OBJECTIVE: Genetic dystonia is a complex movement disorder with diverse clinical manifestations resulting from pathogenic mutations in associated genes. A recent paradigm shift emphasizes the functional convergence among dystonia genes, hinting at a shared pathomechanism. However, the neural dynamics supporting this convergence remain largely unexplored.
METHODS: Herein, we analyzed microelectrode recordings acquired during pallidal deep brain stimulation surgery from 31 dystonia patients with pathogenic mutations in the AOPEP, GNAL, KMT2B, PANK2, PLA2G6, SGCE, THAP1, TOR1A, and VPS16 genes.

Low-intensity ultrasound stimulation of the retina has the ability to modulate neural activity in the primary visual cortex (V1), however, it is currently unclear how different intensities and durations of ultrasonic stimulation of the retina modulate neural activity in V1. In this paper, we recorded local field potential (LFP) signals in the V1 brain region of mice under different ultrasound intensities and different stimulation times. The amplitude of LFP corresponding to 1 s before ultrasound stimulation to 2 s after stimulation (-1-2 s) was analyzed, including the power and sample entropy

PURPOSE: To evaluate the feasibility of automated segmentation of pigmented choroidal lesions (PCLs) in optical coherence tomography (OCT) data and compare the performance of different deep neural networks.
METHODS: Swept-source OCT image volumes were annotated pixel-wise for PCLs and background. Three deep neural network architectures were applied to the data: the multi-dimensional gated recurrent units (MD-GRU), the V-Net, and the nnU-Net. The nnU-Net was used to compare the performance of two-dimensional (2D) versus three-dimensional (3D) predictions.
RESULTS: A total of 121 OCT volumes w

BACKGROUND: Accumulating evidence indicated that anhedonia as a transdiagnostic construct might be an inherent feature of obsessive-compulsive disorder(OCD). Moreover, our recent study demonstrated that OCD patients exhibited consummatory anhedonia but not anticipatory anhedonia. However, neural mechanisms of consummatory anhedonia in OCD has not been explored. This study aimed to investigate this issue using resting-state functional magnetic resonance imaging (fMRI).
METHODS: 44 OCD patients with high consummatory anhedonia(OCD-HCA), 41 OCD patients with low consummatory anhedonia(OCD-LCA) a

Altered brain structure and function is evident in adults with multisite chronic pain. Although many such adults trace their pain back to childhood, it has been difficult to disentangle whether central nervous system alterations precede or are consequences of chronic pain. If the former is true, aberrant brain activity may identify children vulnerable to developing chronic pain later in life. We examined structural and functional brain magnetic resonance imaging metrics in a subset of children from the first 2 assessments of the Adolescent Brain and Cognitive Development Study. Children (aged

Narcissistic Personality Disorder (NPD) entails severe impairments in interpersonal functioning that are likely driven by self-beneficial and exploitative behavior. Here, we investigate the underlying motivational and neural mechanisms of prosocial decision-making by experimentally manipulating motivational conflict between self-beneficial and prosocial incentives. One group of patients diagnosed with NPD and a group of healthy controls (CTL) were scanned using functional magnetic resonance imaging while performing a prosocial decision-making task. In this task, we systematically varied the le

Multiple sclerosis and neuromyelitis optica spectrum disorders are both neuroinflammatory diseases and have overlapping clinical manifestations. We developed a convolutional neural network model that differentiates between the two based on magnetic resonance imaging data. Thirty-five patients with relapsing-remitting multiple sclerosis and eighteen age-, sex-, disease duration-, and Expanded Disease Status Scale-matched patients with anti-aquaporin-4 antibody-positive neuromyelitis optica spectrum disorders were included in this study. All patients were scanned on a 3-T scanner using a multi-d

INTRODUCTION: Alterations in narrow-band spectral power of electroencephalography (EEG) recordings are commonly reported in patients with schizophrenia (SZ). It is well established however that electrophysiological signals comprise a broadband scale-free (or fractal) component generated by mechanisms different from those producing oscillatory neural activity. Despite this known feature, it has not yet been investigated if spectral abnormalities found in SZ could be attributed to scale-free or oscillatory brain function.
METHODS: In this study, we analyzed resting-state EEG recordings of 14 SZ

