Effect of beta-endorphin on fetal breathing movements in sheep.

It has been suggested that endogenous opioids, such as beta-endorphin (beta-EP), act to depress respiration in the fetus and newborn. We have investigated the effect of infusing beta-EP either intravenously or into a lateral cerebral ventricle on breathing movements and electrocortical activity in eight fetal lambs between 116 and 133 days gestation. Intravenous infusion of beta-EP (200 or 500 micrograms over 1 h) increased plasma beta-EP concentrations 2- to 230-fold and was associated with a small decrease in the percent time spent breathing, from 57.8 +/- 9.1 to 51.3 +/- 8.2%/h (n = 6 exp).

Effect of HI-6, applied into the cerebral ventricles, on the inhibition of brain acetylcholinesterase by soman in rats.

When applied to rats (intraperitoneally) immediately after subcutaneous injection of soman (120 micrograms/kg) HI-6 (100 mg/kg) protected about 40% of the activity of acetylcholinesterase (AChE) in the motor end plate region of the diaphragm but did not protect AChE in the brain. However, a partial protection of AChE in brain against inhibition by soman was obtained in anaesthetized, atropinized rats by the oxime injected into the cerebral ventricle 5 min before parenteral exposure to soman. The AChE activity in brain of rats pretreated with HI-6, analyzed 60 min after the injection of soman w

Surgical stress inhibits physostigmine-induced pressor effect.

Physostigmine-induced pressor response was studied in adrenalectomized rats. The pressor response to intracerebroventricular administration of physostigmine was found to be inhibited in both acutely adrenalectomized and sham-operated rats, but not in animals adrenalectomized 24 h earlier. Laparatomy performed just before the experiment also inhibited the pressor effect of physostigmine. This inhibition was completely prevented by naloxone pretreatment. Endogenous opioid peptides released as a response to the stress induced by surgical manipulation, could be accounted for inhibition of the pres

Factors influencing the secretion of vasopressin into cerebrospinal fluid.

Vasopressin (VP) has been found in the cerebrospinal fluid (CSF) of several species of animals. Although it is known that hemorrhage, hypertonicity of body fluid, hypoxia, and hypercapnia all increase VP in plasma, little is known regarding the stimuli that cause the secretion of VP into the CSF. We therefore performed several studies to examine whether stimuli capable of increasing plasma levels of VP can also increase VP in the CSF of anesthetized dogs. We found that hemorrhage, intracerebroventricular infusion of hypertonic artificial CSF, hypoxia, and hypercapnia all produced increases in

On the prolactin-inhibiting effect of neurotensin. The role of dopamine.

Neurotensin (NT) when injected in a dose of 5 micrograms into the third cerebral ventricle of conscious, unrestrained male rats, outfitted with chronic third ventricle and jugular cannulae, decreased the resting prolactin (PRL) levels. NT (5 micrograms) also markedly inhibited the PRL-releasing effect of activation of central serotonin receptors by 5-hydroxytryptophan (5HTP, 15 mg/kg) in animals pretreated with fluoxetine (4 mg/kg). however, interruption of dopamine (DA) neurotransmission by either alpha-methyl-p-tyrosine (alpha MT, 250 mg/kg) or spiroperidol (0.3 mg/kg) led to a blockade of t

Inhibition by morphine and morphine-like drugs of nicotine-induced emesis in cats.

The effect of morphine, methadone and pethidine injected into the cerebral ventricle of the unanesthetized cat upon emesis produced by nicotine induced similarly was investigated. Morphine and morphine-like drugs depress or abolish the emetic effect of nicotine. The inhibitory effect of morphine, methadone and pethidine is observed after a transient emetic action of these drugs. The emetic and anti-emetic action of morphine, methadone and pethidine can perhaps be ascribed to an agonist/antagonist activity. Further, the possible site of inhibitory action of morphine and morphine-like drugs on t

Increased sodium appetite in the rat induced by intracranial administration of components of the renin-angiotensin system.

