Effects of chemical lesion of the ventral noradrenergic bundle or the medial preoptic area on preovulatory LH release in rats.

The noradrenergic innervation of the medial preoptic area (MPO) and the hypothalamus is provided by mesencephalic neurons via the ventral noradrenergic tract. Fibers of these neurons emerge through the MPO. Bilateral microinjections of 6-OHDA into the ventral noradrenergic bundle (VNB) depletes large parts of the diencephalon of norepinephrine (NE) and dopamine (DA). Since the total hypothalamic DA content is of intrahypothalamic origin, 6-OHDA injection into the VNB does not reduce hypothalamic DA content. Similarly microinjections of 6-OHDA into the MPO reduces hypothalamic and preoptic NE c

The influence of ATP and AMP injected into the lateral ventricles of the brain on behavior in rats.

The influence of ATP and AMP injected into the lateral cerebral ventricle in doses of 50, 100 and 200 mug on behavior in rats was studied. ATP in the doses administered had no effect on behavior of rats. AMP in these doses enhanced motor activity of the rats. Chemical sympathectomy by intracerebral injection of 6-hydroxy-dopamine did not affect the action of either of the compounds on behavior of rats. AMP potentiated the action of intracerbrally administered noradrenaline on behavior of the animals.

[Gynostemma pentaphyllum ethanol extract ameliorates motor dysfunction in a Parkinson's disease mouse model through inhibiting neuronal apoptosis].

OBJECTIVES: To investigate the protective effects and underlying mechanisms of Gynostemma pentaphyllum (GP)ethanol extract on motor dysfunction in a mouse model of Parkinson's disease (PD). METHODS: Eighty C57BL/6 male mice were randomly divided into five groups: control group, model group, levodopa group (positive control group), low-dose GP group, and high-dose GP group, with 16 mice per group. The PD model was induced by injection of 6-hydroxydopamine into the substantia nigra pars reticulata of the mice. Two weeks after 6-hydroxydopamine, positive control group received intraperitoneal in

Uptake of biogenic amines by thyroid glands.

The biogenic amines, serotonin (5-HT) and norepinephrine, activate follicular cells and are endogenous thyroid constitutents. Both amines are taken up by throid lobes incubated in vitro. Uptake of 5-HT is temperature and concentration dependent, is inhibited by chlorimipramine more than by desmethylimipramine, and although it is inhibited by norepinephrine, 5-HT uptake is unaffected by chemical sympathetomy with 6-hydroxydopamine. Electron microscopic radioautography reveals that follicular cells are responsible for uptake of 5-HT and adrenergic nerves for uptake of norepinephrine. These data

Effect of subarachnoid hemorrhage on serotonin uptake and metabolism in rabbit basilar artery.

The effects of subarachnoid hemorrhage (SAH) on neuronal uptake and metabolism of serotonin (5-HT) in the rabbit basilar artery were examined. Extracted 3H-amines from the isolated arteries after incubation with [3H]5-HT were separated by column chromatography. Radioactivity of 5-HT and 5-hydroxyindoleacetic acid was, respectively, 52.7 +/- 13.9 and 22.9 +/- 5.4 x 10(2) dpm/mg tissue in the control group (n = 8); 32 and 18% of control after denervation (n = 6); 99 and 12% of control after treatment with pargyline (n = 7); and 65 and 76% of control after SAH (n = 7). These results suggest that

Alcohol intake and ethanol intoxication in the rate: effect of a 6-OHDA-induced lesion of the ascending noradrenaline pathways.

The ascending noradrenaline (NA) pathways were lesioned by injecting 6-hydroxydopamine (6-OHDA) 16 micrograms/4 microliters bilaterally into the posterior mesencephalon in male Long Evans rats. Another group of rats was pretreated with protriptyline (25 mg/kg), a NA uptake blocking agent, 15 min before they received the intracerebral injections of 6-OHDA. The controls received the vehicle only. Spectrofluorimetric determination of the catecholamine concentrations in various parts of the brain revealed a marked degeneration of the ascending NA systems in the group receiving 6-OHDA. Unexpectedly

The involvement of noradrenergic nerves in the cardiovascular reflex responses to lower body negative pressure in the anaesthetised rabbit.

