[Comparative study of lipid peroxidation in the eye pigment epithelium of pigmented and albino animals].

The induction of lipid peroxidation in the eye pigment epithelium of pigmented rabbits by the Fe2+ + ascorbate and xanthine--xanthine oxidase systems did not result in accumulation of malonic dialdehyde. In albino rabbits the lipid peroxidation of the non-pigmented pigment epithelium and retina occurred at a high rate under the same conditions. The rate of lipid peroxidation also showed an increase after removal of melanoprotein granules from the pigmented pigment epithelium. The latter inhibited the rate of lipid peroxidation in the eye retina. In albino animals the rate of lipid peroxidation

Erythropoietic protoporphyria: lipid peroxidation and red cell membrane damage associated with photohemolysis.

The mechanism by which long wavelength ultraviolet light hemolyzes red cells obtained from patients with erythropoietic protoporphyria (EPP) was investigated. Previous studies had suggested that irradiation of these red cells with wavelengths of light capable of eliciting dermatological manifestations led to oxygen-dependent colloid osmotic hemolysis through the formation of peroxides. In the present report, lipid peroxidation during in vitro irradiation of EPP red cells with long ultraviolet light was demonstrated by: (a) the formation of 2-thiobarbituric acid reactants; (b) the presence of c

Sperm Antioxidant Capacity Discriminates Between Fertile and Infertile Men and Is Strictly Related to Lipid Peroxidation and Lipid Mediator Production.

Oxidative stress (OS) and inflammation are common causes of male infertility; antioxidants in spermatozoa and especially in seminal plasma play a protective role. The study aims to evaluate whether OS lipid mediators (F2-isoprostanes: F2-IsoPs; Resolvin D1: RvD1) measured in seminal plasma, and Trolox equivalent antioxidant capacity (TEAC), measured in both seminal plasma and spermatozoa, are associated with male infertility. Semen samples (18 fertile and 62 infertile subjects) were examined following WHO guidelines and with a mathematically elaborated transmission electron microscopy analysis

The Effects of Salicyluric Acid, the Main Metabolite of Aspirin, on Lipid Peroxidation Induced by Iron and Copper Ions in a Lipid Membrane Model.

Salicyluric acid (SUA), the main metabolite of aspirin and a natural product, is known for its ability to chelate iron and other metal ions. In particular, the chelation and increased excretion of iron by SUA may contribute to the aspirin-induced iron deficiency anemia observed in long-term aspirin users. The redox activity of iron and copper complexes of drugs and also drug metabolites, such as SUA, is an important parameter of their overall toxicity profile, including the induction of ferroptosis, which has been associated with many diseases. In this context, the effect of SUA on iron- and c

DS0384 Alleviates Necrotizing Enterocolitis: Secretes N-carbamyl glutamic Acid and Participates in Lipid Metabolism and Lipid Peroxidation Processes.

Necrotizing enterocolitis (NEC) is a life-threatening inflammatory bowel disease linked to gut microbiome dysbiosis. This study evaluates the efficacy of Limosilactobacillus reuteri strain DS0384, which secretes N-carbamyl glutamic acid (NCG), in modulating lipid peroxidation and inflammatory pathways in NEC. After pretreatment with DS0384, NEC mouse model was induced by gavage with bacteria-containing formula. NCG levels in the ileum were measured via CE-TOFMS metabolomic analysis. Additionally, rat small intestinal epithelial IEC-6 cells were exposed to lipopolysaccharide (LPS), treated with

Iron-associated lipid peroxidation in Alzheimer's disease is increased in lipid rafts with decreased ferroptosis suppressors, tested by chelation in mice.

INTRODUCTION: Iron-mediated cell death (ferroptosis) is a proposed mechanism of Alzheimer's disease (AD) pathology. While iron is essential for basic biological functions, its reactivity generates oxidants which contribute to cell damage and death. METHODS: To further resolve mechanisms of iron-mediated toxicity in AD, we analyzed post mortem human brain and ApoEFAD mice. RESULTS: AD brains had decreased antioxidant enzymes, including those mediated by glutathione (GSH). Subcellular analyses of AD brains showed greater oxidative damage and lower antioxidant enzymes in lipid rafts, the site o

Preferential incorporation of docosahexaenoic acid into nonphosphorus lipids and phosphatidylethanolamine protects rats from dietary DHA-stimulated lipid peroxidation.

In a previous study, we found that dietary docosahexaenoic acid (DHA)-stimulated tissue lipid peroxide formation was suppressed to a lesser extent than expected from the peroxidizability index of tissue total lipids. This suppression was presumed to be potentiated by mechanisms other than the lipid peroxide-scavenging system. In this study, we focused primarily on the incorporation of DHA into tissue nonphosphorus lipids and phospholipid species. DHA and different levels of dietary vitamin E (VE; 7.5, 54, 134 and 402 mg/kg of diet) were fed to rats for 32 d. In rats with poor VE status, liver

Effects of vitamin E-enriched egg yolk on lipid peroxidation, hemolysis and serum lipid concentration in young and old rats.

