Active site stoichiometry of L-phenylalanine: tRNA ligase from Escherichia coli K(-10).

The existence of two active siter per molecule of L-phenylalanine:tRNA ligase from Escherichia coli K(-10) has been demonstrated by isolation of the E-aminoacyl adenylate and tel filtration and the nitrocellulose filter assay at pH 5.0 revealed the same stoichiometry for the E-tRNAPhe comples as protection against degradation by snake venom phosphodiesterase and equilibrium gel filtration at pH 7.5. Using a fluorescence titration technique, it was found that the dissociation constant for ligase-tRNAPhe complex is decreased 20-fold when the hydrogen ion concentration is changed from pH 6.0 to p

The gene expression and activity of calpains and the muscle wasting-associated ubiquitin ligases, atrogin-1 and MuRF1, are not altered in patients with primary hyperparathyroidism.

Hyperparathyroidism (HPT) can be associated with muscle atrophy and weakness. Muscle atrophy is typically caused by increased muscle protein breakdown. The influence of HPT on calpains and the ubiquitin-proteasome pathway, which are important regulators of muscle proteolysis, is not yet known. We examined the expression in skeletal muscle of mu- and m-calpain and the ubiquitin ligases, atrogin-1 and MuRF1, in patients with primary HPT. A biopsy was obtained from the sternohyoid muscle in patients undergoing surgery for primary HPT (n=8) and in normocalcemic control patients undergoing thyroid …

HECT E3 ubiquitin ligase SMURF2 orchestrates FcεRI-dependent mast cell activation and allergic responses.

BACKGROUND: Cross-linking of IgE to its high-affinity receptor FcεRI on mast cells initiates intracellular signaling that drives the pathogenesis of allergic disorders, including asthma, allergic rhinitis, and atopic dermatitis. Precise regulation of mast cell signaling is essential for immune homeostasis and controlling allergic inflammation. OBJECTIVE: We sought to explore the functional role of HECT E3 ubiquitin ligase SMURF2 in FcεRI-dependent mast cell activation and allergic responses, as well as the expression profile in allergic disease. METHODS: We examined SMURF2 expression in mast

Structural basis of diverse substrate recruitment by the HIV-1 Vpr-hijacked CRL4(VprBP) E3 ligase.

HIV-1 hijacks the host CUL4-RING E3 ligase (CRL4) complex to trigger the degradation of various restriction factors. The viral accessory protein Vpr is the key protein that recruits diverse substrates to the receptor VprBP for proteasomal degradation. However, the underlying molecular mechanism remains poorly understood. Here, we characterize the molecular architecture of the DDB1-VprBP substrate recognition unit alone and in complex with different Vpr-substrates. Our results pinpoint that the LisH domain of VprBP mediates the DDB1-VprBP dimerization. Of note, the Armadillo-like (ARM-like) dom

Neddylation-dependent RAB40-Cullin5 E3 ligase complex controls ubiquitin-mediated proteasomal degradation of DNAJC13.

DNAJC13 is a member of DNAJ co-chaperone family and its missense mutation causes a rare familial form of Parkinson's disease. DNAJC13 is indispensable for neuronal homeostasis by regulation of autophagy and the regulation of receptor expression on the cell surface. However, the regulatory mechanisms governing DNAJC13 expression remain poorly understood. We previously demonstrated that DNAJC13 binds to RAB40 small GTPase. Unlike other RABs, RAB40 proteins interact with activated Cullin-5 (CUL5) via the SOCS box, forming an Cullin-RING E3 ubiquitin ligase (CRL) complex that regulates ubiquitylat

A fish-specific E3 ubiquitin ligase NHRE3 suppresses antiviral immunity by degrading ISG15 via ubiquitination.

Grass carp (Ctenopharyngodon Idella) hemorrhagic diseases (GCHD), caused by grass carp reovirus (GCRV), has posed a serious threat to the grass carp aquaculture industry in China. Ubiquitination plays a central role in viral evasion of the host interferon response; therefore, elucidating the mechanisms by which viruses exploit ubiquitination to achieve immune escape is of significant scientific importance and translational value. In a previous study, we identified an E3 ubiquitin ligase-like gene associated with susceptibility to hemorrhagic disease through random mutagenesis in rare minnow (G

Modulation of the E3-ubiquitin ligase RNF217 alters ferroportin levels in enterocytes and iron loading in murine hemochromatosis.

