Effect of inosine monophosphate dehydrogenase-1 gene polymorphisms on mycophenolate mofetil effectiveness in neuromyelitis optica spectrum disorder patients.

BACKGROUND: Inosine monophosphate dehydrogenase-1 is the target of mycophenolate mofetil. This research investigated the association between the gene polymorphism of inosine monophosphate dehydrogenase-1 and effectiveness of mycophenolate mofetil therapy in neuromyelitis optica spectrum disorder patients. METHODS: Fifty-nine neuromyelitis optica spectrum disorder patients accepted Mycophenolate Mofetil therapy for 1 year at least were divided into two groups: relapsing (n=21) and non-relapsing (n=38). Four single-nucleotide polymorphisms (SNPs: rs2228075, rs2278294, rs2288550, and rs3793165)

Challenging the traditional approach for interpreting genetic variants: Lessons from Fabry disease.

Fabry disease (FD) is an X-linked genetic disease due to pathogenic variants in GLA. The phenotype varies depending on the GLA variant, alpha-galactosidase residual activity, patient's age and gender and, for females, X chromosome inactivation. Over 1000 variants have been identified, many through screening protocols more susceptible to disclose non-pathogenic variants or variants of unknown significance (VUS). This, together with the non-specificity of some FD symptoms, challenges physicians attempting to interpret GLA variants. The traditional way to interpreting pathogenicity is based on a

HLA diversity in Brazil.

Brazil is the fifth largest country in the world in area and the fifth most populous. The Brazilian voluntary Bone Marrow Donor Registry is the third largest in terms of number of donors in the world, being a valuable source of HLA genetics to characterize the donor population of Brazil as well. The genetic background of the Brazilian population is quite heterogeneous, resulting from 5 centuries of admixture among Native Americans, Europeans and Africans, making the Brazilian population unique in terms of genetic ancestry. The unique characteristics of populations in different Brazilian region

Historic exposure to plague and present-day frequency of CCR5del32 in two isolated island communities of Dalmatia, Croatia.

AIM: To assess the frequency of deletion of 32 base pairs in a CCR5 gene, shown to confer resistance to HIV infection, in two isolated island communities of Dalmatia, Croatia, with different histories of exposure to (plague) during the medieval period and beyond. METHODS: Random samples of 100 individuals from highly isolated communities of Lopar (island of Rab) and Komiza (island of Vis) were selected in 2002 and their DNA was extracted. An extremely high level of 3-generational endogamy was found in both communities (98% and 91%, respectively), indicating very limited gene flow, which was c

Dermatoglyphic study in children with phenylketonuria.

Dermatoglyphic findings in 19 patients with phenylketonuria (11 male and 8 female), 39 of their relatives (18 female and 21 male) and 500 controls (TRC) were not statistically significant among the three groups studied. There was no definite relationship between the phenylketonuric gene and the dermatoglyphic patterns. The parents of half the phenylketonuria cases are not consanguineous; thus the phenylketonuria gene may be more frequent in Turkey than other European countries.

beta S-haplotypes and alpha-thalassemia along the coastal belt of Kenya.

In this paper, we present data on studies of beta S-haplotypes and alpha-thalassemia gene in subjects from the indigenous population of the Coast Province of Kenya Of the 7SS patients studied, four were homozygous for beta S-haplotype 20 characteristically associated with the severe form of sickle cell anemia found in the Central African Republic and Western Kenya. Two had haplotype 20 combined with haplotype 19 (Benin Type) and one had haplotype 20 combined with a new haplotype (20x). Alpha thalassaemia-2 gene (-3.7kb deletion) was detected in 45.6% of the 57AA subjects studied. An alpha glob

Beta-thalassemia associated with alpha-thalassemia in Thailand.

In Thailand alpha-thalassemia (thal), beta-thal, hemoglobin (Hb) E and Hb Constant Spring (Hb CS) are prevalent. The incidences are 20-30% for alpha-thal (3.5% for alpha-thal-1 and 16% for alpha-thal-2), 3-9% for beta-thal, up to 54% for Hb E and nearly 8% for Hb CS. Different combinations of these genes result in a spectrum of thalassemia syndromes ranging in severity from asymptomatic to intrauterine death. From the known gene frequencies the numbers of thalassemic patients per generation (total population of 50 million) are as follows: Hb Bart's hydrops fetalis 20,000; homozygous beta-thal

The DAM chicken: a model for spontaneous postnatal cutaneous and ocular amelanosis.

