A rapid ICP-MS screen for heavy metals in pharmaceutical compounds.

A robust general inductively coupled plasma-mass spectrometry (ICP-MS) based method was developed as an alternative to the wet chemical heavy metals test prescribed in the United States Pharmacopoeia (USP), British Pharmacopoeia (BP), Japanese Pharmacopoeia (JP) and European Pharmacopoeia (EP). The described method provides specific detection and quantitation for each of the elements expected to give rise to a positive response in the compendial methods: arsenic (As), selenium (Se), cadmium (Cd), indium (In), tin (Sn), antimony (Sb), lead (Pb), bismuth (Bi), silver (Ag), palladium (Pd), platin…

Interactions of metals affect their distribution in tissues of Phragmites australis.

Culms of Phragmites australis were grown in vermiculite in a greenhouse. Some plants were exposed to 1000 microg/g Cu or Pb or Zn, or combinations of two or three of those metals. When plants reached senescence, they were harvested and analyzed for metal concentrations in upper leaves, lower leaves, stems and roots. While all metals accumulated in highest concentrations in the roots, Zn accumulated in aboveground tissues far more than the other metals. Furthermore, the concentration of any one metal in the different tissues was affected by the presence of other metals. The amount of copper in …

[Function and toxicity of trace metals in the central nervous system].

Ход выполнения metals such as zinc, manganese and iron usually serve the function of metalloproteins in neurons and glial cells, while a portion of trace metals exists in the presynaptic vesicles, and may be released with neurotransmitters. Zinc released into the synaptic cleft may serve as an inhibitory neuromodulator of glutamate release in the hippocampus, while neuromodulation by other trace metals such as manganese and copper might mean both functional and toxic aspects in the synapse. Dietary zinc deficiency affects zinc homeostasis in the brain, followed by an enhanced susceptibility to excitoto…

Heavy metals in sandy sediments of the Rías Baixas (NW Spain).

We studied the concentrations of certain heavy metals in the total and labile fractions of sandy sediments from four embayments situated on the northwestern coast of Spain. In order to compensate for the effects of the heterogeneity in texture of the different samples, data were normalized. Organic carbon was the most suitable normalizing factor for copper, lead and zinc in the total fraction and copper in the labile fraction, and iron was the most suitable for the other metals in both fractions. Enrichment of copper, lead and zinc in these sediments may have been caused by sewage contaminatio…

The neuromelanin of human substantia nigra and its interaction with metals.

Neuromelanin (NM) is a peculiar biochemical component of several neurons in the Substantia Nigra (SN), the target area of the degenerative process in Parkinson Disease (PD). SN NM has peculiarities as to its composition and an impressive capacity of chelating metals, iron in particular, but not exclusively. Gaining insights into the structural and functional characteristics of NM should help understanding the reasons of selective vulnerability of nigral neurons in many parkinsonian conditions. From the present data a protective role of NM can be postulated until the buffering capability toward…

Effects of metals on the nervous system of humans and animals.

Several metals have toxic actions on nerve cells and neurobehavorial functioning. These toxic actions can be expressed either as developmental effects or as an increased risk of neurodegenerative diseases in old age. The major metals causing neurobehavioral effects after developmental exposure are lead and methylmercury. Lead exposure in young children results in a permanent loss of IQ of approximately 5 to 7 IQ points, and also results in a shortened attention span and expression of anti-social behaviors. There is a critical time period (<2 years of age) for development of these effects, afte…

Chromium-resistant soil actinomycetes: their tolerance to other metals and antibiotics.

Chromium occurs widely in most soils, but generally in trace amounts. Actinomycetes, one of the important components of the microbial population in soils interact with a variety of metals including chromium. This study was aimed to evaluate the tolerance of soil actinomycetes to Cr6+, other metals and antibiotics. Thirty-two actinomycete isolates were screened for their tolerance to Cr6+ on tryptone yeast extract agar medium supplemented with Cr6+ at concentrations ranging from 100 to 2000 micrograms ml-1. Thirteen Cr-tolerant isolates were selected on the basis of their growth at the highest …

Human exposure to toxic metals: factors influencing interpretation of biomonitoring results.

An important approach to assessment of risk from environmental and occupational exposures is biomonitoring which provides an estimate of the total dose absorbed and gives indirect access to determination of target site concentrations. It is also a tool for assessing residual exposure, when respiratory protection is used. The interest in biological effects from toxic metals has increased during the last decades, as large amounts of metals have been released into industrial areas, and it is estimated that approximately 150,000 Danish workers are exposed to various metals. Since biomonitoring res…

Production of metallothionein and heat-shock proteins in response to metals.

Acute stress, such as heat, and some metals, such as arsenite, will induce a specific group of stress proteins referred to as heat-shock proteins. The heat-shock proteins contribute to the survival of cells following a variety of stresses. Similarly, metals such as cadmium and zinc, will increase the levels of metallothionein (MT). The metal-binding protein, MT, has also been found to have a protective role in the cellular response to acute stresses like heavy metals. The purpose of the present study was to examine the production of these proteins in response to metals. Rat hepatocytes were ma…

Darkfield illumination improves microscopic detection of metals in Timm's stained tissue.

Deposits of trace or toxic metals can be quickly identified by light microscopical surveys of tissue sections stained for metals by variants of Timm's silver enhancement method. The present work shows that the small, isolated silver grains that label isolated deposits of metal in tissue are undetectable in brightfield light microscopy but are easily detected in darkfield microscopy. Darkfield illumination is therefore recommended for improving the detection of trace or toxic metals in tissue.

[Biological aspects of occupational exposure to cadmium and several other metals].

We have performed several cross-sectional epidemiological surveys among workers exposed to cadmium, mercury vapour or manganese in order to assess : their early biological or functional effects; the biological tests allowing an assessment of the amount of metal absorbed or stored in the body; the acceptable exposure levels. Studies have also been carried out among persons exposed to inorganic arsenic in order to define its inactivation mechanism and to develop a biological test of exposure. The kidney is the main critical organ following long-term exposure to cadmium. To prevent the occurrence

Relationship between exposure to heavy metals and prevalence of renal dysfunction.

Total proteinuria (biuret method) and the urinary excretion of specific proteins (albumin, transferrin, orosomucoid and IgG measured by an automated immunoprecipitin reaction and beta 2-microglobulin determined by radioimmunoassay) have been assessed in four groups of workers: a control group (n = 88) and a group exposed to cadmium (n = 148), mercury vapour (n = 63) and lead (n = 25) respectively. The results demonstrate that a moderate exposure to lead (lead concentration in blood < 62 micrograms/100 ml) does not change the prevalence of renal dysfunction, whereas mercury vapour exposure incr

Seizure disorders and trace metals: manganese tissue levels in treated epileptics.

We determined the concentrations of manganese in whole blood and hair in 52 epileptics, 6 blood relatives, and 24 normal controls. Blood, and possibly hair manganese content, was significantly lower in treated epileptics than in controls (p less than 0.002). Although not all patients showed reduced tissue manganese levels, most of those with frequent seizures had manganese levels falling below the lowest control level, suggesting a relationship between manganese tissue levels and high seizure activity. These differences in manganese levels were not correlated with the type, dose, or plasma lev
