Genetic evidence supports primary biliary cholangitis as a risk factor for lacunar stroke.

Primary biliary cholangitis (PBC) is a chronic autoimmune liver disease, while lacunar stroke is a common cerebrovascular subtype. Understanding potential systemic vascular consequences of PBC is crucial for patient management. Using a two-sample Mendelian randomization (MR) approach, we investigated the causal effect of PBC on lacunar stroke risk. We utilized genetic instruments for PBC from large-scale genome-wide association studies and tested their association with lacunar stroke in independent datasets. In the discovery analysis, genetically predicted PBC was associated with an increased

Genetic predisposition to systemic inflammatory proteins is causally associated with inflammatory bowel disease: Insights from multi-omics association study and single-cell RNA-sequencing analysis.

Systemic inflammatory proteins have been reported to be related to inflammatory bowel disease (IBD) in previous observational research. However, their causal links remain obscure. Herein, we performed a Mendelian randomization (MR) analysis to analyze the causality between systemic inflammatory proteins and IBD. Genetic variants related to systemic inflammatory proteins were extracted from a meta-analysis of genome-wide association study (GWAS) data of 8293 European participants. Summary statistics of IBD diverse subtypes were obtained from the international IBD genetic consortium (IIBDGC). We

The impact of educational attainment on the risk of three types of atherosclerosis: A two-sample mendelian randomization analysis.

BACKGROUND: Atherosclerosis is a leading contributor to cardiovascular disease and mortality worldwide, influenced by both genetic and environmental factors. Educational attainment, as a key socioeconomic indicator, has been consistently associated with cardiovascular risk in observational studies. However, the causal nature of this relationship remains uncertain. This study aimed to investigate the potential causal association between educational attainment and the risk of atherosclerosis using Mendelian randomization (MR) methods. METHODS: We performed a two-sample MR analysis using genome-

A two-sample Mendelian randomization analysis of the impact of GABA receptor subtypes on human height.

Gamma-aminobutyric acid (GABA) supplements are increasingly marketed for promoting growth, yet the genetic influence of GABA receptor subtypes on human height remains unexplored. This Mendelian randomization (MR) study aimed to investigate the causal relationships between genetically predicted plasma levels of 2 GABA receptor subtypes (GABRA4 and GABRB2), and height-related outcomes, including body height (BH), idiopathic short stature (ISS), and constitutional tall stature (CTS). This study employed a two-sample MR approach to assess the causal effect of GABA receptor subtypes, specifically G

Lipids, lipid-lowering drugs and the risk of herpes zoster: a Mendelian randomization study.

Observational studies have shown that the risk of developing herpes zoster (HZ) increases with the use of statins. However, there are many confounding factors in observational studies. Therefore, our Mendelian randomization (MR) study aimed to explore the causal role of lipids in HZ and to assess the causal impact of lipid-lowering drug targets on HZ risk. Our study used low-density lipoprotein (LDL) as a biomarker, and MR analysis was applied to study the effects of genetic inhibition of 3-hydroxy-3-methylglutaryl CoA reductase (HMGCR, targeted by statins), Niemann-Pick C1-like 1 (NPC1L1, tar

Single-cell expression quantitative trait loci-based Mendelian randomization identifies MAN1A2 in naive CD4+ T cells as a candidate therapeutic target for osteoarthritis.

BACKGROUND: Osteoarthritis (OA) is a leading cause of chronic pain and disability worldwide, with growing evidence implicating immune dysregulation in its pathogenesis. However, the specific peripheral immune cell populations contributing causally to OA risk remain poorly characterized. METHODS: We performed a multi-stage two-sample Mendelian randomization (MR) study integrating single-cell cis-expression quantitative trait loci (cis-eQTLs) for 14 peripheral immune cell types from the OneK1K cohort with two independent OA GWAS datasets - the Million Veteran Program for discovery and the UK Bi

Revealing the effects of various immune cells in anorexia nervosa: Evidence from Mendelian randomization.

The aim of this study was to assess the causal relationship between immune cells and anorexia nervosa (AN) by Mendelian randomization (MR). Data on immune cell phenotypes and AN were obtained from genome-wide association studies. Next, single nucleotide polymorphisms included in the MR analysis were screened according the basic assumptions. Furthermore, inverse variance weighted was used as the main method for MR analysis to evaluate the causal effect of immune cell phenotypes on AN. Finally, MR-Egger intercept, Cochran Q, and leave-one-out sensitivity analyses were used to assess horizontal p

Identification and replication of sex-dimorphic protein quantitative trait loci across multiple ancestries and their associations with diseases.

Males and females exhibit differences in proteome profiles associated with disease risk. However, sex-dimorphic protein quantitative trait loci (SD-pQTL) and their effects on sex differences in health disorders have not been thoroughly investigated. We conducted a sex-stratified, genome-wide association study on 2,922 proteins using data from 30,272 individuals of Caucasian ancestry from the UK Biobank and compared the estimated effects on protein levels of these variants in the men and women to identify SD-pQTLs. The identified SD-pQTLs were replicated using data from two Japanese cohorts (co

Genetic causal role of body mass index in multiple neurological diseases.

Body mass index (BMI) is a crucial health indicator for obesity. With the progression of socio-economic status and alterations in lifestyle, an increasing number of global populations are at risk of obesity. Given the complexity and severity of neurological diseases, early identification of risk factors is vital for the diagnosis and prognosis of such diseases. In this study, we employed Mendelian randomization (MR) analysis utilizing the most comprehensive genome-wide association study (GWAS) data to date. We selected single nucleotide polymorphisms (SNPs) that are unaffected by confounding f

Pleiotropy analysis between lobar intracerebral hemorrhage and CSF β-amyloid highlights new and established associations.

BACKGROUND AND AIMS: Combining biologically related traits in genome-wide association studies (GWAS) increases the power for genetic discovery. Given the established relationship between lobar intracerebral hemorrhage (ICH) and cerebral amyloid angiopathy (CAA), and between the latter and levels of cerebrospinal fluid amyloid-β 42 (CSF-Aβ42), we leveraged genetic predisposition for lower CSF-Aβ42 levels as a proxy phenotype for CAA to identify new genes associated with lobar ICH. METHODS: We used publicly available GWAS data for CSF-Aβ42 levels (n = 3146) and for lobar ICH (n = 2094). First,

New target-HMGCR inhibitors for the treatment of primary sclerosing cholangitis: A drug Mendelian randomization study.

BACKGROUND: No intervention definitively extends transplant-free survival in primary sclerosing cholangitis (PSC). Statins, inhibitors of 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGCR), may enhance PSC prognosis, but their efficacy is debated. METHODS: We analyzed HMGCR single-nucleotide polymorphisms from published genome-wide association studies using Mendelian randomization to assess the causal relationship between HMGCR and PSC risk. Effects of HMGCR were compared with proprotein convertase subtilisin kexin 9 (PCSK9) inhibitors, common lipid-lowering drugs, using coronary heart d

Calcium homeostasis and human evolution.

The maintenance of calcium homeostasis is critical to survival. The systems dependent upon the availability of calcium include the musculoskeletal, nervous, and immune systems, all of which utilize the ionic properties of divalent calcium to perform critical functions. The widespread utilization of calcium in essential biological processes across a wide range of phyla argues strongly for its adoption early in the evolution of life. In the broad context of evolution, the use of calcium in the structure of the skeleton is a relatively late, or derived trait. It is not surprising, therefore, that
