This study reports the identification of a rare allele of TabZIP45-4B (TabZIP45-4Bpnd1) that confers tolerance to nitrogen starvation. Knockout of TabZIP45-4B increases spike number and grain yield per plant in wheat under nitrogen-starvation conditions by downregulating TaDWARF4. Introgression of TabZIP45-4Bpnd1 into a modern commercial cultivar improves multiple agronomic traits with minimal nitrogen input.

The Prunus mume species has undergone a long process of domestication and introduction in China, resulting in marked differences in freezing tolerance among populations across a latitudinal gradient of approximately 1,000 km. However, the introduction and domestication process is time-consuming, constraining the efficiency of species introduction. In this study, P. mume samples with the same genetic background spanning over 1,000 km from north to south were selected as experimental materials. The molecular mechanisms underlying cold-acclimation-mediated freeze tolerance in P. mume were investi

Mineral nutrients are essential for plant growth and development. Sulfur (S), as a macronutrient, is incorporated into numerous critical S-containing metabolites that play key roles in mitigating both abiotic and biotic stresses. Understanding how plants regulate S homeostasis and integrate it with other physiological processes is crucial for developing crops that can better withstand environmental challenges. Here, we demonstrate that the conserved sulfate starvation transcriptional response across Arabidopsis, tomato, rice, and Setaria is limited to only seven genes. We further characterize

Fruit shape is a key determinant of pepper (Capsicum annuum L.) fruit commercial value, and the shoulder is a crucial feature of the fruit's proximal end. Despite the fruit shoulder being an important parameter in morphological variation, the genes and regulatory mechanisms underlying fruit shoulder morphology remain elusive. Here, we identified a fruit shoulder (fs) mutant that primarily exhibits a shoulderless fruit phenotype, with an ∼40% reduction in fruit weight. Our data demonstrated that OVATE family protein 20 (CaOFP20) controls this phenotype. Silencing CaOFP20 in various fruit types

Reduced seed shattering is a key goal in crop domestication and improvement. Seed shattering 1 (Sh1) was parallel selected during the domestication of the Poaceae family. Investigating the regulatory network of OsSh1 in controlling seed shattering in rice (Oryza sativa L.) offers biological insights and potential for agricultural applications. Here, we identified a transcription factor of CONSTANS, CONSTANS-LIKE, and TOC1 (CCT) family, OsCCT22, that physically interacts with OsSh1 in rice. oscct22 mutants exhibited a phenotype similar to ossh1 mutants, characterized by incomplete development o

Lignification is one of the key factors restricting the postharvest storage quality of okra. Analyzing the key factors inducing lignin deposition is beneficial for targeted regulation of the quality deterioration caused by postharvest lignification of okra. The aim of this study is to elucidate the molecular mechanisms of key v-myb avian myeloblastosis viral oncogene homolog (MYB) transcription factors involved in postharvest lignification of okra in response to carboxymethyl chitosan (CMCS). Subcellular localization experiments confirmed that AeMYB6, AeMYB315-1, and AeMYB35-2 are all nuclear

Iron (Fe) is essential for plant development throughout the life cycle. Rosette leaves are responsive to Fe supply in Arabidopsis thaliana. Little is known about the dynamics of Fe deficiency (-Fe) responses of rosette leaves during the reproductive stages. We studied the dynamics of Fe-dependent responses at four consecutive reproductive stages (rosette, bolting, flowering, and mature silique stages, herein named RS, BS, FS, and MS). We examined the growth of rosette leaves, elemental contents, and gene expression patterns of Fe homeostasis genes belonging to differently regulated groups. We

Plant height is a key trait that significantly influences plant architecture, disease resistance, adaptability to mechanical cultivation, and overall economic yield. Galactinol synthase (GolS) is a crucial enzyme involved in the biosynthesis of raffinose family oligosaccharides (RFOs). It plays a significant role in carbohydrate transport and storage, combating abiotic and biotic stresses, and regulating plant growth and development. The present study employed CRISPR/Cas9 gene-editing technology to create the gols4 mutant in tomato (Solanum lycopersicum), which exhibits a semi-dwarf phenotype.

UV-A, an important part of sunlight radiation, is typically absent in experiments on plant-endophyte interactions. We examined the impact of UV-A in the 350-400 nm range (UV-A1 waveband) on the plant interactions with fungal endophytes belonging to different taxonomic groups: Paraphoma chrysanthemicola, Phomopsis columnaris, Diaporthe eres, Mucor sp., and yeast Sporobolomyces ruberrimus. Physiologically relevant levels of UV-A did not substantially affect the colonisation of shoots and roots by endophytes. UV-A upregulated the expression of genes involved in the establishment of symbiosis. Spe

