The Oral-Gut-Immune-Nutrition Axis in Rheumatoid Arthritis: Molecular Mechanisms and Therapeutic Implications.

Rheumatoid arthritis is a chronic systemic autoimmune disease that arises from complex interactions among genetic susceptibility, environmental factors, and immune dysregulation. Growing evidence indicates that microorganisms residing in the oral cavity and gastrointestinal tract, together with dietary factors, play a central role in shaping inflammatory and autoimmune responses in rheumatoid arthritis, forming an interconnected microbiome-immune-nutrition axis. Alterations in the composition and function of oral and intestinal microbial communities are associated with disruption of mucosal ba

Spontaneous development of autoimmune arthritis due to genetic anomaly of T cell signal transduction: Part 1.

A point mutation of the gene encoding ZAP-70, a key signal transduction molecule in T cells, results in spontaneous development of T cell-mediated autoimmune arthritis in mice homozygous for the mutation. The genetic anomaly alters differentiation and selection of T cells in the thymus, leading to thymic production of arthritogenic autoimmune T cells. The arthritogenic T cells persist in the periphery and elicit arthritis when activated by microbial agents that stimulate innate immunity. This model is instrumental in understanding how genetic variations in T cell signal transduction, together

Host diets drive microbiome evolution and functional divergence.

Substantial variation in gut microbiome function across individuals underscores the pivotal role of the host in shaping microbial diversity. Increasing evidence indicates that diet is a major selective force driving the evolutionary trajectory of the gut microbiome. In response to dietary inputs, the microbiome adapts by shifting the relative abundance of taxa, species, and sub-strains. The gut microbiome also acquires novel genetic features through selecting advantageous mutations and genes, as well as colonizing exogenous bacterial species that enhance nutrient utilization. These adaptive pr

Distribution and influencing factors of antibiotic resistance genes in two mussel species along the coasts of the East China Sea and the Yellow Sea.

Antibiotic resistance genes (ARGs) raise a global public health concern. The ARGs profile in marine aquaculture environments was well reported, yet it is poorly revealed in marine bivalves. This study investigated the microbiota, resistome, and environmental factors within the digestive glands of two mussel species (Mytilus coruscus and Mytilus galloprovincialis) cultivated in the East China Sea and Yellow Sea. The microbial communities in the digestive glands of mussels exhibit significant variations across different sampling sites and between the two seas. The three bacterial phyla that pred

Structural and functional shifts of mercury methylating microorganisms during an exceptional drought can elevate ecosystem risk of nearshore area.

Mercury (Hg) is a global pollutant and its transformation to the highly toxic methylmercury (MeHg) is mediated by anaerobic microbes carrying hgcA gene, which can affect human health risk through fish consumption. The Pearl River Estuary in southern China is productive but suffers from increasing anthropogenic stresses. However, it remains poorly known regarding the environmental response of hgcA+ microorganisms to climatic variations. We demonstrated that saltwater intrusion driven by prolonged drought led to elevated sedimentary MeHg concentrations, particularly in the nearshore zone, compar

X-Ray Photoelectron Spectroscopy on Microbial Cell Surfaces: A Forgotten Method for the Characterization of Microorganisms Encapsulated With Surface-Engineered Shells.

Encapsulation of single microbial cells by surface-engineered shells has great potential for the protection of yeasts and bacteria against harsh environmental conditions, such as elevated temperatures, UV light, extreme pH values, and antimicrobials. Encapsulation with functionalized shells can also alter the surface characteristics of cells in a way that can make them more suitable to perform their function in complex environments, including bio-reactors, bio-fuel production, biosensors, and the human body. Surface-engineered shells bear as an advantage above genetically-engineered microorgan

Penicillin and cephalosporin resistance in gonococci.

Non-penicillinase producing Neisseria gonorrhoeae isolated at St Mary's Hospital, London were examined for the prevalence of resistance to penicillin and for decreased susceptibility to cefuroxime. Of the 941 non-PPNG tested 100 (10.6%) were resistant to penicillin (minimum inhibitory concentration, MIC, greater than or equal to 1 mg/l) and were considered to be chromosomally-resistant N gonorrhoeae (CMRNG). Decreased susceptibility to cefuroxime (MIC, greater than or equal to 0.5 mg/l) was detected in 79% of the CMRNG. The CMRNG were also more often prototrophic and of serogroup IB than the r

Contribution of parasite and host genotype to immunopathology of schistosome infections.

BACKGROUND: The role of pathogen genotype in determining disease severity and immunopathology has been studied intensively in microbial pathogens including bacteria, fungi, protozoa and viruses but is poorly understood in parasitic helminths. The medically important blood fluke Schistosoma mansoni is an excellent model system to study the impact of helminth genetic variation on immunopathology. Our laboratory has demonstrated that laboratory schistosome populations differ in sporocyst growth and cercarial production in the intermediate snail host and worm establishment and fecundity in the ver

Location-specific signatures of Crohn's disease at a multi-omics scale.

