A cDNA clone for a pathogenesis-related protein 1 from barley.

A barley cDNA clone (PRb-1) corresponding to an mRNA differentially induced in resistant compared to susceptible barley cultivars by powdery mildew infection was isolated and characterised. The deduced amino acid sequence revealed 24 amino acids comprising the signal peptide and 140 amino acids of the mature peptide (15 kDa). This showed close homology to PR-1-like proteins, which have been isolated from maize, tobacco, tomato and Arabidopsis thaliana. Northern blot analysis showed accumulation of the corresponding mRNA 12 h after inoculation of resistant barley cultivars with Erysiphe gramini

Solution structure of gamma 1-H and gamma 1-P thionins from barley and wheat endosperm determined by 1H-NMR: a structural motif common to toxic arthropod proteins.

The complete assignment of the proton NMR spectra of the homologous gamma 1-hordothionin and gamma 1-purothionin (47 amino acids, 4 disulfide bridges) from barley and wheat, respectively, has been performed by two-dimensional sequence-specific methods. A total of 299 proton-proton distance constraints for gamma 1-H and 285 for gamma 1-P derived from NOESY spectra have been used to calculate the three-dimensional solution structures. Initial structures have been generated by distance geometry methods and further refined by dynamical simulated annealing calculations. Both proteins show identical

Overexpression of barley heat stress transcription factor HvHsfA6a provides thermotolerance by modulating transcriptome and metabolome.

Global climate change has adverse impacts on crop production. Plants have an innate mechanism to combat the ill effects of temperature alterations. Heat shock factors (HSFs) are crucial transcription factors involved in developmental processes and various heat stress-associated rescue mechanisms. Here, we functionally characterize a highly heat stress-responsive HvHsfA6a in barley by developing constitutively overexpressing transgenic lines. The transgenic lines show a heat-tolerant phenotype via improved photosynthesis, antioxidants, and upregulation of HSPs and metabolites involved in stress

A novel oral immunotherapy strategy using transgenic barley induces Culicoides allergen-specific immune responses in horses.

INTRODUCTION: Insect bite hypersensitivity (IBH) is a seasonal, IgE-mediated allergic dermatitis of horses caused by salivary gland proteins of biting midges (Culicoides spp.). Current management relies on relief of clinical signs and on physical protection. In a previous pilot study, healthy horses were fed transgenic barley expressing a Culicoides allergen via a special spiral bit, which successfully induced allergen-specific antibody responses. Building on this concept, the present study aimed to evaluate a more practical feeding approach, delivering transgenic barley expressing the major C

Influence of yeast culture and feed antibiotics on ruminal fermentation and site and extent of digestion in beef heifers fed high grain rations1.

The study objective was to investigate the effects of site of delivering Saccharomyces cerevisiae fermentation product (SCFP) on ruminal pH and fermentation characteristics, and the site and extent of feed digestion in the digestive tract of beef heifers fed high-grain diets. Examining the ruminal and postruminal effects of SCFP is important for understanding the potential use of SCFP as an alternative for current industry-standard antibiotics used in beef cattle rations. Five beef heifers (initial BW = 561 ± 11.7 kg) equipped with ruminal and duodenal cannulas were used in a 5 × 5 Latin squar

Immunoglobulin E antibodies to ingested cereal flour components: studies with sera from subjects with asthma and eczema.

The specificity of immunoglobulin E for different cereal grain proteins was investigated using sera from twenty paediatric patients with asthma and/or eczema. Close correlations were observed between radioallergosorbent test values for grain extracts of wheat, rye and barley, and, to a lesser extent, oats. Of the different wheat four fractions tested, the globulins and glutenins consistently bound higher levels of IgE than the gliadins and albumins. This is in contrast both with bakers' asthma (an allergy to inhaled flour where the albumins are important allergens) and with coeliac disease (in

Treatment of grain with organic acids at 2 different dietary phosphorus levels modulates ruminal microbial community structure and fermentation patterns in vitro.

Recent data indicate positive effects of treating grain with citric (CAc) or lactic acid (LAc) on the hydrolysis of phytate phosphorus (P) and fermentation products of the grain. This study used a semicontinuous rumen simulation technique to evaluate the effects of processing of barley with 50.25 g/L (wt/vol) CAc or 76.25 g/L LAc on microbial composition, metabolic fermentation profile, and nutrient degradation at low or high dietary P supply. The low P diet [3.1g of P per kg of dry matter (DM) of dietary P sources only] was not supplemented with inorganic P, whereas the high P diet was supple

Identification of common released peptide and allergenic cross-reactivity of protein fractions among cereals.

Cross-reactivity between wheat and related cereals is a clinically relevant phenomenon, potentially driven by conserved digestion-resistant peptides. In this study, proteins extracted from wheat, barley, and rye were subjected to in vitro digestion and assessed by indirect enzyme-linked immunosorbent assay (ELISA) and competitive ELISA, using sera from wheat-allergic patients immunized mice, respectively. Peptides released during simulated digestion were analyzed by mass spectrometry, and the potential allergenicity of the cereals was evaluated using cell lines. Results showed high cross-react
