Hydrogen sulfide plays an important role by regulating endoplasmic reticulum stress in myocardial diseases.

Endoplasmic reticulum (ER) is an important organelle for protein translation, folding and translocation, as well as the post-translational modification and assembly of newly synthesized secreted proteins. When the excessive accumulation of misfolded and/or unfolded proteins exceeds the processing capacity of ER, ER stress is triggered. The integrated intracellular signal cascade, namely the unfolded protein response, is induced to avoid ER stress. ER stress is involved in many pathological and physiological processes including myocardial diseases. For a long time, hydrogen sulfide (H2S) has be

Phytohormone-microbiome interactions and epigenetic regulation of plant stress responses and priming.

Plants continuously face fluctuating environmental conditions, requiring tightly coordinated regulatory systems to balance growth and stress responses. This review synthesizes current knowledge on phytohormones as central integrators of plant-microbiome interactions, highlighting their dual role as internal regulators and ecological gatekeepers that shape microbiome assembly and function. Phytohormones, such as auxins, cytokinins, gibberellins, abscisic acid, ethylene, salicylic acid, and jasmonates, dynamically regulate plant development, immunity, and rhizosphere chemistry, thereby influenci

Integrating Plant Physiology and Microbiome Engineering for Climate-Resilient Crops: Bridging Knowledge Gaps in Multi-Stress Tolerance.

Climate change is intensifying the frequency and co-occurrence of abiotic and biotic stresses, posing significant challenges to global crop productivity and stability. Conventional approaches based on single-stress responses are increasingly insufficient for addressing complex field environments where plants experience multiple simultaneous stresses. This review synthesizes current knowledge on plant physiological responses, microbiome interactions, and emerging technological interventions to develop an integrative framework for climate-resilient agriculture. It highlights how stress perceptio

Decoding Short- and Long-Term Cellular Adaptations to Cr(VI) Exposure Through High-Throughput Transcriptomics.

Hexavalent chromium (Cr(VI)) is a well-established environmental and occupational carcinogen, but its time-dependent molecular effects remain poorly characterized. This study aims to elucidate the transcriptional responses triggered by acute versus chronic Cr(VI) exposure through an integrated analysis of two publicly available transcriptomic datasets: GSE16349 (short-term exposure, 16 hours) and GSE24025 (long-term exposure, 4 weeks). We identified 250 differentially expressed genes across both exposure models. MetaCore pathway enrichment analysis revealed shared activation of apoptosis, surv

Huntingtin (HTT) interactome in regulation of DNA repair/remodeling and RNA processing pathways.

Huntington's disease (HD), an uncurable neurodegenerative disorder, is caused by CAG repeat expansion in the HD gene encoding mutant huntingtin protein. DNA damage response is implicated in HD pathogenesis. We used multiple approaches to assess normal and mutant HTT interactomes in the context of genotoxic stress. We show that double-strand break (DSB) repair response is impaired in HD neurons, which are more vulnerable to DSB-induced stress. We found that S1181 phosphorylation of HTT is regulated by DSB, and can be carried out by DNA-PK. Functional interaction of HTT with a major DSB kinase D

Adverse Childhood Events, Post-Traumatic Stress Disorder, Infectious Encephalopathies and Immune-Mediated Disease.

Adverse Childhood Events (ACE), Post-Traumatic Stress Disorder (PTSD), and infectious encephalopathies are associated with immune-mediated diseases. Data supporting this are reviewed, and an integrated hypothesis is provided. All three can be associated with intrusive symptoms and temporal lobe pathology. ACE and PTSD are accompanied by an impaired mental capacity to differentiate external danger vs. safety. Infectious encephalopathies are accompanied by a failure of adaptive immunity and an impaired immune capacity to differentiate internal danger vs. safety. All three conditions are associat

Seed priming-mediated molecular responses enhance thermo-osmotic resilience in Halophila ovalis for climate-resilient restoration.

Marine heatwaves pose significant threats to seagrass ecosystems, particularly during early life stages where recruitment governs population stability. In this study, we investigated propagule quality and stress tolerance mechanisms in the tropical seagrass Halophila ovalis by integrating density-based seed selection, antimicrobial storage, thermal priming and gene expression analysis. Density-gradient fractionation revealed clear stratification of seed quality, with high-specific-gravity seeds (SG > 1.20) exhibiting 78% germination compared to <12% in low-density fractions, indicating strong

SnRNA-seq and genome-wide CRISPR screening define the complete transcriptional trajectory and functional drivers of podocyte stress.

