An SMC-domain protein regulates eupyrene spermiogenesis and sperm migration in a cosmopolitan insect, Plutella xylostella.

Reproductive capacity is a key determinant of the global success of agricultural insect pests and depends fundamentally on efficient spermatogenesis. In Lepidoptera, however, the molecular mechanisms regulating sperm development and migration remain largely unknown. Here, we functionally characterized an SMC-domain-containing gene, PxSMC, in the diamondback moth, Plutella xylostella. PxSMC exhibited strong testis-specific expression, suggesting a role in male reproductive development. CRISPR/Cas9-mediated disruption of PxSMC caused complete male sterility without affecting female fertility. Mu…

Determination of Cyclaniliprole in Fruits and Vegetables Using Disposable Pipette Extraction Cleanup and Ultrahigh-Performance Liquid Chromatography-Tandem Mass Spectrometry.

Despite an outstanding agent for control of Lepidoptera, the diamide insecticide cyclaniliprole (CYCP) is a suspected carcinogen. In the present study, an analytical method was developed for the determination of CYCP in six fruits and vegetables (apple, grape, peach, bell pepper, lettuce, and tomato) using ultrahigh-performance liquid chromatography coupled with tandem mass spectrometry. Sample preparation was carried out by the acetonitrile-salting-out extraction followed by simple and fast cleanup of disposable pipette extraction tip containing styrene divinyl benzene and/or graphitized carb…

Laboratory and field tests of spray-dried and granular formulations of a Bacillus thuringiensis strain with insecticidal activity against the sugarcane borer.

Formulations of Bacillus thuringiensis Berliner (Bt) with insecticidal activity against the sugarcane borer, Diatraea saccharalis Fabricius (Lepidoptera: Pyralidae), were developed and tested under laboratory and field conditions. The formulations were prepared using biodegradable polymers such as modified corn starch as an encapsulating agent, gelatin as an adherent, powdered sugarcane as a feeding stimulant and a Bt var. kurstaki GM-34 strain from a non-sugarcane region as the active ingredient. The spore-crystal complex of this strain was mixed at three different concentrations (30, 70 and …

Cryptococcus neoformans capsular enlargement and cellular gigantism during Galleria mellonella infection.

We have studied infection of Cryptococcus neoformans in the non-vertebrate host Galleria mellonella with particular interest in the morphological response of the yeast. Inoculation of C. neoformans in caterpillars induced a capsule-independent increase in haemocyte density 2 h after infection. C. neoformans manifested a significant increase in capsule size after inoculation into the caterpillar. The magnitude of capsule increase depended on the temperature, being more pronounced at 37°C than at 30°C, which correlated with an increased virulence of the fungus and reduced phagocytosis at 37°C. C

Altering the fate of stem cells from midgut of Heliothis virescens:the effect of calcium ions.

Cultured stem cells from larval midgut tissue of the lepidopteran Heliothis virescens respond to alterations in external calcium ion concentration (Ca(2+) (out)) by changing the rate of stem cell proliferation and by differentiating to larval or non-larval phenotypes. Decreasing the external concentration of Ca(2+) with the Ca(2+) chelating agent EGTA increased proliferation of stem cells in culture, and doubled the proportion of cells differentiating to columnar and goblet cells typical of larval midgut compared to controls. In contrast, increasing inward transport of Ca(2+) into the cells by…

Vertical transmission of TnSNPV, TnCPV, AcMNPV, and possibly recombinant NPV in Trichoplusia ni.

Four viruses were tested for vertical transmission in Trichoplusia ni: T. ni nucleopolyhedrovirus (TnSNPV), T. ni cypovirus (TnCPV), Autographa californica nucleopolyhedrovirus (AcMNPV), and AcMNPV engineered to express a scorpion toxin (AcMNPV.AaIT). Fifth instars were exposed to each virus, the survivors were reared and mated, and second-generation (F(1)) insects were examined for infection. TnSNPV was transmitted to offspring at a prevalence rate of 15.4%, TnCPV at 10.2%, and AcMNPV at 10.1%. Only one of 2484 F(1) insects was infected with AcMNPV.AaIT; this experiment was repeated, and none

Facilitated diacylglycerol exchange between insect hemolymph lipophorins. Properties of Manduca sexta lipid transfer particle.

Manduca sexta hemolymph lipid transfer particle (LTP) is a very high density lipoprotein (d = 1.23 g/ml) containing 14% lipid and 5% carbohydrate. Each of three apoprotein components, apoLTP-I (Mr approximately 320,000), apoLTP-II (Mr = 85,000), and apoLTP-III (Mr = 55,000), is glycosylated. Carbohydrate analysis revealed the presence of mannose and N-acetylglucosamine in a ratio of 4.5:1. A native Mr greater than 670,000 was determined by pore limiting gradient gel electrophoresis. Lipid analysis of LTP revealed the presence of phospholipid, diacylglycerol (DAG), free fatty acid, and triacylg

Selection of organosilicone surfactants for tank-mixed pesticides considering the balance between synergistic effects on pests and environmental risks.

In this study, the bioactivities of binary mixtures of organosilicone surfactants and indoxacarb against two Lepidopteran pests were investigated along with their environmental risks. All of the tested organosilicone surfactants had obvious synergistic effects on the contact toxicity of indoxacarb against Spodoptera exigua and Agrotis ipsilon. However, all of the organosilicone surfactants exhibited certain antagonism for indoxacarb against S. exigua in terms of stomach & contact toxicity; both Silwet-408 and Silwet-806 exhibited additivity against A. ipsilon, whereas Silwet-618 and Silwet-DRS…

Purification of Recombinant Human Tyrosinase from Insect Larvae Infected with the Baculovirus Vector.

