Escherichia coli (E. coli) F18 is the main pathogen responsible for post-weaning diarrhea (PWD) in piglets. Resistance to E. coli F18 depends on the expression of the cognate receptors in the intestinal epithelial cells. However, the molecular mechanism of E. coli F18 resistance in weaned piglets remains unclear. Here, we performed a comparative transcriptome study of the duodenal tissue from Sutai E. coli F18 sensitive and resistant pigs by RNA-seq, and pig α(1,2) fucosyltransferase 2 (FUT2) was identified as a host differentially expressed gene controlling the E. coli F18 infection. The distribution of alpha 2,3-sialyltransferase (alpha 2,3-ST) mRNA in the rat iris and ciliary body was investigated with in situ hybridization histochemistry. Strong expression of alpha 2,3-ST mRNA was detected in the inner epithelial layer of the ciliary body and weak expression in the iris epithelium. Since the synthesis of sialoglycoconjugates is completed by terminal sialylation by the action of sialyltransferase (ST), the ST-expressed portions are considered to produce sialoglycoconjugates. Hence, the source of the sialoglycoconjugates found in the inner epithelial layer of the ciliary

Studies on blood samples from a 30-year-old woman of Korean origin, her husband and children, showed the presence of the cis-AB genotype in the mother and one child. The cis-AB red cells showed abnormal reactions with both polyclonal and monoclonal anti-B reagents and an elevated level of H antigen. The serum contained weak anti-B that reacted with normal B antigen but not with that on cis-AB cells. Normal levels of A substance but reduced levels of B and H substances were present in the saliva. Conversely, the serum showed reduced A and normal B and H blood group transferase activity.

Estrogen deficiency after menopause accelerates bone loss by stimulating osteoclast formation and activity, but the molecular pathways that link estrogen signaling to osteoclast regulation remain incompletely defined. Here, we identify the sialyltransferase ST3GAL-I as a key mediator of RANKL-induced osteoclastogenesis. RANKL activates c-FOS to drive ST3GAL1 transcription, whereas estrogen-bound ERα competes with TRAF6 and suppresses this c-FOS-dependent induction. In a clinical cohort of pre-menopausal and post-menopausal women with or without osteoporosis, serum total and α-2,3-linked sialic

BACKGROUND: Pancreas fat deposition, measured by pancreatic proton density fat fraction (PDFF) in the MRI, has emerged as a critical area of research within the metabolic field. However, its genetic basis and how genetic predisposition impacts the pancreatic disorders remain poorly understood.
METHODS: We conducted a genome-wide association study (GWAS) in 20,040 European individuals (excluding those with acute/chronic pancreatitis and pancreatic cancer) from the UK Biobank. Subsequent post-GWAS analyses including colocalization, transcriptome-wide association studies (TWAS) and integrative a

BACKGROUND: Efficiency of engraftment after liver cell transplantation is less than 1% under conventional conditions. Our aim was to develop a high-efficiency, nonsurgical, no-genetic-advantage mouse model of liver repopulation with transplanted cells.
METHODS: Mice were conditioned with nonlethal doses of a cell cycle inhibitor, retrorsine, 70 mg/kg, to irreversibly block proliferation of native hepatocytes. After the drug was eliminated, 2 million freshly isolated beta-galactosidase-labeled liver cells were transplanted into the spleens of C57BL/6J recipient mice. To stimulate donor cell pr

Mutations in the presenilin genes are associated with early onset familial Alzheimer's disease and lead to accumulation of beta-amyloid peptide in the brain of patients, suggesting that presenilin abnormalities induce pathological processing of amyloid precursor protein (APP) in Alzheimer's disease. For the understanding of pathogenesis in this type of familal Alzheimer disease, it is important to know whether presenilins are directly involved in the metabolism of beta-amyloid or not. To test whether presenilin 1 (PS1) directly binds to APP, we performed two-hybrid interaction assays between t

Cytochrome P450 oxidoreductase (POR) acts as an electron donor for all cytochrome P450 enzymes. Knockout mouse Por(-/-) mutants, which are early embryonic (E9.5) lethal, have been found to have overall elevated retinoic acid (RA) levels, leading to the idea that POR early developmental function is mainly linked to the activity of the CYP26 RA-metabolizing enzymes (Otto et al., Mol. Cell. Biol. 23, 6103-6116). By crossing Por mutants with a RA-reporter lacZ transgene, we show that Por(-/-) embryos exhibit both elevated and ectopic RA signaling activity e.g. in cephalic and caudal tissues. Two s

BACKGROUND: The para-Bombay phenotype is characterized by H-deficient or H-partially deficient red blood cells (RBCs) in persons who secrete ABH antigens in their saliva. The studies that determined the genotypes for two Chinese individuals with the para-Bombay phenotype are described.
STUDY DESIGN AND METHODS: RBC phenotypes were characterized by conventional serologic methods. Exons 6 and 7 of the ABO gene were amplified, as well as the entire coding region for FUT1 and FUT2, with four independence polymerase chain reactions (PCRs) from genomic DNA. PCR products were excised, purified from