1. Resting cardiovascular status and the reflex responses to lower body negative pressure (LBNP) were examined in control rabbits and in rabbits sympathectomised with 6-hydroxydopamine (6-OHDA). 2. Resting systemic arterial pressure and total peripheral resistance were significantly higher in control than in 6-OHDA treated animals. 3. Control animals responded to LBNP at -25 mm Hg with a peripheral vasoconstriction and tachycardia. These responses were absent in 6-OHDA-treated animals. 4. Control animals responded to LBNP at -70 mm Hg with a peripheral vasoconstriction and a bradycardia follow

Preclinical models of Parkinson's disease.

Parkinson's disease (PD) is a neurodegenerative disorder in which pigmented midbrain neurons progressively die producing a dopamine (DA) deficit in the striatum, which manifests as an akinetic movement disorder. Experimentally induced striatal DA depletion in animals is a valid model of parkinsonism. The capacity of certain substances to damage catecholaminergic neurons has been used extensively to produce DA deficiency in animals. This unit describes methods for inducing parkinsonism in nonhuman primates and rodents using the neurotoxins 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) and

L-DOPA inhibits spontaneous acetylcholine release from the striatum of experimental Parkinson's model rats.

Acetylcholine (ACh) release was measured by microdialysis. Addition of 10 nM L-DOPA to the perfusate significantly decreased ACh release, from the striatum of rats lesioned with 6-hydroxydopamine (6-OHDA), but not sham-operated rats. The L-DOPA-induced decrease was not affected by (-)-sulpiride which completely blocked D2- and D3-agonist-induced decrease in ACh release in lesioned rats. Neither 10 nM D-DOPA nor 100 nM dopamine caused by any change in ACh release. These findings suggest that L-DOPA-sensitive mechanisms are supersensitized in Parkinson's disease model rats.

Pharmacologic studies on the mechanism of melatonin-induced vasorelaxation in rat aorta.

The influence of several classes of drugs on the melatonin-induced vasorelaxation of isolated rat aorta were examined. Melatonin caused a dose-dependent relaxation of precontracted (30 mM KCl) aorta. This relaxant response was blocked by preincubating vessels with antagonists of vasoactive intestinal peptide. Substance P antagonists did not alter the response to melatonin. Pretreatment of vessels with 6-hydroxydopamine, lidocaine, or tetrodotoxin antagonized the relaxant response to melatonin. On the other hand, pretreatment with atropine + propranolol did not alter the response to melatonin.

Terguride, a mixed dopamine agonist-antagonist, in animal models of Parkinson's disease.

Terguride, an analogue of lisuride, decreased locomotor activity, produced cataplexy, and blocked apomorphine-induced stereotypic behavior. It did not induce stereotypies in rodents or emesis in dogs. However, in rats with unilateral 6-hydroxydopamine lesions of the substantia nigra, terguride produced contralateral rotation. Terguride bound to D-2 striatal dopamine receptors. Terguride has both agonist and antagonist actions at striatal dopamine receptors, but chronic administration did not produce behavioral supersensitivity. These pharmacologic properties differ from those of other antipark

Therapeutic gene transfer with herpes-based vectors: studies in Parkinson's disease and motor nerve regeneration.

We have examined the possibility of using herpes simplex virus (HSV)-based vectors to prevent neuronal cell death and enhance functional recovery after injury. In the 6-hydroxydopamine (6-OHDA) model of Parkinson's disease (PD) and after proximal spinal root injury, direct stereotactic injection of HSV-based vectors constructed to express the glial cell derived neurotrophic factor (GDNF) or the anti-apoptotic peptide Bcl-2 prevented neuronal death and enhanced recovery. Gene transfer may be useful in the treatment of neurologic disorders in which neuronal cell death occurs in a restricted anat

Fatty acid incorporation depicts brain activity in a rat model of Parkinson's disease.

