of glutathione reductase (GR) and total superoxide dis…

Fusarium species are reported frequently as the most common causative agents of fungal keratitis in tropical countries such as India. Sixty-five fusaria isolated from patients were subjected to multilocus DNA sequencing to characterize the spectrum of the species associated with keratitis infections in India. Susceptibilities of these fusaria to ten antifungals were determined in vitro by the broth microdilution method. An impressive phylogenetic diversity of fusaria was reflected in susceptibilities differing at species level. Typing results revealed that the isolates were distributed among s…

To improve our knowledge of the role of microRNAs (miRs) in responses of the porcine digestive system to two Fusarium mycotoxins, zearalenone (ZEN) and deoxynivalenol (DON), we examined the expression of 7 miRs (miR-9, miR-15a, miR-21, miR-34a, miR-122, miR-125b, and miR-192), previously found to be deregulated in diseased liver and colon cells. In this study, immature gilts were exposed to NOEL doses of ZEN (40 μg/kg/d), DON (12 μg/kg/d), ZEN + DON (40 + 12 μg/kg/d), andplacebo (negative control group) for 7, 14, 21, 28, 35, and 42 days. Before the treatment, expression levels of the selected…

BACKGROUND: Fusarium onychomycoses are weakly responsive or unresponsive to standard onychomycosis treatments with oral terbinafine and itraconazole.
OBJECTIVE: To examine whether the use of terbinafine and itraconazole, which are highly effective in fighting Trichophyton onychomycoses, could be a cause of the high incidence of Fusarium nail infections.
METHODS: Polymerase chain reaction methods were used to detect both Fusarium spp. and Trichophyton spp. in nails of patients who had either received treatment previously or not.
RESULTS: No significant microbiological differences were found …

The ergosterol biosynthesis pathway is well characterized in Saccharomyces cerevisiae, while little is known about the pathway in filamentous fungi. In this study, we isolated and genetically documented biological functions of FgErg3 and FgErg5, which are located upstream of FgErg4, the enzyme catalyzing the final step of ergosterol synthesis in Fusarium graminearum. Our results demonstrated that F. graminearum contains two paralogous FgERG3 and two FgERG5 genes. FgErg3, but not FgErg5, is involved in ergosterol biosynthesis. Double deletion mutants of FgERG3 alleles or the double deletion mut…
