A responder-informed gut microbial consortium enhances anti-PD-1 efficacy in a mouse cancer model.

Aim: Immune checkpoint inhibitors (ICIs), particularly anti-programmed cell death protein 1 (PD-1) therapy, have improved cancer treatment outcomes, yet durable benefit is achieved in only a subset of patients. Growing evidence implicates the gut microbiome as a modulator of ICI responsiveness, but defined and experimentally validated microbial strategies remain limited. This study aimed to identify responder-associated gut microbes and to evaluate a defined bacterial consortium for enhancing PD-1 blockade efficacy. Methods: Publicly available shotgun metagenomic datasets from anti-PD-1-treate

Sustained anaerobic valorization of refractory wastewater into bioenergy via abiotic-biotic hybrid systems: Unraveling synergistic hydrolysis and optimized interspecies electron transfer.

Anaerobic digestion (AD) of refractory industrial wastewater remains challenging due to structural recalcitrance and thermodynamic limitations. While abiotic-biotic hybrid systems incorporating electroactive materials (EMs) offer promise, their long-term efficacy and mechanisms at a system level, particularly with real industrial waste streams, are underexplored. This study demonstrates the sustained, system-level enhancement of anaerobic valorization using magnetite (Mag)- or pyrogenic carbon (PC)-amended reactors during 140-day continuous operation treating real membrane-making wastewater la

Synergistic removal of morpholine fungicides and cadmium from agricultural water by a biochar-immobilized bacterial-duckweed system: Quantifying roles of biodegradation, adsorption, and phyto-uptake.

The co-contamination of agricultural water by morpholine fungicides (e.g., flumorph and dimethomorph) and cadmium (Cd) poses significant ecological threats, challenging conventional treatment approaches. This study developed an innovative bioremediation system integrating biochar-immobilized microbial consortia with phytoremediation, and quantified the individual contributions of biodegradation, biosorption, phyto-uptake, and biochar-adsorption to the synergistic removal of pesticide and Cd co-contaminants. A novel Cd-tolerant and flumorph-degrading bacterium, Alcaligenes faecalis X4, was comb…

Impact of oral typhoid vaccination on the human gut microbiota and correlations with s. Typhi-specific immunological responses.

The resident microbial consortia of the human gastrointestinal tract play an integral role in modulating immune responses both locally and systemically. However, detailed information regarding the effector immune responses after vaccine administration in relation to the gastrointestinal microbiota is absent. In this study, the licensed oral live-attenuated typhoid vaccine Ty21a was administered in a clinical study to investigate whether oral immunization resulted in alterations of the microbiota and to identify whether a given microbiota composition, or subsets of the community, are associated…

Importance of exposure dynamics of metal-based nano-ZnO, -Cu and -Pb governing the metabolic potential of soil bacterial communities.

Metal-based engineered nanomaterials (ENMs) are known to affect bacterial processes and metabolic activities. While testing their negative effects on biological components, studies traditionally rely on initial exposure concentrations and thereby do not take into consideration the dynamic behavior of ENMs that ultimately determines exposure and toxicity (e.g. ion release). Moreover, functional responses of soil microbial communities to ENMs exposure can be caused by both the particulate forms and the ionic forms, yet their relative contributions remain poorly understood. Therefore, we investig

Optimization of volatile fatty acids and hydrogen production from Saccharina japonica: acidogenesis and molecular analysis of the resulting microbial communities.

Response surface methodology (RSM) was used to optimize the production of volatile fatty acids (VFAs) and hydrogen from mixed anaerobic cultures of Saccharina japonica with respect to two independent variables: methanogenic inhibitor concentration and temperature. The effects of four methanogenic inhibitors on acidogenic processes were tested, and qualitative microbial analyses were carried out. Escherichia, Acinetobacter, and Clostridium were the most predominant genera in samples treated with chloroform (CHCl3), iodoform (CHI3), 2-bromoethanesulfonate (BES), or β-cyclodextrin (β-CD), respect

Degradation of atrazine by microbial consortium in an anaerobic submerged biological filter.

Atrazine (2-chloro-4-ethylamino-6-isopropylamino-1,3,5-triazine) (ATZ) is one of the components of S-triazine. Due to its certain characteristics, ATZ causes pollution in various ecosystems and has been of concern for its probable carcinogenic effects on humans. Researchers have used chemical and physical methods for removing ATZ from the environment. Although these methods are quick, they have not been capable of complete mineralization. Therefore, researchers are looking for methods with lower energy consumption and cost and higher efficiency. In this study, biodegradation of ATZ by microbia

Integrated bioremediation of microplastics-cadmium co-contamination via chitosan-sugarcane residue encapsulated fungal consortia.