Podocyte injury is a central driver of proteinuria and progressive kidney dysfunction. Although podocytes are continuously exposed to diverse stressors in both physiological and pathological contexts, the dynamic processes underlying their adaptation and eventual failure remain poorly defined. Here, we performed integrative single-nucleus RNA sequencing of kidney tissues from patients with six types of representative chronic glomerulonephritis, capturing a spectrum of podocyte injury states. We identified distinct podocyte subpopulations and reconstructed a dynamic trajectory characterized by

Functions of Basic Helix-Loop-Helix (bHLH) Proteins in the Regulation of Plant Responses to Cold, Drought, Salt, and Iron Deficiency: A Comprehensive Review.

Abiotic stresses including cold, drought, salt, and iron deficiency severely impair plant development, crop productivity, and geographic distribution. Several bodies of research have shed light on the pleiotropic functions of BASIC HELIX-LOOP-HELIX (bHLH) proteins in plant responses to these abiotic stresses. In this review, we mention the regulatory roles of bHLH TFs in response to stresses such as cold, drought, salt resistance, and iron deficiency, as well as in enhancing grain yield in plants, especially crops. The bHLH proteins bind to E/G-box motifs in the target promoter and interact wi

OMA1-Mediated Mitochondrial Dynamics Balance Organellar Homeostasis Upstream of Cellular Stress Responses.

In response to cellular metabolic and signaling cues, the mitochondrial network employs distinct sets of membrane-shaping factors to dynamically modulate organellar structures through a balance of fission and fusion. While these organellar dynamics mediate mitochondrial structure/function homeostasis, they also directly impact critical cell-wide signaling pathways such as apoptosis, autophagy, and the integrated stress response (ISR). Mitochondrial fission is driven by the recruitment of the cytosolic dynamin-related protein-1 (DRP1), while fusion is carried out by mitofusins 1 and 2 (in the o…

Multiple Roles of G3BP1 in Regulating STING-Dependent Interferon and Cytokine Induction by Cytosolic dsDNA and HSV-1 Infection.

Virus infection requires coordinated activation of pathogen-sensing, innate immune, and cellular stress response pathways to mount an effective antiviral defense. Recognition of nucleic acid pathogen-associated molecular patterns (PAMPs) by pattern recognition receptors (PRRs) initiates signaling cascades that drive the production of type I interferons (IFNs) and proinflammatory cytokines. These responses are often accompanied by the activation of integrated stress response pathways that help optimize host defense. Cytosolic double-stranded dsDNA, generated during viral infection or released f…

HOG Signaling: A Multifunctional Regulator of Stress Homeostasis in Saccharomyces cerevisiae.

During growth and metabolism, the budding yeast Saccharomyces cerevisiae is continuously exposed to diverse environmental stresses, and conserved mitogen-activated protein kinase (MAPK) cascades execute core functions in stress sensing and adaptive regulation. The high-osmolarity glycerol (HOG) pathway, a classical MAPK cascade in S. cerevisiae, was initially identified as the core regulator of hyperosmotic stress responses. Over recent decades, accumulating evidence has revealed that the HOG pathway is not merely an osmoregulatory module but a versatile signaling hub that integrates multiple

Crosstalk of heat shock proteins and antioxidants with peroxisome biogenesis supports wheat thermotolerance.

Rising temperatures globally impose coupled proteotoxic and oxidative stresses that limit spring wheat (Triticum aestivum) productivity, yet how protein quality control, redox homeostasis, and peroxisome dynamics are coordinately regulated during thermotolerance remains unclear. Here, we investigated the integrated roles of heat shock proteins (HSPs), antioxidant defenses, and peroxisome biogenesis in two contrasting wheat genotypes differing in heat stress tolerance at anthesis: Misr2 (tolerant) and Line4 (susceptible). Heat responses were characterized by osmoprotectant accumulation (total s

Hypothalamic Regulation of Cardiorespiratory Functions: Insights into the Dorsomedial and Perifornical Pathways.

The dorsomedial hypothalamus nucleus (DMH) plays a pivotal role in the orchestration of sympathetic nervous system activities. Through its projections to the brainstem and pontomedullary nuclei, it controls heart rate, contractility, blood pressure, and respiratory activity, such as timing and volumes. The DMH integrates inputs from higher brain centers and processes these signals in order to modulate autonomic outflow accordingly. It has been demonstrated to be of particular significance in the context of stress responses, where it orchestrates the physiological adaptations that are necessary

Early life stress exposure alters brain vasculature transcriptomic profiles in areas regulating stress resilience.

Early life stress (ELS) events during sensitive postnatal time periods can recalibrate future stress responsiveness and precipitate mental disorders. Neurovascular adaptations can influence cognition, mood, and stress responses. Disruption of blood-brain barrier (BBB) integrity, which is formed by endothelial cells, astrocytes, and pericytes, has been implicated in affective disorders such as depression, which often arise from chronic stress experiences. Despite the BBB undergoing critical maturation stages during development, it remains poorly known how ELS influences brain vascular function,