Effects of a diet containing different amount of vitamin E-enriched egg yolk powder on lipid peroxidation, hemolysis, and serum lipid concentration were examined in young and old rats. Young and old rats were fed experimental diets containing 5.6% or 22.5% of normal egg yolk powder, and 5.6% or 22.5% of vitamin E-enriched egg yolk powder. When young rats were fed these diets, their tocopherol concentrations in testes, heart, kidneys, and retroperitoneum fat pads increased according to the diet tocopherol contents. Lipid peroxide levels in these tissues were inversely proportional to the tocoph

Effects of PUVA therapy on skin surface lipids: skin surface lipid peroxidation in psoriasis vulgaris and its biological significance.

PUVA therapy (Psoralen + UVA irradiation) is an effective mode of photochemotherapy for psoriasis vulgaris. The biological significance of PUVA therapy for psoriasis vulgaris has mainly been considered to be based on DNA, especially the formation of DNA crosslinks between complementary DNA stands. On the other hand, we have already reported that skin surface lipids were oxidized by UVA irradiation in vitro and this reaction was enhanced in the presence of 8-methoxy-psoralen by a singlet oxygen mechanism. Keeping this in mind, we conducted an experiment to determine whether lipid peroxide can b

Triple glutathione depletion-augmented disulfide and lipid peroxide accumulation to potentiate disulfidptosis and ferroptosis.

UNLABELLED: The adaptive regulation of abnormal redox homeostasis endows tumor cells with robust survival capabilities, with glutathione (GSH) serving as one of the antioxidant cornerstones, which significantly impairs the efficacy of many regulated cell death-related therapies, including disulfidptosis and ferroptosis. METHODS: Herein, Au nanodot-loaded iron-dithiodiglycolic acid (SS) nanoparticles (FeSS@Au NPs) were engineered to potentiate disulfidptosis and ferroptosis via triple GSH depletion for oncotherapy. FeSS NPs were pre-prepared by dominant coordination-driven self-assembly betwee

High temperature induces lipid peroxidation and PPARα-dependent peroxisomal remodeling in fish cells.

High-temperature (HT) stress induces female-to-male sex reversal (masculinization) in the teleost fish medaka (Oryzias latipes). Previous studies have shown that activation of peroxisome proliferator-activated receptor α (PPARα) signaling promotes masculinization and that experimentally induced oxidative stress elicits a similar response in a PPARα-dependent manner, suggesting that PPARα is a key mediator of temperature-induced sex reversal. Because PPARα is a lipid-sensing regulator of metabolic adaptation, it may translate oxidative stress into remodeling of metabolic organelles, including p

Spatial Association Between Polyamine Metabolism and Lipid Peroxidation in Different Pathological Regions of Glioblastoma.

Background: Glioblastoma is highly heterogeneous, highly invasive, and resistant to temozolomide, with an extremely low 5-year survival rate. The spatial association between polyamine metabolism and ferroptosis remains unclear. Spatial metabolic heterogeneity is a core challenge in glioblastoma treatment. Methods: In this study, 17 patients with primary or recurrent high-grade glioma were included. Spatial metabolomics (n = 9), multiplex immunofluorescence (exploratory experiment n = 10, observational experiment n = 1), flow cytometry (n = 13), and mouse orthotopic models (n = 3) were combined

Molecular signatures of lipid peroxidation in cardiomyopathy: unraveling oxidative mechanisms in cardiac cell dysfunction.

Cardiomyopathy, like other major cardiovascular diseases, is becoming increasingly prevalent and represents a growing public health challenge. Although both genetic and acquired factors contribute to its development, obesity and diabetes remain among the most significant modifiable risks. These conditions share key pathological features, including oxidative stress, inflammation, and elevated lipid peroxidation, all of which can disrupt cellular metabolism and function. Lipid oxidation products, irrespective of their origin, are characterized by increased oxygenation and hydrophilicity compared

LncPEDS1-AS promotes UTUC resistance to lipid peroxidation by regulating PEDS1 expression via DDX23.

Upper tract urothelial carcinoma (UTUC) is a rare malignancy with a significantly poorer prognosis than bladder cancer (BC). One distinguishing feature of UTUC is its enhanced resistance to reactive oxygen species (ROS)-induced lipid peroxidation, a phenomenon closely associated with adverse clinical outcomes. However, the molecular mechanisms underlying this resistance remain largely unexplored. In this study, we identify LncPEDS1-AS, an ultra-long (>6900 nt) antisense lncRNA, as a key regulator of ROS resistance in UTUC. Mechanistically, LncPEDS1-AS interacts with the splicing factor DDX23,