A mutant line of chickens (DAM line) has been developed that is characterized by a high incidence of a spontaneous, postnatal, cutaneous amelanosis. Amelanotic individuals also have a high incidence of blindness and a low incidence of an integumentary defect expressed as a variable loss of feathers. A low incidence of hypothyroidism is also present but is not limited to the amelanotic phenotype. All line associated traits appear to be controlled by multiple autosomal genes. Selection experiments showed that the amelanosis and eye defect are highly heritable and partially share a common genetic

Heterogeneity of glucose-6-phosphate dehydrogenase deficiency in Algeria. Study in Northern Algeria with description of five new variants.

Glucose-6-phosphate dehydrogenase (G6PD) deficiency was found in 3.2% of the male population living in the urban area of Algiers. The deficient subjects originated from multiple geographic regions of Northern Algeria, with prevalence of individuals of Berber-Kabyle origin. Red blood cell G6PD was partially purified and characterized in deficient males from 17 families, and six different variants were found. Among them, only one, the Gd(-) Kabyle variant, had been previously described. It was detected in nine families. The other five variants were new: Gd(-) Laghouat (four cases), Gd(-) Blida (

Red cell glucose-6-phosphate dehydrogenase deficiency and haemoglobin variants among ten endogamous groups of Maharshtra and West Bengal.

Over 900 individuals from ten endogamous groups in the Indian states of Maharashtra and West Bengal were studied for G-6-PD deficiency and haemoglobin variants. The incidence of G-6-PD varied from nil to 17.3%, while that of Hb-S varied from nil to 22.3%. In general, the tribal populations of Maharashtra are characterized by the presence of a high incidence of both Hb-S and G-6-PD deficiency. The caste Hindus showed an absence of Hb-S and rather low G-6-PD deficiency. Immigrant Parsis possessed the highest incidence of G-6-PD deficiency (17.3%).

A survey of glucose-6-phosphate-dehydrogenase deficiency in the North Central Province of Sri Lanka (formerly Ceylon).

A high frequency of the G6PD deficient gene was detected in the North Central Province of Sri Lanka. The frequency in the ancient villages is much higher than that of the recently colonised areas. The Sinhalese and Ceylon Moors have a significantly higher frequency as opposed to the Ceylon Tamils. The distribution appears to be related to a history of exposure to malarial endemicity.

[Temperature-sensitive autosomal dominant lethal mutations in Drosophila melanogaster induced by ethylmethane sulfonate].

Cold- and heat-sensitive dominant autosome and recessive sex-linked lethals were scored using C(1)RM, y; vg bw; e ss tester stock. The frequencies of heat-sensitive mutations were 1.43, 0.30, 0.07% and of cold-sensitive ones were 0.39, 0.16 and 0.09% in the 1-st, 2-nd and 3-rd chromosomes, respectively. For the first time, dominant cold-sensitive lethals were obtained in chromosome 3. The data from genetic analysis point to the fact that penetrance of such mutations strongly depends on the genetic background. That may be the reason, why they were not obtained using some of the balancer-3 chrom

Segregation analysis of juvenile myoclonic epilepsy.

We examined the inheritance of juvenile myoclonic epilepsy (JME). We looked at both the trait of "epilepsy" and the trait of "epilepsy-plus-EEG abnormalities," since EEG abnormalities are frequently found in the clinically unaffected sibs of JME patients. We tested several modes of inheritance including the fully penetrant recessive and several two-locus models. We could reject all models tested (fully penetrant single-locus and two-locus models) when abnormal EEGs were classified as "unaffected." We could also reject the fully penetrant single locus models when family members with abnormal EE

Genome Aggregation Database Version 4-Allele Frequency Changes and Impact on Variant Interpretation in Dystonia.

BACKGROUND: Population-scale databases majorly contribute to variant interpretation. The recently released Genome Aggregation Database (gnomAD) v4 offers a >5-fold increased sample size compared to v2.1.1. Pathogenic variants absent from v2.1.1 are now registered in v4 at a considerable rate. The implications on variant interpretation in dystonia are unknown. METHODS: All curated variants linked to the most common dominant forms of isolated dystonia were extracted from the International Parkinson's Disease and Movement Disorder Society Gene database. We compared variant population-frequencies