The litchi genome has five anthocyanidin reductase (LcANR) and two leucoanthocyanidin reductase (LcLAR) members. The high expression of LcANR1a/2a and LcLAR1/2 is significantly positively correlated with the abundant proanthocyanidins and (-)-epicatechin (EC) in the pericarp, leaf, root, etc. The recombinant LcANR1a/2a converts cyanidin to both EC and (+)-catechin (CT) (EC:CT ≈ 1:1) and converts delphindin to (+)-gallocatechin and (-)-epigallocatechin; the recombinant LcLAR1/2 converts leucocyanidin to CT. The enzymatic kinetics of the four enzymes are presented, with the respective Km of LcLA

Dwarfism is an agronomic trait that has substantial effects on crop yield, lodging resistance, planting density, and a high harvest index. Ethylene plays an important role in plant growth and development, including the determination of plant height. However, the mechanism by which ethylene regulates plant height, especially in woody plants, remains unclear. In this study, a 1-aminocyclopropane-1-carboxylic acid synthase (ACC) gene (ACS), which is involved in ethylene biosynthesis, was isolated from lemon (Citrus limon L. Burm) and named CiACS4. Overexpression of CiACS4 resulted in a dwarf phen

Plant glutathione S-transferases are an ancient protein superfamily having antioxidant activity. These proteins are primarily involved in diverse plant functions such as plant growth and development, secondary metabolism, signaling pathways and defense against biotic and abiotic stresses. The current study aimed to comprehensively identify and characterize the GST gene family in the medicinally important crop Papaver somniferum. A total of 93 GST proteins were identified belonging to eight GST classes and found to be majorly localized in the cytoplasm. All GST genes were found on eleven opium

It is of great importance to better understand how trees regulate nitrogen (N) uptake under N deficiency conditions which severely challenge afforestation practices, yet the underlying molecular mechanisms have not been well elucidated. Here, we functionally characterized PuHox52, a Populus ussuriensis HD-ZIP transcription factor, whose overexpression greatly enhanced nutrient uptake and plant growth under N deficiency. We first conducted an RNA sequencing experiment to obtain root transcriptome using PuHox52-overexpression lines of P. ussuriensis under low N treatment. We then performed multi

BACKGROUND: Cucumber green mottle mosaic virus (CGMMV) causes substantial global losses in cucurbit crops, especially watermelon. N6-methyladenosine (m6A) methylation in RNA is one of the most important post-transcriptional modification mechanisms in eukaryotes. It has been shown to have important regulatory functions in some model plants, but there has been no research regarding m6A modifications in watermelon.
RESULTS: We measured the global m6A level in resistant watermelon after CGMMV infection using a colorimetric method. And the results found that the global m6A level significantly decr

WRKY transcription factors, one of the largest transcription factor families, play important roles in regulating the synthesis of secondary metabolites. In sweet osmanthus (Osmanthus fragrans), the monoterpenes have been demonstrated as the most important volatile compounds, and the W-box, which is the cognate binding site of WRKY transcription factors, could be identified in most of the terpene-synthesis-related genes' promoters. However, the role of the WRKY family in terpene synthesis in sweet osmanthus has rarely been examined. In this study, 154 WRKY genes with conserved WRKY domain were

The tomato Mi-1 gene mediates plant resistance to whitefly Bemisia tabaci, nematodes, and aphids. Other genes are also required for this resistance, and a model of interaction between the proteins encoded by these genes was proposed. Microarray analyses were used previously to identify genes involved in plant resistance to pests or pathogens, but scarcely in resistance to insects. In the present work, the GeneChip™ Tomato Genome Array (Affymetrix®) was used to compare the transcriptional profiles of Motelle (bearing Mi-1) and Moneymaker (lacking Mi-1) cultivars, both before and after B. tabaci

Sugarcane production is strongly influenced by drought, which is a limiting factor for agricultural productivity in the world. In this study, the gene expression profiles obtained by de novo assembly of the leaf transcriptome of two sugarcane cultivars that differ in their physiological response to water deficit were evaluated by the RNA-Seq method: drought-tolerant cultivar (SP81-3250) and drought-sensitive cultivar (RB855453). For this purpose, plants were grown in a greenhouse for 60 days and were then submitted to three treatments: control (-0.01 to -0.015 MPa), moderate water deficit (-0.

Antibiotic and herbicide resistance genes are currently the most frequently used selectable marker genes for plant research and crop development. However, the use of antibiotics and herbicides must be carefully controlled because the degree of susceptibility to these compounds varies widely among plant species and because they can also affect plant regeneration. Therefore, new selectable marker systems that are effective for a broad range of plant species are still needed. Here, we report a simple and inexpensive system based on providing transgenic plant cells the capacity to convert a nonmet

In May 2007, a widespread frost event defoliated much of Utah's high elevation aspen. About 5 weeks later, the frost-defoliated aspen produced a second leaf flush. The objective of this study was to characterize changes in leaf morphology and function in re-flush leaves following frost defoliation. Leaf size and thickness, photosynthesis, carbohydrate and nutrient status, and defense chemistry (phenolic glycosides and condensed tannins) were measured in first and second flush leaves. The second flush leaves produced two different morphological responses depending on frost damage severity. Seve