Scintigraphy with radiolabeled J001 as a ligand for macrophage targeting is a new approach for sarcoidosis imaging. J001 is a fully characterized acylated peptido-poly (1,3) galactoside isolated from Klebsiella membrane proteoglycans and able to bind electively recruited macrophages. Its physiochemical properties allow rapid absorption by the respiratory tract when this agent, labeled by 99m technetium, is administered as an aerosol. Images are obtained within 3 to 5 h after inhalation. In the present study, we determined the ability of J001 scintigraphy to localize areas of sarcoidosis involv

Administration of thyrotropin to porcine thyroid follicles, obtained in a serum-free chemically defined medium, provoked marked increases in the activities of several glycosyltransferases involved in protein N-glycosylation. The coincidence of these effects with a previously demonstrated enhancement of thyroglobulin production renders a relationship between these events likely. The most important stimulation was for peptide oligosaccharyltransferase (3-fold). Among the enzymes involved in the synthesis of the lipid oligosaccharide donor, Dol-P glycosyl- and mannosyltransferases were increased

INTRODUCTION: Galectin-9, a β-galactoside-binding protein, is elevated in various kidney diseases. While exogenous galectin-9 attenuates kidney injury in experimental models, it remains unclear whether endogenous plasma galectin-9 reflects renal histopathology and independently predicts long-term adverse outcomes in chronic kidney disease (CKD).
METHODS: Plasma galectin-9 levels were prospectively measured in 366 participants from the Taipei Renal Transcriptomics and Outcomes Investigation (TRTOI) cohort. Renal histopathology and chronicity were assessed using a modified Mayo Clinic Chronicit

Galectin-3 (Gal-3) is a β-galactoside-binding lectin implicated in metabolic inflammation, cardiovascular and renal dysfunction, neurodegenerative disorders, and obesity-related pathologies. Although Gal-3 is recognized as a clinically relevant biomarker, the mechanisms controlling its tissue expression and circulating abundance remain poorly defined. O-GlcNAcase (Oga; encoded by Mgea5), the enzyme that removes O-linked β-N-acetylglucosamine (O-GlcNAc) from proteins, regulates nutrient-sensitive signaling and transcriptional processes that overlap with Gal-3 associated disease pathways. Galectins, a structurally conserved family of β-galactoside-binding lectins, have emerged as key regulators of cancer progression. However, their integrated roles across tumor immunity, metabolic rewiring, and metastasis have yet to be fully defined. In this review, we synthesize current evidence on major galectin family members, particularly galectin (Gal)-1, Gal-3, Gal-4, Gal-7, Gal-9, and Gal-13, and delineate how they drive tumor progression through mechanistically distinct yet convergent pathways. Specifically, galectins induce T-cell dysfunction through T-cell immunoglobulin and mucin-do

Metabolic dysfunction-associated steatohepatitis (MASH) is characterized by hepatic steatosis, inflammation, fibrosis, and metabolic dysfunction, with limited therapeutic options currently available. This study investigated whether pharmacological inhibition of Galectin-3 (Gal-3), a β-galactoside-binding lectin implicated in inflammation and fibrosis, using modified citrus pectin (MCP), attenuates established MASH in ApoE-deficient mice. Mice were assigned to four groups: standard chow (CTL), standard chow plus MCP (CTL + MCP), Western diet with fructose-enriched drinking water (WD), and WD pl

L-asparaginase is a crucial enzyme used in chemotherapy regimens for the treatment of acute lymphoblastic leukemia (ALL), its incorporation in the pediatric treatment protocols helped in achieving a high cure rate. However, immunogenic side-effects restrict its application and frequently result in stopping treatment. There is a current need for the identification of novel L-asparaginase with improved properties and lower adverse effects compared to those available in the market. L-asparaginase gene from Stenotrophomonas maltophilia (S. maltophilia), an isolated organism that was mentioned as a

INTRODUCTION: Galectin-9 is a β-galactoside-binding lectin that functions as a critical pattern recognition receptor (PRR) in the host immune system, initiating immune defense responses by recognizing and binding to pathogen-associated molecular patterns (PAMPs) on the surface of microorganisms. In this study, we identified and characterized a novel galectin-9 cDNA, designated CcGal-9, from Yellow River carp (Cyprinus carpio haematopterus).
METHODS: The full-length CcGal-9 cDNA was cloned and sequenced, and its structural features were analyzed. Tissue distribution of CcGal-9 mRNA was examine

Obesity, type 2 diabetes mellitus (T2DM), and cardiovascular diseases (CVDs) represent major global health burdens with overlapping pathophysiological mechanisms, including chronic low-grade inflammation, oxidative stress, and gut microbiota dysbiosis. Galectins, a family of β-galactoside-binding lectins, have been implicated in immune regulation, inflammation, and tissue remodeling. Among them, Galectin-4 (Gal-4), primarily expressed in the gastrointestinal tract, has emerged as a potential biomarker due to its roles in epithelial integrity, inflammatory signaling, and metabolic regulation.