BACKGROUND: Crohn's disease (CD), an inflammatory bowel disease (IBD) subtype, results from pathologic interactions between host cells and its resident gut microbes. CD manifests in both isolated disease locations (ileum or colon) or a combination of locations (ileocolonic). To date, a comprehensive understanding of how isolated CD subtypes influence molecular profiles remains outstanding. To address this, we sought to define CD location signatures by leveraging a large cross-sectional feature set captured from the stool of over 200 IBD patients and healthy controls using metaproteomics, shotg

GLP-1 agonists and the gut microbiome: A bidirectional relationship.

Glucagon-like peptide-1 (GLP-1) receptor agonists have transformed the management of type 2 diabetes mellitus (T2DM) and obesity, yet their interactions with the gut microbiome remain an emerging frontier in pharmacological and metabolic research. Mounting evidence suggests that the gut microbiota modulates GLP-1 secretion via microbial metabolites, including short-chain fatty acids and bile acid derivatives, while GLP-1 agonists reciprocally reshape microbial composition, influencing metabolic outcomes beyond their classical incretin functions. This bidirectional interplay has profound implic

Root Transporters Shape Rhizosphere Microbiomes to Enhance Nitrogen Acquisition Efficiency in Plants.

Root nitrogen acquisition is a central belowground process that determines how efficiently plants capture nitrogen from the rhizosphere and influences fertilizer demand and environmental nitrogen losses. Root N transporters, including nitrate, ammonium, amino acid, and peptide transporters, provide the molecular basis for inorganic and organic N uptake. However, root N acquisition is not determined solely by plant transport systems but is also shaped by rhizosphere microbial communities that regulate N mobilization, transformation, and availability. In addition to bacteria and archaea, saprotr

Skin microbiome characterization in acne vulgaris across urban and rural Egyptian populations.

BACKGROUND: Cutibacterium acnes is recognized as a key contributor to acne, but recent evidence suggests that shifts in skin microbial diversity, rather than simple overgrowth, are critical in disease progression. Despite the unique genetic and environmental characteristics of the Middle East and North Africa, microbiome data on acne remain scarce. OBJECTIVE: To characterize the skin microbiome associated with acne vulgaris in Egyptian urban and rural populations and assess the influence of acne severity and lifestyle factors on microbial diversity. METHODS: We recruited 45 acne patients (ur

Microbiota in cholestatic diseases: crosstalk among bile composition, the biliary microbiome, and host immunity.

Cholestatic liver diseases are a heterogeneous group of hepatobiliary disorders caused by impaired bile formation, secretion, or excretion, leading to hepatocyte injury, biliary inflammation, fibrosis, and eventually cirrhosis. Traditional studies have largely focused on isolated mechanisms, including bile acid toxicity, immune dysregulation, and genetic susceptibility. However, recent advances in metagenomics, metabolomics, and immunology have highlighted the critical role of the gut and biliary microbiota in disease pathogenesis. This review proposes the core concept of a "tripartite interpl

Unveiling the role of phages in shaping the periodontal microbial ecosystem.

The oral microbiome comprises various species and plays a crucial role in maintaining the oral ecosystem and host health. Phages are an important component of the periodontal microbiome, yet our understanding of periodontal phages remains limited. Here, we investigated oral periodontal phages using various advanced bioinformatics tools based on genomes of key periodontitis pathogens. Prophages were found to encode various auxiliary genes that potentially enhance host survival and pathogenicity, including genes involved in carbohydrate metabolism, antibiotic resistance, and immune modulation. Molecular characterization of microbial contaminants isolated from Umbilical Cord Blood Units for transplant.

Disposal of Umbilical Cord Blood Units due to microbial contamination is a major problem in Cord Blood Banks worldwide as it reduces the number of units available for transplantation. Additionally, economic losses are generated as result of resources and infrastructure used to obtain such units. Umbilical Cord Blood Units that showed initial microbial contamination were subject to strains isolation, identification, and characterization by sequencing the 16S rRNA gene and Enterobacterial Repetitive Intergenic Consensus (ERIC-PCR). Moreover, tests of antimicrobial resistance/sensitivity and phen

Multiparameter analysis of immunogenetic mechanisms in clinical diagnosis and management of inflammatory bowel disease.

The integrity of the intestinal mucosa depends on a functional coordination of the epithelium, lumenal microorganisms, and the local immune system. The mammalian immune system is superbly organized for innate and adaptive recognition of microbial antigens, a defensive capacity that must be balanced against the tissue damage produced by immune activity to preserve normal intestinal function. Inflammatory bowel disease (IBD) is generally thought to reflect an impairment in this balance, due to a combination of host genetic traits that shift the balance of immune and epithelial function to commen
