Genetic factors are major determinants of phenotypic variability in a mouse model of the DiGeorge/del22q11 syndromes.

The del22q11 syndrome is associated with a highly variable phenotype despite the uniformity of the chromosomal deletion that causes the disease in most patients. Df1/+ mice, which model del22q11, present with reduced penetrance of cardiovascular defects similar to those seen in deleted patients but not with other del22q11-like findings. The reduced penetrance of cardiovascular defects is caused by the ability of mutant embryos to recover from a fourth pharyngeal arch artery growth abnormality that is fully penetrant in early embryos. Here we show that genetic background has a major effect on p

Genetic determinants of severity of acute withdrawal from diazepam in mice: commonality with ethanol and pentobarbital.

Potentially life-threatening seizures can occur following withdrawal from benzodiazepines, ethanol, or barbiturates. In animals, withdrawal severity has been shown to be partially genetically determined for each drug class. Susceptibility to these drugs is partially determined by common genetic factors, but the evidence is conflicting. We tested the hypothesis that acute benzodiazepine withdrawal convulsions are influenced by at least some genes that also affect withdrawal from ethanol and pentobarbital. Results in inbred mouse strains demonstrate that strain susceptibility is genetically corr

Genetic background determines the nature of immune responses and experimental immune-mediated blepharoconjunctivitis (EC).

PURPOSE: Experimental immune-mediated blepharoconjunctivitis (EC) was induced in Lewis rats by immunization with ovalbumin (OVA) in complete Freund's adjuvant (CFA) or aluminum hydroxide [Al(OH)3]. To investigate the affect of genetic factors on the susceptibility of EC, we tested different strains of rats for the development of EC. METHODS: Lewis and Brown Norway (BN) rats were immunized once with 100 microg of OVA in CFA or Al(OH)3. Three weeks later they were challenged with OVA in eye drops; 24 hours after the challenge they were sacrificed and their eyes, blood, and lymph nodes were harv

Genetic determinants of type 2 diabetes mellitus: lessons learned from family studies.

Non insulin dependent diabetes mellitus (Type 2) is a multifactorial disease, with a polygenic inheritance and environmental factors contributing to its clinical expression. The search for the genetic determinants of Type 2 diabetes began when several genes involved in the mechanisms of insulin secretion or action were cloned, localized in the human genome, and when informative polymorphisms were described within or in the vicinity of these genes. It then became possible to compare, in groups of patients and normoglycaemic controls from various populations, the frequency of the different allel

[Clinical and morphological aspects of the state of the sclera in various types of genetically determined corneal changes].

The authors analyze the results of a morphologic examination of 5 trephination scleral disks in various types of corneal curvature changes: with ectasia of the lower half of the cornea (1), with uneven corneal ectasia (1), with true keratoconus (2), and with keratoglobus (1), and come to a conclusion that involvement of the scleral connective tissue presenting as extracellular dysproteinosis is the morphologic substrate of deterioration of the elastic properties of the sclera, clinically observed in patients with various genetic corneal ectasias. Involvement of the sclera in the dystrophic pro

Genetic mapping of novel virulence determinants of Salmonella typhimurium to the region between trpD and supD.

Salmonella typhimurium strains are known to vary greatly in their virulence for mice. A few of the genes affecting their virulence have been described. In this report we have localized at least two genes that affect the ability of S. typhimurium to grow in BALB/cByJ to a 6 unit section of the salmonella chromosome which does not contain any previously described virulence determinants. The genetic mapping was done by interrupted matings using Hfr strains made in a virulent LT2 strain. The Hfr strains were constructed by inserted the plasmid F'(TS)114 lac+ Tn::10 into the LT2 chromosome at speci

A guide to host genetic determinants of female microbiome composition across distinct anatomical niches.

The female microbiome is a complex, dynamic and site-specific ecosystem that plays a vital role in maintaining host health, influencing immune regulation, metabolism, reproductive function and dermatological health. While significant attention has been directed towards modifiable lifestyle and environmental influences, host genetics, including inherited genetic variants in immune, cell signalling, metabolic, hormonal pathways and epithelial structure/barrier integrity, are increasingly recognised as contributors to microbial composition across distinct anatomical niches in individuals. It is t

Genetic determinants of fatigue up to 2 years after radiotherapy in prostate cancer patients.

