PURPOSE: The current study was done to elucidate the in vivo mechanism of action of intravesical instillation of oxybutynin to treat overactive bladder.
MATERIALS AND METHODS: In rats receiving oral and intravesical oxybutynin we measured muscarinic receptors in the bladder and other tissues by radioligand binding assay using [3H]NMS ([N-methyl-3H] scopolamine methyl chloride) with the simultaneous measurement of plasma concentrations of oxybutynin and its active metabolite N-desethyl-oxybutynin. The kidney is well documented as the target organ for mercuric ion. Mechanisms by which this ion accumulates in renal tissue, however, are less well understood. Sulfhydryl groups in renal tissue might well bind this metal and serve as a sink for its accumulation. Camel whey protein (CWP) is rich in potent antioxidant and antimicrobial compounds, while the iron-based metal-organic framework (MOF)-235 offers high biocompatibility for drug delivery. In this study, sewage sludge-derived activated carbon (SDAC) was synthesized, characterized, and tested for its potential as an adsorbent of methyl tert-butyl ether (MTBE). The SDAC was produced by activating the sludge with zinc chloride and subsequently subjected to various ranges of pyrolytic temperatures. It was then characterized using SEM/EDX, BET, and TGA. A rescue window for glutamate-insulted cells with regard to Ca(2+) influx was first investigated. The addition of EGTA, an impermeable calcium chelator to the culture within 5 min after the beginning of glutamate stimulation potently suppressed the neuronal cell death examined at 22 h.
