Rigidity-Activity Relationships of bisQPC Scaffolds against Pathogenic Bacteria.

Biscationic quaternary phosphonium compounds (bisQPCs) represent a promising class of antimicrobials, displaying potent activity against both Gram-negative and Gram-positive bacteria. In this study, we explored the effects of structural rigidity on the antimicrobial activity of QPC structures bearing a two-carbon linker between phosphonium groups, testing against a panel of six bacteria, including multiple strains harboring known disinfectant resistance mechanisms. Using simple alkylation reactions, 21 novel compounds were prepared, although alkene isomerization as well as an alkyne reduction …

Advances and updates in antibiotic combination therapy.

INTRODUCTION: While antibiotic combination therapy remains a common clinical practice, its scientific foundation is fragile. The available evidence is fragmented, biased, and fails to capture the complexity of modern infectious disease scenarios. This perspective reinterprets the role of combination therapy through a critical lens, challenging current dogmas and proposing a pathogen-specific approach, focusing also on severe acute infections. AREAS COVERED: This Critical Perspective focused on selected studies from 2018 to 2025 identified through a focused PubMed search on the place in therap

Open Fractures: Are We Still Treating the Same Types of Infections?

Background: To analyze the rate of methicillin-resistant Staphylococcus aureus (MRSA), gram-negative, and polymicrobial infections in open f … had polymicrobial infections. Cephalosporins and β-lactam agents had a 59.2% efficacy rate against gram-positive bacteria and gentamicin showed a 94% sensitivity rate against gram-negative bacteria. Vancomycin (95.8% sensitivity) demonstrated the highest sensitivity for all gram-positive organisms. Amikacin (98.8% sensitivity), meropenem (96.3% sensitivity), and gentam …

Direct identification and susceptibility testing of positive blood cultures using high speed cold centrifugation and Vitek II system.

Compared to routine isolated colony-based methods, direct testing of bacterial pellets from positive blood cultures reduces turnaround time for reporting of antibiotic susceptibility. The aim of this study was to compare the accuracy, and precision, of a rapid method for direct identification and susceptibility testing of blood cultures with the routine method used in our laboratory, using Vitek 2. A total of 60 isolates were evaluated using the candidate and the routine method. The candidate method had 100% accuracy for the identification of Gram negative bacteria, Staphylococcus and Enteroco

Ceftobiprole: a novel cephalosporin with activity against Gram-positive and Gram-negative pathogens, including methicillin-resistant Staphylococcus aureus (MRSA).

Ceftobiprole is a novel broad-spectrum cephalosporin with activity against a wide range of Gram-positive and Gram-negative bacteria, including several resistant species such as methicillin-resistant Staphylococcus aureus and penicillin-resistant Streptococcus pneumoniae. Ceftobiprole is administered intravenously as the prodrug ceftobiprole medocaril, which is almost immediately converted to the active form. It is currently under review by the US Food and Drug Administration (FDA) and is approved in Canada under the trade name Zeftera. The pharmacokinetics of ceftobiprole are non-complex as it

Tigecycline: clinical evidence and formulary positioning.

Tigecycline, is a novel broad-spectrum glycylcycline antibiotic, which has activity against a broad range of Gram-positive, Gram-negative, atypical, anaerobic and antibiotic-resistant bacteria. This includes activity against MRSA, VRE and penicillin resistant Streptococcus pneumoniae. Whilst exhibiting antibacterial activities typical of earlier tetracyclines, it has more potent activity against tetracycline-resistant organisms. Although a bacteriostatic compound in vitro, its effectiveness in clinical trials suggests that traditional laboratory thinking about using bacteriostatic drugs in ser

In vitro and in vivo antibacterial activities of GV129606, a new broad-spectrum trinem.

GV129606 is a new parenteral trinem antibiotic belonging to the beta-lactam class. It combines broad-spectrum activity (against gram-negative and -positive bacteria, aerobes and anaerobes), with high potency and resistance to beta-lactamases. Comparative in vitro and in vivo antibacterial activities were determined for GV129606 against more than 400 recent clinical isolates (aerobes, including beta-lactamase producers, and anaerobes), using representative antibacterial agents (meropenem, piperacillin, ceftazidime, cefpirome, ciprofloxacin, and gentamicin for aerobes and metronidazole, cefoxiti

[Beta-lactamase production and the patterns of bacterial resistance, 1988-1991].

The adequate use of antimicrobial agents depends to a great degree on the results obtained from the continuous surveillance of bacterial res … ive and 2446 were gram-negative. The study also included 1350 nosocomial isolated of which 509 were gram-positive bacteria and 849 were gram-negative bacteria. Overall the gram-negative bacteria were more frequently Beta-lactamase producers than the gram-positive bacteria. Resistance to beta-lactam antibiotics ranged from 64-100% in gram negative bacteria and from 81.96 …

Gram staining for the treatment of peritonsillar abscess.

Objective. To examine whether Gram staining can influence the choice of antibiotic for the treatment of peritonsillar abscess. Methods. Between 2005 and 2009, a total of 57 cases of peritonsillar abscess were analyzed with regard to cultured bacteria and Gram staining. Results. Only aerobes were cultured in 16% of cases, and only anaerobes were cultured in 51% of cases. Mixed growth of aerobes and anaerobes was observed in 21% of cases. The cultured bacteria were mainly aerobic Streptococcus, anaerobic Gram-positive cocci, and anaerobic Gram-negative rods. Phagocytosis of bacteria on Gram stai

Aesculus hippocastanum-Derived Extract β-Aescin and In vitro Antibacterial Activity.

OBJECTIVES: The objective of this study was to investigate the antibacterial activity of β-aescin against common Gram-negative and Gram-positive bacteria. MATERIALS AND METHODS: Agar well diffusion assay was used to determine the antibacterial activity of β-aescin against common Gram-negative and Gram-positive bacteria including Klebsiella pneumoniae, Escherichia coli, Staphylococcus epidermidis, Staphylococcus aureus, and Pseudomonas aeruginosa. Minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) of β-aescin were evaluated by serial dilution method. RESULTS: …

[Occurrence of granulomatous appendicitis in rabbits].

Granulomatous appendicitis was observed in all of the 45 Japanese white rabbits examined. Histologically, multiple microgranulomas were accompanied with foreign body giant cells and focal calcifications in lymph nodules of appendix. Foreign body giant cells contained hair coat and larvae of Passalurus ambiguus. In addition, the sacculus rotundus and mesenteric lymph nodes were affected with the same lesions. PAS- and Gram- positive bacilli were phagocytized in the microgranulomas and macrophages. They were also stained positively with the immunoperoxidase method for the auto-sera of rabbits. I