Excess iron induces tissue toxicity in various conditions, including hereditary hemochromatosis (HH). Hepcidin, a liver-derived hormone encoded by the HAMP gene, plays a pivotal role in regulating systemic iron by mediating the degradation of ferroportin (FPN), the sole cellular iron exporter. Previous research found that the E3 ubiquitin ligase RNF217 is a key regulator of iron homeostasis by directly affecting FPN degradation; however, the role of RNF217 overexpression in iron-overload disorders such as HH is poorly understood. To address this question, we generated both global and intestine

Huntington's disease LIG1 modifier variant increases ligase fidelity and suppresses somatic CAG repeat expansion.

Huntington's disease (HD) is a fatal neurodegenerative disorder caused by inheriting an expanded CAG repeat tract in the huntingtin gene (HTT) that further expands in somatic cells over an individual's lifetime. Genome-wide association studies have provided critical insight into factors that modify the course of disease. These include DNA repair genes that alter the rate of somatic expansion and other genes that do not appear to directly influence this process. One modifier gene is DNA ligase 1 (LIG1), in which a variant specifying a lysine to asparagine substitution (K845N) is associated with

GID/CTLH E3 ligase complex control cell fate programs for sexual development of Plasmodium falciparum.

Transmission of the malaria parasite Plasmodium falciparum requires the formation of specialised sexual cells called gametocytes. A hallmark of P. falciparum gametocyte development is its long duration, during which the parasite undergoes dramatic cellular remodelling including morphological, physiological and metabolic changes which result in the formation of a transmission ready, stage V gametocyte. Here we show that the PfGID E3 ubiquitin ligase complex regulates critical gametocyte cell fate programmes through the targeted ubiquitination of key proteins. Deletion of PfGID complex component

DTX3L E3 ligase: Molecular functions, regulatory mechanisms, and potential as a novel cancer biomarker and therapeutic target.

The ubiquitin-proteasome pathway is a critical regulatory system in eukaryotic cells, which mediates dynamic and reversible protein modifications through the E1-E2-E3 ubiquitination cascade. E3 ubiquitin ligases, which catalyze substrate-specific ubiquitination for proteasomal degradation, play essential roles in maintaining cellular homeostasis. The deltex E3 ubiquitin ligase 3 L (DTX3L) modulates a variety of biological processes, such as proliferation, cell cycle, migration, invasion and apoptosis. Emerging evidence implicates DTX3L dysregulation in the pathogenesis of multiple malignancies…

The herbivore-inducible coumarate-CoA ligase 5 (Zm4CL5) in maize contributes to the production of non-lignin phenylpropanoids.

Maize coumarate-CoA ligase gene expression is differentially regulated by different biotic stresses, and Zm4CL5 is induced by fall armyworm herbivory and impacts plant growth. The phenylpropanoid pathway is important for the synthesis of numerous compounds involved in plant growth and stress resilience, including lignin and flavonoids. The last common step in the core phenylpropanoid pathway is the enzyme 4-coumarate:CoA ligase (4CL). Maize (Zea mays) has five 4CL genes, four of which (Zm4CL1-4) are type I (lignin-associated 4CLs), while one (Zm4CL5) is a type II (non-lignin-associated 4CL). I

Glutamate-cysteine ligase as a diagnostic biomarker and therapeutic target in Parkinson's disease: a multi-omics and experimental study.

Parkinson's disease (PD) is a major global health challenge, and the identification of highly specific and easily detectable blood markers is critical for early diagnosis and intervention in PD. In this study, we recruited 21 patients with Parkinson's disease and 16 healthy controls. We examined blood samples for marker screening using targeted metabolomics and proteomics and machine learning techniques. We found that glutathione metabolism was enriched in Parkinson's disease patients, and the glutathione synthesis rate-limiting enzyme Glutamate-cysteine Ligase catalytic subunit (GCLC) was sig

17-beta estradiol prevents cardiac myocyte hypertrophy by regulating mitochondrial E3 ubiquitin ligase 1.

Cardiac hypertrophy is a cellular process characterized by the increased size of cardiomyocytes in response to a high workload or stress. 17-beta estradiol (E2) has cardioprotective and anti-hypertrophic effects by maintaining mitochondrial network and function. MUL1 is a mitochondrial ubiquitin ligase directly involved in the control of mitochondrial fission and mitophagy. Studies from our group and others have previously shown that cardiomyocyte hypertrophy is associated with mitochondrial fission and dysfunction. These findings led us to study in vitro whether E2 regulates MUL1 to prevent c

Gamma-Glutamyl Cysteine Ligase Activity as a Proxy for Human T Cell Function and Drug-Induced Immunosuppression.