The co-occurrence of microplastics (MPs) and heavy metals in aquatic environments poses a complex challenge for MPs bioremediation, particularly when cadmium (Cd) adsorbs onto MPs surfaces and enhances recalcitrance and microbial stress. This study investigates the biodegradation of MPs based on polyethylene (PE) and polypropylene (PP), under Cd stress using novel fungal consortia (QMFC1 for PE and QMFC2 for PP). To enhance their stability and performance under co-contaminated conditions, both consortia were immobilized in a chitosan-sugarcane residue matrix, resulting in the encapsulated syst

Integration of top-down rhizospheric selection and bottom-up consortium engineering enhances mechanistic biodegradation of crude oil hydrocarbons and soil recovery.

Effective remediation of crude oil-contaminated soils enriched in recalcitrant polycyclic aromatic hydrocarbons (PAHs) requires strategies that couple microbial degradative capacity with ecological compatibility. This study evaluated an ex situ rhizoremediation system integrating Crotalaria pallida with a trait-selected hydrocarbon-degrading bacterial consortium and a biosurfactant amendment, using a combination of bottom-up functional selection and top-down rhizosphere filtering. After 90 days, consortium-amended treatments achieved up to 98.3% PAH and 94.7% total petroleum hydrocarbon (TPH)

MAPK signaling pathway and rhizosphere modulation: mechanisms of arbuscular mycorrhizal symbiosis in enhancing Phragmites communis tolerance to triclocarban stress.

With escalating global health challenges, triclocarban (TCC) contamination has reemerged as a critical ecological threat, especially in wetland ecosystems. Yet, the mechanisms by which arbuscular mycorrhizal fungi (AMF) regulate plant root resistance and the rhizosphere environment under TCC exposure remain poorly understood. Here, we established an AMF-Phragmites communis symbiotic system and conducted TCC exposure experiments. Our results demonstrated that TCC exposure significantly inhibited mycorrhizal colonization, root morphology, and the antioxidant system of P. communis. Specifically,

Metagenomic and Metatranscriptomic Analyses Reveal the Structure and Dynamics of a Dechlorinating Community Containing Dehalococcoides mccartyi and Corrinoid-Providing Microorganisms under Cobalamin-Limited Conditions.

The aim of this study is to obtain a systems-level understanding of the interactions between Dehalococcoides and corrinoid-supplying microorganisms by analyzing community structures and functional compositions, activities, and dynamics in trichloroethene (TCE)-dechlorinating enrichments. Metagenomes and metatranscriptomes of the dechlorinating enrichments with and without exogenous cobalamin were compared. Seven putative draft genomes were binned from the metagenomes. At an early stage (2 days), more transcripts of genes in the Veillonellaceae bin-genome were detected in the metatranscriptome

Identification of specific corrinoids reveals corrinoid modification in dechlorinating microbial communities.

Cobalamin and other corrinoids are essential cofactors for many organisms. The majority of microbes with corrinoid-dependent enzymes do not produce corrinoids de novo, and instead must acquire corrinoids produced by other organisms in their environment. However, the profile of corrinoids produced in corrinoid-dependent microbial communities, as well as the exchange and modification of corrinoids among community members have not been well studied. In this study, we applied a newly developed liquid chromatography tandem mass spectrometry-based corrinoid detection method to examine relationships

Differential proteomic analysis of a polymicrobial biofilm.

Porphyromonas gingivalis, Treponema denticola, and Tannerella forsythia exist in a polymicrobial biofilm associated with chronic periodontitis. The aim of this study was to culture these three species as a polymicrobial biofilm and to determine proteins important for bacterial interactions. In a flow cell all three species attached and grew as a biofilm; however, after 90 h of culture P. gingivalis and T. denticola were closely associated and dominated the polymicrobial biofilm. For comparison, planktonic cultures of P. gingivalis and T. denticola were grown separately in continuous culture. W

Anaerobic oxidation of methane supports a minimal microbial community in a subsurface biofilm at Ginsburg mud volcano.

Deep marine sediments generate large amounts of methane, but most of this gas is consumed by the anaerobic oxidation of methane (AOM) mediated by microscopic consortia of anaerobic methane-oxidizing archaea (ANME) and sulfate-reducing bacteria (SRB). In this study, we investigated the AOM within a sulfate-methane transition zone (SMTZ) at a depth of ~9.6 m at the rim of the Ginsburg mud volcano in the Gulf of Cádiz. The SMTZ is supplied with sulfate from both overlying seawater and an underlying evaporitic deposit, and it coincides with a fracture zone that hosts a visible biofilm. Here, carbo