1. Intracranial injections of components of the renin-angiotensin system in rats in normal water and Na balance caused an immediate thirst followed by a progressive increase in Na appetite during a test session which lasted 18 h. The effect on water and Na intake was dose-dependent. 2. Long-term (7 day) infusions of angiotensin II into the third cerebral ventricle at rates of 1 or 10 pmol h-1 produced large and sustained increases in intake of water and 2 . 7% NaCl. Intakes sometimes exceeded 100 ml 2 . 7% NaCl per day but quickly fell to normal when the infusion was stopped. 3. Intracranial i

Central blood pressure effects of guanylyl-imido-diphosphate and cyclic guanosine-monophosphate.

Injections of guanylyl-imido-diphosphate (250, 500 and 1,000 microgram/kg) into the lateral cerebral ventricle of the anaesthetized cat induced increases in blood pressure and heart rate while the intravenous injections of the same doses were ineffective, thus indicating a central mechanism of action of this compound which activates adenylcyclase at the catalytic subunit. The results support the hypothesis that the activity of cardiovascular centres depends on the prevailing concentration of cAMP. Intracerebroventricular injection of cGMP (125, 250 and 500 microgram/kg) caused hypotension and

Comparison of the thermoregulatory responses to intracerebroventricularly injected dopamine, noradrenaline and 5-hydroxytryptamine in the goat.

The thermoregulatory responses to dopamine (DA), noradrenaline (NA) and 5-hydroxytryptamine (5-HT), injected into the third cerebral ventricle of goats at 20 degrees C ambient temperature (Ta) or during cold exposure, were compared before and after pretreatment with the DA receptor blocker haloperidol or the 5-HT receptor blocker methysergide. At 20 degrees C Ta, intracerebroventricular (i.c.v.) injection of DA (800 microgram), NA (200 microgram) or 5-HT (800 microgram) induced a decrease in body temperature (Tb) and dilatation of the ear vessels. The influence of ATP and AMP injected into the lateral ventricles of the brain on behavior in rats.

The influence of ATP and AMP injected into the lateral cerebral ventricle in doses of 50, 100 and 200 mug on behavior in rats was studied. ATP in the doses administered had no effect on behavior of rats. AMP in these doses enhanced motor activity of the rats. Chemical sympathectomy by intracerebral injection of 6-hydroxy-dopamine did not affect the action of either of the compounds on behavior of rats. AMP potentiated the action of intracerbrally administered noradrenaline on behavior of the animals.

Cholinergic anti-inflammatory pathway in intracerebral hemorrhage.

Stimulated vagus nerve excretes acetylcholine into the peripheral immune organs such as the spleen, reducing innate inflammation. Here, we investigated whether activation of this "cholinergic anti-inflammatory pathway" can be used to reduce cerebral inflammation in a model of hemorrhagic stroke. Experimental intracerebral hemorrhage (ICH) was induced by stereotaxic collagenase injection in rats. Muscarine, an activator of the vagus nerve, or phosphate-buffered saline (control) was injected into the lateral ventricle after induction of ICH. Intraventricular muscarine injection increased heart r

Effects of systemic administration of 6-hydroxydopamine on the circumventricular organs in nonhuman primates. I. Area postrema.

6-Hydroxydopamine (6-OH-DA) has been shown to produce degenerative changes in noradrenergic nerve terminals and preterminals in the CNS following intracisternal, intraventricular or direct injection into the brain parenchyma. Systemic injection of 6-OH-DA is known to result in degenerative changes in noradrenergic terminals in the peripheral nervous system. However, only a few studies have been carried out on the effects of systemic injections of 6-OH-DA on noradrenergic terminals in the CNS. In the present study cynomolgus and squirrel monkeys were injected intravenously on two successive day

Central mediators of the zymosan-induced febrile response.

BACKGROUND: Zymosan is a fungal cell wall protein-carbohydrate complex that is known to activate inflammatory pathways through the Toll-like receptors and is commonly used to induce fever. Nevertheless, the central mediators that are involved in the zymosan-induced febrile response are only partially known. METHODS: The present study evaluated the participation of prostaglandins, substance P, endothelin-1 (ET-1), and endogenous opioids (eOPs) in the zymosan-induced febrile response by using inhibitors and antagonists in male Wistar rats. RESULTS: Both nonselective (indomethacin) and selectiv

Melatonin protects against amyloid-β-induced impairments of hippocampal LTP and spatial learning in rats.