Congenital diaphragmatic hernia (CDH) is associated with significant mortality due to lung hypoplasia and pulmonary hypertension. The role of embryonic pulmonary innervation in normal lung development and lung maldevelopment in CDH has not been defined. We hypothesize that developmental defects of intrapulmonary innervation, in particular autonomic innervation, occur in CDH. This abnormal embryonic pulmonary innervation may contribute to lung developmental defects and postnatal physiological derangement in CDH. To define patterns of pulmonary innervation in CDH, human CDH and control lung auto

The neurotransmitter serotonin (5HT) possesses developmental functions in vertebrates and invertebrates. Rodent embryos express 5HT receptors even before neural development, but the role of this neurochemical seems to be particularly important during axonal morphogenesis and differentiation and in neural crest cell migration. Moreover, 5HT inhibitors are teratogenic in mammals, inducing brain and heart abnormalities. The aim of this study was to investigate the effects of nonphysiological concentrations of 5HT (5HT excess as well as deprivation) on developing rat neural cells using the microma

Many animals in different mammalian orders display skilled forelimb use for prehension, but there has been little study of the rules underlying skilled forelimb use that may provide insight into its neural organization. Inflexibility of movement would imply an underlying fixed neural control characteristic of innate action pattern, whereas flexibility of movement would imply more plastic neural control. In the present study, rat reaching was examined by analyzing movements used to obtain nine different sized food pellets, weighing between 20 and 1000 mg. The prediction was that if the rats' re

BACKGROUND: Social Anxiety Disorder (SAD) is a prevalent mental disorder characterized by altered processing of social stimuli. Intolerance of Uncertainty (IU) has been linked to social anxiety and influences the perception of social information. Yet, the neural correlates of IU in SAD during the processing of emotional stimuli remain insufficiently understood.
METHODS: In this study, fMRI data were collected from 25 individuals with SAD and 25 healthy controls (HC), who completed a face processing task. Differential associations between the IU subdimensions prospective IU, inhibitory IU, and

Conversations can feel effortlessly engaging or, conversely, difficult and unrewarding. Multiple factors contribute to the experienced quality and outcomes of a conversation, among them how interlocutors align with each other. The present study investigated speech-to-speech, brain-to-speech, and brain-to-brain coordination as markers of interpersonal alignment, examining their relationship with jointly perceived interaction quality and mutual affinity between conversational partners. Pairs of previously unacquainted participants (dyads) engaged in multiple short, free-form conversations on top

The dual task cost of gait (DTC) is an accessible and cost-effective test that can help identify individuals with cognitive decline and dementia. However, its neural substrate has not been widely described. This study aims to investigate the neural substrate of the high DTC in older adults across the spectrum of cognitive decline. A total of 336 individuals from the GAIT study cohort were analyzed, including cognitively healthy (N = 122, 71 ± 3.6 years), those with mild cognitive impairment (N = 168, 71 ± 5.3 years), and those with dementia (N = 46, 80 ± 5.7 years). A DTC of 20% or greater was

Neural decoding of others' facial expressions is critical in social interactions and has been investigated using scalp event related potentials (ERPs). However, the impact of task and emotional context congruency on this neural decoding is unclear. Previous ERP studies employed classic statistical analyses that only focused on specific electrodes and time points, which inflates type I and type II errors. The present study re-analyzed the study by Aguado et al. (2019) using robust data-driven Mass Univariate Statistics across every time point and electrode and rejected trials with early reactio

Thyroid hormones (THs) are essential for brain development, and their dysregulation is associated with cognitive deficits and neurodevelopmental disorders. Down syndrome (DS), caused by trisomy 21, is frequently associated with thyroid dysfunction and impaired neurogenesis. Here, we investigated THs signaling dynamics during neural differentiation using human induced pluripotent stem cells (hiPSCs) derived from individuals with DS and controls. We analyzed the gene expression of key THs regulators-deiodinases, transporters, and receptors-and downstream target genes in hiPSCs, hiPSC-derived neu