Following pulse labeling with [3H]arachidonic acid ([3H]AA), its incorporation pattern in brain reflects regional changes in neurotransmitter signal transduction using phospholipase A2, that is, functional activity. In a rat model of Parkinson's disease, unilateral 6-hydroxydopamine lesion in the substantia nigra, [3H]AA acid incorporation from blood was increased in cerebral cortex, caudate putamen, globus pallidus, entopeduncular nucleus, subthalamic nucleus and substantia nigra pars reticulata ipsilateral to the lesion. This increased [3H]AA incorporation likely reflects disinhibition of ba

Central catecholamine, sympathetic nerve and vascular protein in the acute phase of two-kidney, one-clip renovascular hypertension in rats.

Incorporation of 3H-proline into the non-collagenous protein in mesenteric arteries in two-kidney, one-clip hypertensive rats was greater than that in normotensive rats. Splanchnicotomy predominantly over the root of mesenteric arteries or intracranioventricular injection of 6-hydroxydopamine prevented the development of hypertension in 2K-1C rats concomitant with the reduction of incorporation of 3H-proline into the non-collagenous protein in mesenteric arteries. The content of norepinephrine in the hypothalamus in 2K-1C rats was lower than that in normotensive control rats. These findings in

The effect of 6-OHDA lesions of the lateral septum on schedule-induced polydipsia.

In this study the relationship between 6-hydroxydopamine lesions of dopaminergic innervation of the lateral septum and schedule-induced polydipsia was examined. Animals with lesions were found to have a significant increase in water intake during a test involving an intermittent schedule of food delivery. Additional experiments with these animals showed that lesions had no effect on: (i) spontaneous drinking, the amount of drinking after 24 h food or water deprivation, water intake following a hypertonic saline challenge; (ii) eating behavior with or without food deprivation; and (iii) spontan

Abnormalities of cerebral oxidative metabolism in animal models of Parkinson disease.

Abnormalities of cerebral oxidative metabolism were investigated in "animal models" of Parkinson disease by in situ optical measurements of local cerebral blood volume and cytochrome oxidase redox shifts in rats two weeks after unilateral 6-hydroxydopamine lesions of the substantia nigra with or without interruption of ascending noradrenergic pathways. The data demonstrate oxidative metabolic dysfunction of ipsilateral cerebral hemispheres caused by lesions that involve both dopaminergic and noradrenergic systems but not when dopaminergic neurons only are affected. We speculate that the dement

Noradrenergic action of amphetamine following degeneration of descending monoaminergic fibers in the spinal cord.

Amphetamine and methoxamine facilitated the flexor reflex in chronic spinal rats whose descending monoaminergic nerve endings were degenerated. Methoxamine, but not amphetamine, facilitated the flexor reflex in the chronic spinal rat 16-20 h after IV 6-hydroxydopamine treatment. These data indicate that amphetamine's spinal cord facilitative effects are mediated through two noradrenergic mechanisms. In the acute spinal rat, its facilitative effects in these experiments seem to be independent of the sympathetic nervous system and are probably dependent on the presence of the terminal axons of t

Protein gene product 9.5 (PGP 9.5). A new neuronal marker visualizing the whole uterine innervation and pregnancy-induced and developmental changes in the guinea pig.

The guinea pig uterus is supplied by different populations of nerves which can be demonstrated by specific immunocytochemical and histochemical techniques. So far, there has been no single marker displaying entire peripheral innervation patterns. Recently, protein gene product (PGP) 9.5, a cytoplasmic protein in neurons and neuroendocrine cells, was found to visualize both different populations and subtypes of nerves. This prompted the present study of using PGP 9.5 for visualization of the whole uterine innervation. This was performed by the indirect immunofluorescence method using antiserum

Cholecystokinin acts through catecholaminergic mechanisms in the hypothalamus to influence ingestive behaviour in the rat.

Administration of cholecystokinin (CCK) (0.2 - 0.6 micrograms in a volume of 2 microliter) into the lateral cerebral ventricle caused a decrease in intake of food but a relative increase in intake of water (or water-to-food ratio) in rats. To determine whether the anorexic actions of CCK were mediated through the hypothalamic nuclei, rats were infused with CCK (0.02 - 0.12 microgram in a volume of 0.5 microliter) through previously implanted hypothalamic cannulae. Administration of CCK into the lateral hypothalamus, but not the anterior hypothalamus or ventromedial hypothalamus, caused decreas
