Spatial Multiomics Analyses Reveal That Diabetes Promotes Pancreatic Cancer Progression by Stimulating Cholesterol-Induced Neutrophil Extracellular Trap Formation.

UNLABELLED: Patients with pancreatic ductal adenocarcinoma (PDAC) with diabetes mellitus (DM) exhibit poor clinical outcomes. Metabolic reprogramming of both cancer cells and immune compartments plays a crucial role in shaping the antitumor immune response in PDAC. DM-induced metabolic alteration may disrupt the intricate cross-talk between immune cells and tumor-associated immune factors, profoundly influencing PDAC progression. In this study, we performed an integrated, spatially resolved multiomics study to investigate DM-associated, cell-specific metabolic remodeling within the PDAC tumor …

Spatiotemporal multiomics reveal a default CD4 fate and a stem-like CD8 T cell subset in the thymus.

BACKGROUND: T cell development in the thymus proceeds through coordinated transcriptional and spatial transitions. While key stages have been described, the precise timing and regulatory logic of lineage bifurcation-such as the separation between αβ and γδ T cells, and the commitment to CD4 or CD8 fate remains incompletely understood. A high-resolution, integrated view is needed to clarify how distinct T cell identities are established. OBJECTIVES: The goal is to construct a comprehensive, multimodal map of thymocyte development that defines early lineage decisions, intermediate states, and s

The multiomics landscape of small peptides encoded by long noncoding RNA-derived small open reading frames in rice.

Long noncoding RNAs (lncRNAs), as a major class of noncoding transcripts, constitute a large proportion of the genomes in complex organisms. Accumulating evidence has revealed that a subset of lncRNAs harbors peptide-encoding potential. Nevertheless, the existence and biological functions of lncRNA-derived small open reading frames-encoded polypeptides (LSEPs) in plants remain largely elusive. In this study, we established an integrated multiomics pipeline combining transcriptomics, translatomics (Ribo-seq), and proteomics to systematically identify LSEPs in rice. Among the 2,764 lncRNAs detec…

Single-cell multiomics reveals exosome-mediated reprogramming and clonotypic remodeling of T cells in triple-negative breast cancer.

Triple-negative breast cancer (TNBC) is an aggressive and immunogenic subtype lacking targeted therapies. While tumor-derived exosomes are known to modulate immune function, their direct impact on human T cell plasticity and antigen specificity remains poorly defined. Here, we conducted a comprehensive single-cell multiomic analysis of primary human T cells exposed to exosomes derived from 17 genomically diverse TNBC cell lines and 35 patient samples. Integrating single-cell RNA-seq, V(D)J sequencing, non-coding RNA profiling, bulk and single-cell cytokine analyses, we uncovered conserved and
