Thrombus leukocytes exhibit more endothelial cell-specific angiogenic markers than peripheral blood leukocytes do in acute coronary syndrome patients, suggesting a possibility of trans-differentiation: a comprehensive database mining study.

BACKGROUND: Current angiogenic therapies for cancers and cardiovascular diseases have not yet achieved expected benefits, which reflects the need for improved understanding of angiogenesis. In this study, we focused on solving the problem of whether tissues have different angiogenic potentials (APs) in physiological conditions and how angiogenesis is regulated in various disease conditions. METHODS: In healthy and diseased human and mouse tissues, we profiled the expression of 163 angiogenic genes, including transcription regulators (TRs), growth factors and receptors (GF/Rs), cytokines and c

Endoplasmic reticulum stress and angiogenesis: is there an interaction between them?

When cells are subjected to stress by changes in their extracellular environment, unfolded proteins accumulate in the endoplasmic reticulum (ER), causing ER stress. This initiates the unfolded protein response (UPR), a signal transduction cascade aiming at restoring cellular homeostasis. The UPR and angiogenesis are involved in the pathogenesis of many diseases such as cancer, pulmonary diseases and chronic liver diseases (CLDs) including alcoholic liver disease, non-alcoholic steatohepatitis and hepatitis B. This review summarizes the upcoming knowledge of the interaction between the UPR and

Magic roundabout is a new member of the roundabout receptor family that is endothelial specific and expressed at sites of active angiogenesis.

We have used bioinformatic data mining to identify a novel, endothelial-specific gene encoding a protein with homology to the axon guidance protein roundabout (ROBO1). The new gene has been called magic roundabout (ROBO4; GenBank acc. no. AF361473) and is smaller than other members of the roundabout gene family. Thus, in the extracellular region, magic roundabout has only two of the five immunoglobulin and two of the three fibronectin domains present in other roundabout genes. Expression of magic roundabout in vitro was detected in only endothelial cells and was greater in cells exposed to hyp

Fibro-Adipogenic Progenitors From Skeletal Muscle Enhance Microvascular Angiogenesis in a 3D Model.

Skeletal muscle tissue is highly vascularized, and angiogenesis plays a key role in both muscle homeostasis and regeneration. Despite their importance in regulating various processes within skeletal muscle, little is known about the role of muscle-resident fibro-adipogenic progenitors (FAPs) in promoting microvascular angiogenesis. In order to better understand the effects of FAPs on angiogenesis, FAPs were co-cultured with adipose-derived microvascular fragments (MVFs) in three-dimensional fibrin hydrogels. We further tested if pre-culturing FAPs in brown adipogenic induction media (BAM) infl

Investigation of mechanical forces during multicellular early angiogenic sprouting by three-dimensional traction force microscopy in hydrogel matrices.

Elucidating the mechanical regulation of angiogenesis remains a challenge owing to the complexities of measuring cellular forces in this dynamic, multicellular and three-dimensional (3D) process. Current methods for force measurements typically involve traction force microscopy (TFM) applied to single cells or monolayers on 2D substrates or to individual cells within 3D extracellular matrix (ECM)-like gels. Here we present a protocol for mimicking and imaging dynamic early angiogenic sprouting into biomimetic matrices compatible with 3D TFM and for visualizing matrix degradation. Given that re

Biomimetic Mechanical Stimulation Enhances Vascular Network Formation in Engineered Cardiac Scaffold.

Myocardial tissue engineering employs cell-loaded biomaterial scaffolds to repair heart damage, but poor vascularization limits effectiveness. Meanwhile, post-infarction cardiac tissue suffers severe vascular deficiency, creating a hypoxic microenvironment that critically impairs the viability of engrafted cells. Current engineered tissues have 7-fold fewer microvessels than natural heart tissue, highlighting the need for better vascularization methods. Mechanical forces can regulate cellular behaviors like proliferation and morphogenesis, especially promote vascular network formation by enhan

The Role and Mechanism of Endothelial Progenitor Cell-Derived Extracellular Vesicles in Vascular Repair of Ischemic Heart Disease.

