Host biological sex directs immune control of the Plasmodium parasite liver stage in mice.

INTRODUCTION: Plasmodium parasites, the causative agents of malaria, undergo a complex liver stage shaped by the spatial heterogeneity of th … epatocytes by 44 hours post-infection. At homeostasis, intact males had lower baseline densities of Kupffer cells, monocytes, and dendritic cells, a disparity that persisted and increased during infection. Although infection induced similar fold increases in Kupffer cell numbers across groups, intact males had reduced absolute numbers of Kupffer cells, diminish …

Therapeutic Targeting of Hepatic Macrophages for the Treatment of Liver Diseases.

Hepatic macrophages play a central role in maintaining homeostasis in the liver, as well as in the initiation and progression of liver diseases. Hepatic macrophages are mainly derived from resident hepatic macrophages called Kupffer cells or circulating bone marrow-derived monocytes. Kupffer cells are self-renewing and typically non-migrating macrophages in the liver and are stationed in the liver sinusoids in contrast to macrophages originating from circulating monocytes. Kupffer cells regulate liver homeostasis by mediating immunity against non-pathogenic blood-borne molecules, while partici…

Does laparoscopic cholecystectomy influence peri-sinusoidal cell activity?

BACKGROUND/AIMS: To investigate the influence of laparoscopic procedures on perisinusoidal cell function. METHODOLOGY: In 31 patients who underwent laparoscopic cholecystectomy for symptomatic cholelithiasis, the serum levels of beta-N-acetyl hexosaminidase (beta-NAH) and hyaluronic acid (HA) were measured. Six female patients, who underwent other laparoscopic procedures, not involving the hepatobiliary system, served as controls. RESULTS: HA serum levels increased significantly in both study and control groups, while beta-NAH serum levels remained within normal values. Post-operative AST an

CD14 and lipopolysaccharide binding protein expression in a rat model of alcoholic liver disease.

Lipopolysaccharide-binding protein (LBP) and CD14 play key intermediary roles in the activation of cells by endotoxin. As endotoxin has been postulated to participate in promoting pathological liver injury in alcoholic liver disease, we investigated the role of LBP and CD14 in alcoholic liver injury. Rats were fed intragastrically ethanol or dextrose and either medium-chain triglycerides, corn oil, or fish oil for 4 weeks. Kupffer cells, endothelial cells, and hepatocytes were isolated. LBP and CD14 mRNA levels were measured in liver and individual cell types. The highest levels of LBP and CD1…

Intrahepatic targeting and sustained release of a Toll like receptor 8 agonist enhances hepatocellular carcinoma therapy.

Hepatocellular carcinoma (HCC) remains a major global health challenge with limited therapeutic options. Liver-resident Kupffer cells, which exhibit strong phagocytic activity and predominantly adopt an immunosuppressive M2 phenotype in the tumor microenvironment, not only promote tumor immune escape but also rapidly clear conventional drug delivery systems, posing a fundamental barrier to effective treatment. Herein, we present a novel strategy that uses these cells as both barriers and therapeutic anchors. We developed a CD47-mimetic self-peptide-modified liposomal system that targets SIRPα …

Acute hepatotoxicity of Pseudomonas aeruginosa exotoxin A in mice depends on T cells and TNF.

The most potent virulence factor of Pseudomonas aeruginosa, its exotoxin A (PEA), inhibits protein synthesis, especially in the liver, and i … alization of TNF or lack of TNF receptors prevented liver injury. In the liver, TNF was produced by Kupffer cells before hepatocellular death occurred. After T cell depletion, Kupffer cells failed to produce TNF. Transaminase release was significantly reduced in perforin knockout mice, and it was even elevated in lpr/lpr mice. These results demonstrate that PEA …

CARD9 orchestrates tissue damage in APAP-induced hepatitis via TREM2-mediated sensing cell death.

During the progression of acetaminophen (N-acetyl-para-aminophenol [APAP])-induced liver injury, the innate immune response is implicated in … hepatocytes, followed by innate immune activation. Our findings reveal that CARD9 expression within Kupffer cells exacerbates liver damage by amplifying the production of proinflammatory factors, which are known to play a pathological role in noninfectious conditions. Furthermore, our study identifies TREM2 receptors on Kupffer cells as capable of recognizing c …

Alcoholic liver disease.

Research has substantiated the role of several mechanisms responsible for alcohol-induced hepatotoxicity. These mechanisms include: oxidative stress and lipid peroxidation; immunogenic processes initiated by formation of protein adducts of acetaldehyde, other aldehydes and 1-hydroxyethyl radicals; and activation of Kupffer cells by endotoxin and subsequent cascade of events that involved cytokines, chemokines, and adhesion molecules. Increasing evidence implicates enhanced intestinal permeability caused by alcohol ingestion as the culprit that leads to endotoxemia. While oxidative stress is im…

Pivotal role of liver sinusoidal endothelial cells in NAFLD/NASH progression.

Liver sinusoidal endothelial cells (LSECs) are involved in the transport of nutrients, lipids, and lipoproteins, and LSEC injury occurs in v … a. LSEC injury appeared during the simple steatosis phase, and preceded the appearance of activated Kupffer cells and hepatic stellate cells (HSCs). Gadolinium chloride pretreatment ameliorates acute cadmium-induced hepatotoxicity.

Cadmium is a known industrial and environmental pollutant. It causes hepatotoxicity upon acute administration. Features of cadmium-induced acute hepatoxicity encompass necrosis, apoptosis, peliosis and inflammatory infiltration. Gadolinium chloride (GdCl3) may prevent cadmium-induced hepatotoxicity by suppressing Kupffer cells. The effect of GdCl3 pretreatment on a model of acute cadmium-induced liver injury was investigated. Male Wistar rats 4-5 months old were injected intraperitoneally with normal saline followed by cadmium chloride (CdCl2; 6.5 mg/kg) or GdCl3 (10 mg/kg) followed by CdCl2 (…

The immunology of fibrogenesis in alcoholic liver disease.

CONTEXT: Alcoholic liver disease in humans frequently leads to cirrhosis. Experimental models of hepatic fibrogenesis are available, but extrapolation of those findings to human ethanol-induced liver injury is difficult. Hepatic ethanol-induced fibrosis in humans has often been studied in relatively small patient populations. During the past decade, several animal models and human studies have attributed fibrogenesis in the liver to the role played by hepatocytes, Kupffer cells, endothelial cells, and especially stellate cells. OBJECTIVE: To determine the contribution of the main liver cell t…

Periodontitis Induces B Cell-Macrophage Crosstalk to Exacerbate Glucose Dysregulation in Obesity.

A growing consensus indicates that periodontitis (PD) adversely affects glycemic control in type 2 diabetes, though its underlying mechanisms remain unclear. Here, we report that PD, induced by long-term oral ligatures, significantly aggravated hyperglycemia and hepatic gluconeogenesis in high-fat diet-induced obese mice, with its impact on insulin resistance being more pronounced in the liver than in adipose or muscle tissue. Immunologically, PD increased systemic B cell abundance, promoted hepatic B2 cell mobilization, facilitated the expansion of Kupffer cells and adipose tissue macrophages…
