In vitro antifungal activity and mechanism of essential oil from fennel (Foeniculum vulgare L.) on dermatophyte species.

Fennel seed essential oil (FSEO) is a plant-derived natural therapeutic against dermatophytes. In this study, the antifungal effects of FSEO were investigated from varied aspects, such as MIC and minimum fungicidal concentration, mycelia growth, spore germination and biomass. The results indicated that FSEO had potent antifungal activities on Trichophyton rubrum ATCC 40051, Trichophyton tonsurans 10-0400, Microsporum gypseum 44693-1 and Trichophyton mentagrophytes 10-0060, which is better than the commonly used antifungal agents fluconazole and amphotericin B. Nardilysin in human brain diseases: both friend and foe.

Nardilysin is a metalloprotease that cleaves peptides, such as dynorphin-A, α-neoendorphin, and glucagon, at the N-terminus of arginine and lysine residues in dibasic moieties. It has various functionally important molecular interaction partners (heparin-binding epidermal growth factor-like growth factor, tumour necrosis factor-α-converting enzyme, neuregulin 1, beta-secretase 1, malate dehydrogenase, P42(IP4)/centaurin-α1, the histone H3 dimethyl Lys4, and others) and is involved in a plethora of normal brain functions. Less is known about possible implications of nardilysin for brain disease

Taenia crassiceps organic acids detected in cysticerci.

Taenia crassiceps cysticerci is used as an experimental model to cysticercosis studies; however there are subcutaneous cases of cysticercosis caused by these cysticerci. It remains unclear in the literature the energetic and fatty acid metabolism in cestodes. Its metabolic study may provide knowledge of pathways that may serve as potential anti-helminthic drugs sites of action. In this work we studied the citric acid cycle organic acids and the fatty acid oxidation in cysticerci removed from mice with 21 and 42 days of infection in two different evolutive stages: growing and final. The organic

Human epididymal secretome and proteome.

The proteins that are neosynthesized and secreted in the different regions of the human epididymis were determined by in vitro biosynthesis of epididymal tubules, and the luminal proteins were collected by microperfusion of each tubule. The preparations were analyzed by two-dimensional gel electrophoresis and the proteins were identified by mass spectrometry. Some of the major proteins identified corresponded to serum compounds such as albumin, transferrin and alpha-1-antitrypsin. The other proteins identified included lactotransferrin, clusterin, PEBP, NCP2/CTP/HE1, HE3, Crisp, actin, calmodu

Brain pyruvate oxidation in experimental thiamin-deficiency encephalopathy.

Pyrithiamine-induced thiamin deficiency has been used in rat as an experimental form of Wernicke-Korsakoff encephalopathy, a disease associated with chronic alcoholism. Although the main etiological factor is known to be the lack of thiamin, the biochemical mechanisms involved in the pathogenesis remain unclear. Thiamin-dependent enzymes were studied in brain mitochondria: alpha-ketoglutarate dehydrogenase activity exhibited 40% reduction, whereas pyruvate dehydrogenase did not change significantly. Polarographic recordings of mitochondrial respiration revealed a decreased State 3, when using

[Differences in the activity of enzymes associated with energy metabolism in the heart atria and ventricles in children].

Tissue specimens from the right atrium and ventricle of the same heart were obtained during surgery in 45 children operated on account of congenital heart disease (tetralogy of Fallot and ventricular septal defect). Significant differences were found in the enzyme activities between the atrial and ventricular musculature. The activities of enzymes associated with aerobic metabolism (citrate synthase, malate dehydrogenase, with lactate metabolism) lactate dehydrogenase (and the fatty acid oxidation) hydroxyacyl-SoA-dehydrogenase) were significantly higher in the ventricular musculature. The chemical composition of standard cotton dust.

Cotton dust samples from Cotton Incorporated were investigated by X-ray fluorescence and proximate analysis methods. These dust samples are known as "standard cotton dust" and have been used by many researchers investigating the causative agent(s) and physiological mechanisms of byssinosis. Silicon, calcium, potassium, and aluminum were present in relatively high concentrations (1-4%) in the dust fractions. The ash content of the dust fractions increased as the fraction particle size decreased. Proximate analyses demonstrate that "noncellulosic organics" are the major class of constituents (35

[Pyruvate metabolism in rats with dietary B1 avitaminosis].

The influence of alimentary deficiency of thiamine on the pyruvate metabolism in the liver and blood of rats was investigated. Note is taken of a fallen activity of pyruvate-dehydrogenase (PDH) and a rising level of pyruvate in the blood and liver. An inverse relationship between the PDH activity and the pyruvate content in the liver, observed during the whole course of the experiment is in evidence, although the substrate level exceeds significantly that of the control only starting from the 20th day. In the normalization of the pyruvate level in the liver all the studied enzymatic systems pa

Irradiation of rainbow trout at early life stages results in a proteomic legacy in adult gills. Part B; the effect of a second radiation dose, after one year, on the proteomic responses in the irradiated and non-irradiated bystander fish.

This study extends the investigation of the legacy effects of exposure to a single radiation dose at one of four early life stages, in adult rainbow trout (Part A), by examining the effects of a second identical dose after one year; i.e. egg 48 h after fertilisation (48 h egg) + 1 year, eyed egg + 1 year, yolk sac larvae (YSL) + 1 year and first feeder + 1 year. This included the induction of a bystander effect in non-irradiated trout which had swam with the irradiated fish. The second radiation dose negated any beneficial proteomic responses following early life stage irradiation only, partic

Real-Time Intracellular Bacterial Metabolism During Shigella Infection Using Hyperpolarized [2-13C]Pyruvate.