Ischemic heart disease (IHD) remains a leading global cause of death, sustained by microvascular dysfunction and defective endogenous repair despite revascularization. Endothelial progenitor cells (EPCs) were initially proposed as regenerative mediators, but direct cell therapy is constrained by poor engraftment, phenotypic heterogeneity, and safety concerns. Accumulating evidence establishes EPC-derived extracellular vesicles (EPC-EVs) as the principal effectors of vascular repair. These vesicles deliver bioactive cargo-including miR-126, miR-210, lncRNAs, and proteins that activate VEGF/PI3K

An Overview and Review of Growth Factors in Wound Healing: Emerging Trends and Innovations.

Wound healing is a complex process involving various cellular and molecular events aimed at restoring tissue integrity. Growth factors play a pivotal role in orchestrating these events by regulating cell migration, proliferation, differentiation, and extracellular matrix synthesis. Several growth factors have been identified as crucial mediators of wound healing, including PDGF, TGF-β, VEGF, FGF, and EGF. PDGF is released by platelets upon injury and stimulates the migration and proliferation of fibroblasts and smooth muscle cells, promoting the formation of granulation tissue. TGF-β regulates

Morphologically adaptive microconical hydrogels of gelatin/polyvinyl alcohol with curcumin for diabetic wound repair.

Conventional planar hydrogels cannot adequately conform to the irregular topography of complex wounds, which limits the efficiency of drug delivery. In this study, a morphologically adaptive microconical hydrogel (G/P@Cur MC) was developed, composed of physically cross-linked gelatin/polyvinyl alcohol and curcumin. Fabricated using a wound-replicating microporous mold, this dressing achieves conformal contact with the tissue and eliminates spatial blind zones in drug delivery. The G/P@Cur hydrogel integrates excellent mechanical properties, biocompatibility, and sustained release characteristi

4D force patterning enables spatial control of angiogenesis.

Engineering organized microvascular networks remains a critical challenge in tissue engineering and regenerative medicine. While biochemical approaches for patterning angiogenesis via growth factor delivery have shown promise, their inability to pattern sustained growth factors with spatiotemporal control limits effectiveness. Here, we demonstrate that dynamically patterned mechanical forces enable precise spatiotemporal control over angiogenic sprouting. We developed a magnetically actuated human vessel-on-a-chip platform that integrates a perfusable endothelialized microchannel within a coll

Hypoxia-activated probiotic vesicle coating enables laminar-shear-protective angiogenesis and antibacterial effects in infectious bone defects.

In treating infectious bone defects, bacterial eradication alone is insufficient, as impaired blood perfusion characterized by low fluid shear stress (FSS) hampers angiogenesis, thereby compromising osteogenesis and delaying bone repair. Herein, we engineered a low-FSS-activated, pro-angiogenic implant coating with antibacterial properties. This coating comprises a Fe3+-tannic acid (Fe3+-TA) chelation network as the adhesive sublayer, anchoring black phosphorus (BP) nanosheets preloaded with metformin (Met), and is further capped by an outer layer of Lactobacillus animalis-derived extracellula

Hierarchical regulation and mechanism of "open-hollow-fibrous network" structures: Osteogenic-angiogenic coupling responses of poly(γ-benzyl-L-glutamate) microspheres.

Biomimetic fibrous microspheres with a high specific surface area hold substantial promise for bone tissue engineering. In this study, asymmetric open-hollow nanofibrous microspheres (HNMs) were fabricated from polypeptide poly(γ-benzyl-L-glutamate) (PBLG) via a combination of emulsion and thermally induced phase separation, yielding PBLG HNMs. To further impart biofunctionality, the copper peptide (GHK-Cu) was covalently grafted onto the microspheres to obtain osteoinductive and pro-angiogenic PBLG-GCu HNMs. The optimized microspheres exhibited an average diameter of 372 ± 102 μm, which is su

Cell-free therapeutics for non-healing wounds: role of MSC-derived exosomes in macrophage polarization, angiogenesis, and fibroblast-mediated ECM remodeling-bridging preclinical insights to clinical translation.