Alzheimer's disease (AD), the most prevalent neurodegenerative disease in the elderly, leads to progressive loss of memory and cognitive deficits. Amyloid-β protein (Aβ) in the brain is thought to be the main cause of memory loss in AD. Melatonin, an indole hormone secreted by the pineal gland, has been reported to produce neuroprotective effects. We examined whether melatonin could protect Aβ-induced impairments of hippocampal synaptic plasticity, neuronal cooperative activity, and learning and memory. Rats received bilateral intrahippocampal injection of Aβ1-42 or Aβ31-35 followed by intrape

Spinal 111Indium-DTPA CSF flow studies in leptomeningeal metastasis.

Sixteen consecutive patients (8 men; 8 women; age range 6-63 years, median 40 years) with leptomeningeal metastasis were found by radionuclide ventriculography to have cerebrospinal fluid (CSF) flow interruption at the following sites: cisterna magna/basal cisterns (6); cervical (5) and thoracic (3) spinal subarachnoid space. Two additional patients had no evidence of interruption of CSF flow. All patients underwent intralumbar injection of 111Indium-DTPA and imaging of spinal ascent of radionuclide, corroborating sites of CSF flow disruption seen by radionuclide ventriculography. Effects of adrenoceptor antagonists on the hyperthermia and hyperglycemia induced by prostaglandin F2 alpha in rats.

We investigated the effects of intraperitoneal administration of adrenoceptor antagonists to the hyperthermia and hyperglycemia induced by prostaglandin F2 alpha (50 micrograms) injected into the third cerebral ventricle in anesthetized rats. Phentolamine inhibited the hyperthermia and hyperglycemia induced by prostaglandin F2 alpha. Prazosin inhibited the hyperthermia induced by prostaglandin F2 alpha, while enhancing the hyperglycemia. Yohimbine inhibited the prostaglandin F2 alpha-induced hyperglycemia without an effect on the hyperthermia. Propranolol had no effect on either prostaglandin

Inhibition of acute inflammation in the periphery by central action of salicylates.

Understanding of the antiinflammatory actions of nonsteroidal drugs is incomplete, but these actions are believed to occur in the periphery, without any contribution from the central nervous system. Recent research on the antipyretic antiinflammatory neuropeptide alpha-melanocyte-stimulating hormone indicates that it can act centrally to inhibit peripheral inflammation; this raises the possibility that other agents, such as nonsteroidal antiinflammatory drugs, may have similar activity. In the present research both lysine acetylsalicylate and sodium salicylate inhibited edema, induced in the m

Cholinergic mechanisms in the rat's hypothalamus mediate the stimulatory effect of thyrotropin-releasing hormone on gastric secretion.

To assess the possible involvement of cholinergic mechanisms in the hypothalamic nuclei in the stimulatory effect of TRH on gastric secretion, rats were infused with thyrotropin-releasing hormone (TRH), cholinergic agonist or antagonist, and normal saline through previously implanted hypothalamic cannulae. Administration of TRH or pilocarpine into the lateral cerebral ventricle or the anterior hypothalamus caused a dose-related increase in gastric volume and acidity in rats. On the other hand, administration of either atropine or D-tubocurarine into the same brain sites caused the opposite eff

Central opioid mechanisms and cardiovascular control in hemorrhagic hypotension.

Bleeding volume during standardized hemorrhagic hypotension was studied in anesthetized, artificially ventilated rats after specific stimulation or blockade of opiate receptor systems. Hypotension was produced by stepwise bleeding to 80, 60, and 40 mmHg systemic arterial pressure. Subcutaneous administration of morphine 15 min before the bleeding resulted in a decrease, whereas subcutaneous administration of naloxone resulted in a significant increase in the required bleeding volume. Intracerebroventricularly administered met-enkephalin, dynorphin, or specific antisera against these peptides c