Fatigue is a common symptom of cancer patients, which can impair quality of life even years after treatment. Little is known about genetic determinants of fatigue, especially in prostate cancer (PCa). This study aims to identify SNPs associated with long-term fatigue in a prospective cohort of PCa patients. A genome-wide association study was conducted in non-metastatic PCa patients treated with radiotherapy in 7 European countries and the USA. A total of 1,381 men recorded fatigue using the EORTC QLQ-C30 and 877 men additionally completed the Multidimensional Fatigue Inventory (MFI) up to two

Genetic and epigenetic determinants of cytochrome P450 activity in psychopharmacology: from pharmacogenetics to functional pharmacogenomics.

Classical pharmacogenetics has explained interindividual variability in psychotropic drug response primarily through inherited polymorphisms in cytochrome P450 enzymes. This framework successfully identified extreme metabolizer phenotypes and informed genotype-guided dosing recommendations. However, genotype-based predictions frequently correlate more strongly with pharmacokinetic parameters than with clinical outcomes. Patients sharing similar CYP genotypes often exhibit divergent therapeutic trajectories, while metabolic phenotypes may change during treatment without corresponding alteration

Genetically determined causality between immune cells and anxiety disorders: A Mendelian randomization study.

Prior investigations have endeavored to unveil the connection between immune cells and anxiety disorders. However, the majority are limited to a single or few indicators and have not comprehensively evaluated the causal relationship between immune cells and anxiety disorders. In this study, we performed a two-sample Mendelian randomization (MR) analysis to assess the relationship between 731 immune phenotypes and anxiety disorders, offering genetic evidence to substantiate the correlation between immune phenotypes and the risk of anxiety disorders. Using publicly available data from genome-wid

Genetic and immunological determinants of Pemphigus vulgaris: integrative analysis of HLA-DRB1 and FCGR2B variants.

Pemphigus vulgaris (PV) is a rare autoimmune blistering disease mediated by pathogenic autoantibodies. Although both HLA and non-HLA loci contribute to disease susceptibility, their combined roles in immune regulation remain incompletely understood. This study investigated the joint contribution of HLA-DRB1 and FCGR2B variants to PV susceptibility within an integrative immunogenetic framework. Genotype data from 286 individuals (200 controls and 86 PV patients) were analyzed using bias-reduced association models, two-locus genotype combination analyses, and cumulative genetic risk modeling. Epidemiology and genetic determination of measures of peripheral vascular health in the Long Life Family Study.

Peripheral artery disease (PAD) is a major contributor to morbidity in older adults. We aimed to determine genetic and non-genetic determinants of PAD and ankle-brachial index (ABI) in the Long Life Family Study (LLFS). 3006 individuals had ABI assessment, including 1090 probands (mean age 89), 1554 offspring (mean age 60) and 362 spousal controls (mean age 61). Outcomes include minimum of right and left ABIs and PAD (ABI <0.9). Stepwise regression determined independent significant non-genetic correlates of ABI and PAD. Genomewide association and linkage analyses were adjusted for age, sex, s

Identification of genetic determinants that drive population differentiation and host adaptation in Campylobacter jejuni in California.

Campylobacter jejuni is a commensal bacterium that colonizes livestock and wild animals and is responsible for more than 80% of campylobacteriosis cases in humans, which all ultimately have an animal source. Its high genome plasticity and recombination rate allow it to adapt to multiple hosts and lead to the rapid emergence of lineages, some of which can be antibiotic resistant. Our aim in this study was to examine the population structure of Campylobacter in California and assess its differentiation between animal hosts and humans. We sequenced 69 human clinical isolates of C. jejuni from Cal

Genetics and environmental determinants of Alzheimer's disease and related dementias in older Africans: a narrative review.

The burden of Alzheimer's disease and related dementias (ADRD) is rising in Africa, yet research remains limited compared to high-income regions. This narrative review investigated the genetic and environmental determinants of ADRD in older African populations, with a focus on the apolipoprotein E (APOE) gene. Although APOE ε4 is a strong risk factor for Alzheimer's disease globally, its role in African populations appears less pronounced and variable, likely due to genetic diversity, evolutionary adaptations, and environmental interactions. We discussed the epidemiology of dementia in Africa,

Genetic determinants of longitudinal behavioural trajectories in rare conditions: The case of fragile X syndrome.

Despite being a monogenic condition, individual variability in the phenotypic profile of fragile X syndrome (FXS) is substantial, with behavioural outcomes differing in severity and frequency. Existing studies have revealed that common variation in 5-HTTLPR (serotonin) and COMT (dopamine) single nucleotide polymorphisms (SNPs) is associated with behavioural variation in FXS when measured cross-sectionally. However, the associations between SNPs and longitudinal behavioural trajectories in FXS remain unknown. This study explored relationships between three SNPs, selected a priori (5-HTTLPR, COM
