Combined Effects of Diazoxide and Moderate-Intensity Exercise on the Restoration of Redox Balance Post-Fatigue in Fast- and Slow-Twitch Skeletal Muscles of Hypertensive Rats.

Muscle fatigue, defined as a decline in force generation, is closely linked to redox imbalance-a condition exacerbated by oxidative stress in hypertension. This study investigated the effects of diazoxide administration and moderate-intensity exercise on skeletal muscle redox status following a fatigue protocol in rats with hypertension. Animals were assigned to eight groups: control (CTRL), diazoxide (DZX), exercise (EX), exercise + diazoxide (EX+DZX), hypertension (HTN), hypertension + diazoxide (HTN+DZX), hypertension + exercise (HTN+EX), and hypertension + exercise + diazoxide (HTN+ EX+DZX…

Treatment with EUK-134 improves sarcoplasmic reticulum Ca2+ release but not myofibrillar Ca2+ sensitivity after fatiguing contraction of rat fast-twitch muscle.

Skeletal muscles undergoing vigorous activity can enter a state of prolonged low-frequency force depression (PLFFD). This study was conducted to examine whether antioxidant treatment is capable of accelerating the recovery from PLFFD, with a focus on the function of the sarcoplasmic reticulum (SR) and myofibril. One hour before fatiguing stimulation (FS) was administered, rats received an intraperitoneal injection of Eukarion (EUK-134), which mimics the activities of superoxide dismutase and catalase. Intact muscles of the hindlimbs were electrically stimulated via the sciatic nerve until the

Superiority of Tetrapeptide Tyr-Gly-Leu-Phe over Amino Acid Mixture: Mechanisms Underlying Enhanced Systemic Amino Acid Delivery, Anti-Fatigue Efficacy, and Muscle Damage Repair.

This study compared the anti-fatigue tetrapeptide YGLF with an equimolar amino acid (AA) mixture across three stages: postingestion absorption, postexercise fatigue, and muscle damage repair. Despite rapid in vitro degradation (<1% intact), YGLF elevated plasma AAs more effectively than the AA mixture 15 min postingestion (51.97 vs 46.34 mg/100 mL). Postexercise, YGLF outperformed the AA mixture by further increasing muscle AAs and blood glucose, reducing metabolic byproducts, and activating AMPK. Following muscle damage, transcriptomics revealed that the AA mixture primarily modulated energy

The increased perceived exertion during the six minute walking test is not accompanied by changes in cost of walking, gait characteristics or muscle fatigue in persons with multiple sclerosis.

BACKGROUND: Persons with Multiple Sclerosis (pwMS) frequently experience walking difficulties, often expressed as a slower walking speed during the 6 Minute Walking Test (6MWT). In addition, slower walking speeds are also related to higher levels of perceived exertion. PwMS are also known to have a higher energetic Cost of walking (Cw) and may experience muscle fatigue during prolonged walking. In this study, we aimed to explore changes in Rate of Perceived Exertion (RPE) and the Cw within participants during the 6MWT in pwMS. Additionally, concomitant changes in the mean and variability of ga…

Cardiac and muscle fatigue due to relative functional overload induced by excessive stimulation, hypersensitive excitation-contraction coupling, or diminished performance capacity correlates with sarcoplasmic reticulum failure.

The development of muscle fatigue due to exhaustive exercise is associated with impaired sarcoplasmic reticulum (SR) Ca-transport activity. This study tested the hypothesis that SR failure is a consistent feature of cardiac and skeletal muscle fatigue owing to relative functional overload regardless of the method of induction: excessive stimulation, diminished performance capacity, or excessive excitation-contraction coupling. The Ca-transport activity was determined using three unique models of muscle fatigue: chronic and rapid ventricular pacing in dogs; metabolic inhibition caused by global…

Biomechanical mechanisms of multidirectional dynamic compensatory muscle fatigue induced by abnormal cervical curvature: a cross-sectional case-control study based on surface electromyography and Cobb angle.

INTRODUCTION: Cervical lordosis, a core indicator of spinal sagittal balance, is closely associated with cervical dysfunction when abnormal (straightening or kyphosis). The muscular system, including the upper trapezius (UT) and sternocleidomastoid (SCM), contributes to the mechanical stability of the cervical spine. However, how abnormal curvature affects dynamic muscle fatigue mechanisms remains unclear. This study investigated how abnormal curvature influences the performance of the UT and SCM during movement and their correlations. METHODS: Employing a cross-sectional case-control design,…

Spatiotemporal characteristics of lower back muscle fatigue during a ten minutes endurance test at 50% upper body weight in healthy inactive, endurance, and strength trained subjects.

In modern developed societies, heavy physical demands are decreasing and getting replaced by longer periods of static, low-exertion activities such as sitting or standing. To counteract this lack of physical activity, more and more people are engaging in physical activity through exercise and training. Virtually opposite training modalities are endurance and strength. We asked if back muscle endurance capacity is influenced by training mode. 38 healthy male subjects (age range 19-31 years, mean age 22.6 years) were investigated: sedentary (Control, n = 12), endurance trained (ET, n = 13), and

Wavelet-based time-frequency intermuscular beta-band coherence decreases with age but increases after mental fatigue in ankle muscles during gait independent of age.

Mental fatigue can affect cognitive function and interfere with motor performance. We examined if mental fatigue affected gait through age-specific modulation of wavelet-based time-frequency intermuscular beta-band coherence in muscles while walking on a treadmill at 1.2 m·s-1. The Psychomotor Vigilance Task, and the AX-Continuous Performance and the Stroop tests were used to induce mental fatigue in groups of healthy young and older participants. Mental fatigue reduced stance time, stride length, and marginally step width and increased cadence, stride length and stance time variability. In ol