The purification of an enzyme from insect larvae infected with a baculovirus vector is described. The enzyme tyrosinase is of biomedical importance and catalyzes the first rate-limiting steps in melanin production. Tyrosinase mutations can result in oculocutaneous albinism type 1 (OCA1), an inherited eye disease associated with decreased melanin pigment production and vision defects. To simplify expression and subsequent purification, the extracellular domain is expressed in insect cells, produced in Trichoplusia ni larvae, and purified using affinity and size-exclusion chromatography. Comprehensive physiology and toxicology of ecdysogens--The metabolically activated porphyrin-ecdysteroid complexes in insects.

The polyhydroxylated derivatives of 6-keto,7-dehydrocholesterol (ecdysone, ecdysteroids, Ecd) are natural compounds widely distributed in plants. They exhibit strong anabolic, vitamin D-like, pharmacological effects in vertebrate animals and in the human body. In the larval stages of insects, injections of pure Ecd cause serious pathophysiological, "hyperecdysonic" syndromes associated with neuromuscular paralysis, premature cuticular apolysis and complete inhibition of ecdysis. Ecds do not penetrate insect cuticle. For this reason, all previous attempts to induce ecdysone responses by topical

Tropical dermatology: Venomous arthropods and human skin: Part I. Insecta.

Although many tropical insects carry infectious diseases, cutaneous injury can occur by other mechanisms, for example erucism (envenomation by caterpillars) or lepidopterism (dermatitis from moths). Pararama is a unique form of erucism seen in workers in contact with rubber trees in the Amazon, and it is caused by Premolis larvae, resulting in progressive periarticular fibrosis, ankylosis, and the loss of articulation. Ants and aquatic insects of the Belostomatidae family can cause painful bites and stings. Anaphylactic shock and death can result from the venom of bees and wasps. Beetles can c

[Intoxications specific to the Aquitaine region].

Some intoxications are more specifically linked to the Aquitaine region than to other regions of France, due to environmental circumstances (fauna, flora, climate) or traditional activities (gastronomy). Three types of intoxications are particular in this area. Pine processionary caterpillar envenomations (Thaumetopoea pityocampa), a Southern Europe pinewood parasite, are frequently encountered in the Landes' forest. They are responsible of ocular and/or skin lesions with urticaria or contact dermatitis, seldom associated with immediate IgE hypersensitivity. According to the south Atlantic coa

Food quality, competition, and parasitism influence feeding preference in a neotropical lepidopteran.

We surveyed Lepidoptera found on 11 species of Inga (Fabaceae:Mimosoideae) co-existing on Barro Colorado Island, Panama, to evaluate factors influencing diet choice. Of the 47 species of caterpillars (747 individuals) recorded, each fed on a distinct set of Inga. In the field, 96% of the individuals were found on young leaves. Growth rates of caterpillars that were fed leaves in the laboratory were 60% higher on young leaves compared to mature leaves. When caterpillars were fed leaves of nonhost Inga, they grew more slowly. These data provide support for a link between preference and performan…

Comparative venom toxicity between Pteromalus puparum and Nasonia vitripennis (Hymenoptera: Pteromalidae) toward the hemocytes of their natural hosts, non-target insects and cultured insect cells.

Crude venoms from two parasitoid species, Pteromalus puparum and Nasonia vitripennis (Hymenoptera: Pteromalidae) were assayed for biological activities toward hemocytes from two species of their natural hosts and eight species of their non-natural hosts as well as two lines of cultured Lepidoptera cells, respectively. By inhibiting the spreading and viability of insect hemocytes, the venom from P. puparum displayed significantly higher activities toward plasmatocytes and granular cells from both larvae and pupae of two natural hosts, Pieris rapae and Papilio xuthus, and lower activity toward t…

Evidence for the independent evolution of a rectal complex within the beetle superfamily Scarabaeoidea.

Rectal or cryptonephridial complexes have evolved repeatedly in arthropods, including in beetles where they occur in ∼190,000 species of Cucujiformia + Bostrichoidea, and Lepidoptera where they occur in ∼160,000 species. Sections of the Malpighian/renal tubules coat the outer surface of the rectum, acting as powerful recycling systems of the gut contents, recovering water and specific solutes. There are hints that a rectal complex evolved independently within another beetle group, Scarabaeoidea. Here we report our observations of rectal complexes in Scarabaeoidea, which support this view. We d…

An Overview of Ovarian Calyx Fluid Proteins of Toxoneuron nigriceps (Viereck) (Hymenoptera: Braconidae): An Integrated Transcriptomic and Proteomic Approach.

The larval stages of the tobacco budworm, Heliothis virescens (Fabricius) (Lepidoptera: Noctuidae), are parasitized by the endophagous parasitoid wasp, Toxoneuron nigriceps (Viereck) (Hymenoptera: Braconidae). During the injections of eggs, this parasitoid wasp also injects into the host body the secretion of the venom gland and the calyx fluid, which contains a polydnavirus (T. nigriceps BracoVirus: TnBV) and the Ovarian calyx fluid Proteins (OPs). The effects of the OPs on the host immune system have recently been described. In particular, it has been demonstrated that the OPs cause hemocyte…