T cell effector functions are critical for immune defense, but their dysregulation can cause diseases like immune exhaustion in cancer and loss of tolerance in autoimmunity. Curtailing these functions is essential in therapies such as chimeric antigen receptor T-cell (CAR-T) therapies or organ transplantation to avoid hyperactivation and rejection. A major challenge in the field is the precise, live measurement of T cell function at the single-cell level, limiting the prediction of immune responses, the development of effective immunotherapies, and optimization of immunosuppressive regimens. RING-Box E3 Ligase Target N-Terminal Lysine 55 to Regulate Turnover of Sp7 Protein.

Specificity protein 7 (Sp7) is essential for osteoblast differentiation and bone formation. In humans, Sp7 gene mutations are associated with skeletal abnormalities, including osteogenesis imperfecta. Ubiquitylation regulates the cellular levels of Sp7 protein. However, the role of the largest class of E3 ubiquitin ligases in the turnover of Sp7 protein remains unknown. Here, we report for the first time that both catalytic subunits of multi-subunit RING-box E3 ligase, Rbx1 and Rbx2, are expressed in the skeletal tissues and during osteoblast differentiation. In situ immunofluorescence and bio…

Cotargeting of thioredoxin 1 and glutamate-cysteine ligase in both imatinib-sensitive and imatinib-resistant CML cells.

Chronic myeloid leukemia (CML) is a type of malignancy characterized by harboring the oncogene Bcr-Abl, which encodes the constitutively activated tyrosine kinase BCR-ABL. Although tyrosine kinase inhibitors targeting BCR-ABL have revolutionized CML therapy, native and acquired drug resistance commonly remains a great challenge. Thioredoxin 1 (Trx1) and glutamate-cysteine ligase (GCL), which are two major antioxidants that maintain cellular redox homeostasis, are potential targets for cancer therapy and overcoming drug resistance. However, how their inhibition is implicated in CML is still unc

Subtractive proteomics unravel the potency of D-alanine-D-alanine ligase as the drug target for Burkholderia pseudomallei.

Melioidosis, also known as Whitmore's disease, is caused by the deadly pathogen Burkholderia pseudomallei and remains a significant global health concern, particularly in South Asia. The disease is contracted through exposure to contaminated soil, water, air, and food. Infected individuals often present with abscesses in internal organs such as the lungs, spleen, and liver, and in soft tissues, with severe cases leading to septic shock and acute pneumonia. The rising incidence and mortality rates, coupled with B. pseudomallei's ability to form biofilms and develop resistance to antibiotics lik

Inactivation of mitochondrial MUL1 E3 ubiquitin ligase deregulates mitophagy and prevents diet-induced obesity in mice.

Obesity is a growing epidemic affecting millions of people worldwide and a major risk factor for a multitude of chronic diseases and premature mortality. Accumulating evidence suggests that mitochondria have a profound role in diet-induced obesity and the associated metabolic changes, but the molecular mechanisms linking mitochondria to obesity remain poorly understood. Our studies have identified a new function for mitochondrial MUL1 E3 ubiquitin ligase, a protein known to regulate mitochondrial dynamics and mitophagy, in the control of energy metabolism and lipogenesis. Prevention of age-related truncation of γ-glutamylcysteine ligase catalytic subunit (GCLC) delays cataract formation.

A sharp drop in lenticular glutathione (GSH) plays a pivotal role in age-related cataract (ARC) formation. Despite recognizing GSH's importance in lens defense for decades, its decline with age remains puzzling. Our recent study revealed an age-related truncation affecting the essential GSH biosynthesis enzyme, the γ-glutamylcysteine ligase catalytic subunit (GCLC), at aspartate residue 499. Intriguingly, these truncated GCLC fragments compete with full-length GCLC in forming a heterocomplex with the modifier subunit (GCLM) but exhibit markedly reduced enzymatic activity. Crucially, using an a

CUL4A-DDB1-circRFWD2 E3 ligase complex mediates the ubiquitination of p27 to promote multiple myeloma proliferation.

Multiple myeloma (MM) is an incurable disease characterized by the abnormal expansion of plasma cells in the bone marrow (BM). Numerous studies have shown that BM tumor cells can influence the tumor microenvironment (TME) through communication with extracellular vesicle circular RNAs (circRNAs), a type of noncoding RNA. Our study revealed that a circular RNA, circRFWD2 (hsa_circ_0015361), is expressed by MM cells and translated into a new protein, circRFWD2_369aa. We found that elevated levels of circRFWD2_369aa in MM peripheral blood samples were closely associated with poor outcomes in MM pa