Non-healing wounds, such as diabetic foot ulcers, burns, and pressure injuries, represent a persistent clinical challenge, affecting an estimated 2-5% of the global population and accounting for annual healthcare costs exceeding USD 25 billion. Their failure to heal is driven by unresolved inflammation, inadequate vascularization, and progressive disorganization of extracellular matrix (ECM) architecture, which collectively disrupt the coordinated cascade of tissue regeneration. Mesenchymal stem cell (MSC)-derived exosomes have emerged as a potent cell-free therapeutic approach capable of addr

Prevascularized grafts with spatially organized MSC spheroids to accelerate therapeutic angiogenesis in ischemic disease.

OBJECTIVE: Ischemic diseases, characterized by impaired blood flow and progressive tissue necrosis, remain a major challenge in regenerative medicine. Surgical revascularization remains the gold standard for restoring blood flow in major vessels, but still shows limited effects on microvascular regeneration. To address this unmet need, various strategies in therapeutic angiogenesis have been explored to induce microvascular formation, including delivery of bioactive molecules, stem cells, or pre-vascularized grafts. However, injection-based approaches often suffer from off-target effects, and

LRG1 Alters Pericyte Phenotype and Compromises Vascular Maturation.

Upregulation of leucine-rich alpha-2-glycoprotein-1 (LRG1) contributes to aberrant neovascularization in many different diseases. In contrast, LRG1 is not involved in developmental angiogenesis. Here, we investigated the vasculopathic properties of LRG1 by examining its effect on developing retinal blood vessels. By injecting recombinant protein or an expression vector into the mouse retina during vascular development, we showed that exogenous LRG1 reduces pericyte coverage and NG2 expression. It leads to diminished collagen IV sheathing, fewer adhesion and gap junctions, and reduced vessel ca

Gene dysregulation impairs placental angiogenesis in allogeneic pig pregnancies.

Embryo transfer (ET) is a valuable reproductive technology in pigs, albeit its efficiency remains significantly lower than that of natural mating or artificial insemination (AI), owing to high embryonic death rates. Critical for embryo survival and pregnancy success is the placenta, which supports conceptus development through nutrient exchange, hormone production, and immune modulation. Alterations in placental development and function may therefore underlie the reduced efficiency of ET. To investigate the molecular mechanisms underlying this dysfunction, crossbred (Landrace × Large White) re

Glucocorticoid-induced osteoporosis in children: emerging vascular and molecular targets.

PURPOSE OF REVIEW: Glucocorticoid-induced osteoporosis (GIO) poses a significant risk to pediatric patients undergoing chronic glucocorticoid therapy for inflammatory, autoimmune, and genetic conditions. This review highlights the pathophysiology of GIO in children and emerging insights into the role of bone vasculature in skeletal health during growth. RECENT FINDINGS: GIO arises from a dual mechanism of increased bone resorption and decreased bone formation, leading to rapid declines in bone mineral density. Recent studies emphasize the importance of angiogenic-osteogenic coupling, particul

Near-infrared-activatable angiogenic proteinaceous nanomicelles for sequential augmentation of bone repair in periodontal defects.

The highly vascularized architecture of natural bone has inspired sequential augmentation of vascularization and osteogenesis to promote bone tissue regeneration, particularly in periodontal defects characterized by severely diminished bone quantity and quality, often worsened by systemic conditions such as osteoporosis. However, current growth factor delivery systems frequently lack precise control over release profiles and allow limited modification of predetermined kinetics. Herein, near-infrared (NIR) light-activatable angiogenic proteinaceous nanomicelles (NMs) bioengineered from vascular

Mechano-immunomodulation of macrophages influences the regenerative environment of fracture healing through the regulation of angiogenesis and osteogenesis.

Successful completion of the initial inflammatory phase is critical for the establishment of a regenerative environment conducive to long-term fracture healing. Mechanical signals are among the most potent regulators of bone repair, yet whether local mechanics can modulate inflammation and associated immune response remains poorly understood. In this study, we develop a 3D in vitro model comprising of a purpose-built bioreactor that can replicate distinct loading conditions experienced during ambulation of fixated or unfixed large bone defects, and a haematoma mimetic fibrin hydrogel mirroring
