Interferon-alpha is an effective agent in the treatment of chronic myelogenous leukaemia and hairy cell leukaemia. Because interferon-alpha does not inhibit DNA synthesis, we have used it to treat these diseases during pregnancy. We report here two patients with chronic myelogenous leukaemia and two patients with hairy cell leukaemia managed with interferon-alpha during pregnancy. This paper aims to summarize current experience with alpha interferon and provide direction for future study. There are four areas in which alpha interferon has proven or potential activity: antiviral, premalignant, adjuvant and advanced disease settings. In a series of studies, recombinant interferon-alpha 2a (rIFN alpha 2a, Roferon-A) was administered alone (273 men) or combined with vinblastine (91 men) to patients with acquired immunodeficiency syndrome (AIDS)-related Kaposi's sarcoma (KS). Ten patients suffering from discoid lupus erythematosus (DLE) or subacute cutaneous lupus erythematosus (SCLE) were treated with interferon alpha 2a. A marked improvement or clearing of cutaneous lupus erythematosus lesions was observed in eight of them. Tumour necrosis factor (TNF) induces the lysis of many malignant cells in vitro and regression of some tumours in vivo. 15 patients with myeloproliferative diseases and thrombocythaemia (7 Polycythaemia vera, 5 essential thrombocythaemia, 3 chronic myelogenous leukaemia) were assigned to a therapy with recombinant interferon (recombinant IFN-alpha-2C) in a prospective, controlled trial. Under therapy all patients showed a significant decrease in thrombocyte values. 51% of the patients revealed thrombocyte values within the normal range after 3 months of IFN therapy. Noted side effects of IFN therapy were dose-dependent and clinically well tolerated by the patients.

PURPOSE: Protein aggregates are thought to be involved in the immunogenicity of recombinant proteins in humans. To probe human IFN-alpha formulations for the presence of soluble protein aggregates, enzyme-linked immunosorbent assays (ELISA) were developed.

BACKGROUND: Recent genome-wide association studies revealed the rs12979860 single nucleotide polymorphism (SNP) of the IL28B gene (CC genotype) to be the strongest pre-therapeutic predictor of therapy response to interferon alpha in patients with chronic hepatitis C infection.