Intracellular bacterial infections are challenging to study metabolically, as traditional methods often rely on indirect indicators. Hyperpolarized magnetic resonance spectroscopy (MRS) enables noninvasive, real-time metabolic monitoring, while being able to distinguish between pathogenic bacterial metabolism and host metabolism. Using dDNP-NMR, we demonstrate real-time conversion of hyperpolarized [2-13C]pyruvate to [1-13C]acetate within Shigella-infected HeLa cells. The conversion of hyperpolarized [1,4-13C2]fumarate to [1,4-13C2]malate was used to verify host cell integrity during infection

Targeted delivery of NK007 to macrophages to treat colitis.

Macrophages are important therapeutic targets for various disorders, including infectious diseases, inflammatory diseases, metabolic diseases, and cancer. In this study, we report a novel oral delivery system for the targeted delivery of anti-inflammatory therapeutics to macrophages. Using this formulation, the model drug tylophorine malate (NK007) was tightly incorporated inside beta-glucan particle shells by the formation of colloidal particles with chitosan, tripolyphosphate, and alginate via electrostatic interactions. This formulation specifically delivered NK007 to macrophages in vivo af

Isoenzyme and ultrastructural characterization of Leishmania tropica axenic amastigotes and promastigotes.

Leishmania tropica is one of the main etiological agents of cutaneous leishmaniasis in Iran. For ultrastructural and isoenzyme study, axenic amastigotes were cultured in a brain-heart infusion medium containing 20 % fetal calf serum, pH 4.5, and incubated at 37 °C in 5 % CO(2). Different stages of L. tropica revealed the same isoenzyme profiles after comparing four enzyme systems including phosphoglucomutase, 6-phosphogluconate dehydrogenase, malate dehydrogenase, and nucleoside hydrolase II. Different isoenzyme patterns for glucose-phosphate isomerase, glucose-6-phosphate dehydrogenase, nucle

Screening of nitrosative stress resistance genes in Coxiella burnetii: Involvement of nucleotide excision repair.

Coxiella burnetii, an obligate intracellular Gram-negative bacterium, is the etiological agent of Q fever. This work takes advantage of a hypersensitive Escherichia coli genetic system to identify genes involved in resistance to nitrosative stress imposed by reactive nitrogen intermediates. Among the ten candidate genes identified, the transposase, UvrB and DNA topoisomerase IV are involved in DNA transaction; the sigma-32 factor and the putative DNA-binding protein may be involved in transcriptional regulation; IF-2 is involved in protein translation; malate dehydrogenase and carbamoyl-phosph

[Catalytic properties of 6-phosphogluconate dehydrogenase from rat liver in normal state and during toxic hepatitis].

Experimental toxic hepatitis is accompanied by a decrease of 6-phosphogluconate dehydrogenase (6PGD, 1.1.1.44) activity in rat liver. Enzyme preparations of 6PGD, obtained from livers of control group rats and animals with toxic exhibit the same molecular mass of 114.2 +/- 5.8 kDa. However G6PD isolated from liver of animals with toxic hepatitis exhibited altered Km and pH-optimum values. Reduced glutathione inhibited 6PGD-activity from the liver of control group rats and did not affect the enzyme from liver of animals with toxic hepatitis. Inhibition effect of oxidized glutathione is stronger

Biochemical evaluations in skeletal muscles of primates with MPTP Parkinson-like syndrome.

The toxic effects of the neurotoxin MPTP (1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine) in primates can be exploited for investigating the physiopathology of Parkinson's disease which may also cause functional alterations of skeletal muscles, whose biochemical modifications have been studied very little. Some enzyme activities related to energy transduction in skeletal muscles were evaluated (gastrocnemius, soleus and biceps) from MPTP-treated monkeys. Systemically administered MPTP altered the enzyme activities related to: (i) the anaerobic glycolytic pathway (decrease in hexokinase and phosp

Sequential morphologic and biochemical studies of naturally occurring wheat-sensitive enteropathy in Irish setter dogs.

This study has investigated the potential role of wheat in the pathogenesis of a naturally occurring enteropathy in Irish setter dogs. At eight months on a cereal-containing diet, jejunal biopsies from affected animals exhibited partial villus atrophy, increased intraepithelial lymphocytes, and distinct biochemical abnormalities in the brush border. Activities of alkaline phosphatase and leucyl-2-naphthylamidase were almost undetectable while disaccharidases were unaltered. Activity of 5'-nucleotidase (basolateral membrane) was low, and reduced malate dehydrogenase reflected a loss of mitochon

Selective hepatic vagotomy eliminates the hypophagic effect of different metabolites.

To test the hypothesis that vagally mediated metabolic signals from the liver affect feeding, we investigated the influence of selective hepatic vagotomy on the anorectic effect of different metabolites in rats. Subcutaneous injection of glycerol (GLY), D-3 hydroxybutyrate (DHB), L-malate (MAL), L-lactate (LAC), or pyruvate (PYR) (5.7 mmol/kg metabolic body weight = b.wt.0.75, each) reduced food intake in sham-vagotomized rats. Selective hepatic vagotomy eliminated the hypophagic effect of all metabolites. In an additional experiment, LAC retained its potency to inhibit feeding in sham-vagotom
