Restricted expression of protocadherin 2A in the developing mouse brain.

Protocadherins are cell-cell adhesion molecules that are thought to be involved in neural development. Here, we report the expression pattern of protocadherin 2A (Pc2A) in the developing mouse brain as determined by the in situ hybridization technique. In the postnatal day 2 brain, various regions expressed Pc2A including the cerebellar cortex, ventral posterior thalamic nucleus, dorsal lateral geniculate nucleus, hippocampus and cerebellum (Purkinje cells). In particular, some ependymal cells that form the lining of the lateral ventricle and the third ventricle and floor plate cells lining th

Effects of prenatal ethanol exposure on neuronal migration, neuronogenesis and brain myelination in the mice brain.

BACKGROUND: One of the most severe consequences of maternal alcohol consumption is the damage to the developing central nervous system. MATERIAL AND METHODS: To evaluate the effect of prenatal alcohol exposure on neuronal migration, neuronogenesis and myelination in the brain, pregnant BALB/C mice were maintained on either liquid diets containing ethanol (12 g/kg body weight) beginning on gestation day 6 or isocoloric diet (control group). The dams were sacrificed on gestation day 17 and the brain sections of the pups were analyzed using immunohistochemical method to evaluate the number of ne

Role of GPI-anchored NCAM-120 in rabies virus infection.

Although the neural cell adhesion molecule (NCAM) -140 and -180 have been shown to serve as a receptor for rabies virus (RV), it was not known whether the other major isoform of NCAM, GPI-anchored NCAM-120 functions as RV receptor. In this study, we have established HEp-2 cells stably expressing NCAM-120 or NCAM-140, and their susceptibilities to RV infection were compared. The results demonstrated that NCAM-120 served as virus attachment protein; however, the cells expressing NCAM-120 did not support efficient RV replication. Furthermore, the level of IFN-ss mRNA was apparently elevated in NC

L1CAM enhances autophagy via inhibition of p38 phosphorylation in colorectal cancer development and progression.

BACKGROUND: Colorectal cancer (CRC) is a common gastrointestinal tumor with a high incidence worldwide. L1 cell adhesion molecule (L1CAM) is highly expressed in CRC tissues and can promote tumor growth and metastasis. Autophagy regulates oncogenesis and tumor progression through context-dependent mechanisms and is regulated by multiple signaling pathways, including the p38 MAPK signaling pathway. OBJECTIVE: However, the relationship between L1CAM and autophagy is not known yet. In this study, we are trying to dissect the role of L1CAM-autophagy interaction in the development and progression o

Change in neuron aggregation and neurite fasciculation on EVAL membranes modified with different diamines.

In this study, we modified poly(ethylene-co-vinyl alcohol) (EVAL) membranes by the covalent bonding of diamines via epoxidation of surface hydroxyl groups of EVAL to analyze the effect of immobilized diamines with different carbon chain length on the cultured cerebellar granule neurons. Morphological studies showed that neurons seeded on the diamine-immobilized EVAL membrane were able to survive and regenerate with formation of an extensive neuritic network. Furthermore, cultured neurons showed that the presence of diamine with different carbon chain length was able to effectively regulate the

Fabrication and Evaluation of a Xenogeneic Decellularized Nerve-Derived Material: Preclinical Studies of a New Strategy for Nerve Repair.

The repair and regeneration of transected peripheral nerves is an important area of clinical research, and the adhesion of anastomosis sites to surrounding tissues is a vital factor affecting the quality of nerve recovery after nerve anastomosis. This study involves the generation of a novel nerve repair membrane derived from decellularized porcine nerves using a unique, innovative technique. The decellularized nerve matrix was verified to be effective in eliminating cellular components, and it still retained some neural extracellular matrix components and bioactive molecules (collagens, glyco

Novel mechanisms and approaches in the study of neurodegeneration and neuroprotection. a review.

Cellular mechanisms involved in neurodegeneration and neuroprotection are continuing to be explored, and this paper focuses on some novel discoveries that give further insight into these processes. Oligodendrocytes and activated astroglia are likely generators of the pro-inflammatory cytokines, such as the tumor necrosis factor family and interleukin family, and these glial support cells express adhesion receptors (e.g., VCAM) and release intercellular adhesion molecules (ICAM) that have a major role in neuronal apoptosis. Even brief exposure to some substances, in ontogeny and sometimes in ad

Bi-allelic variants in FSD1L cause a neurodevelopmental disorder overlapping with L1 syndrome.

Disruption of the complex processes underlying central nervous system development leads to a broad spectrum of brain malformations and neurodevelopmental disorders, often with a genetic cause. Here, we report bi-allelic pathogenic variants in fibronectin type III and SPRY domain-containing 1-like (FSD1L), encoding a protein of unknown function, in eleven individuals, including five fetuses from six unrelated families. The phenotype ranges from severe hydrocephalus, corpus callosum agenesis, and absent pyramid decussation to a neurodevelopmental syndrome characterized by severe intellectual dis

Role of CNTN6 in neurodevelopment and neuropathology.

Contactin 6 (CNTN6) is a recently discovered member of the contactins family, which belongs to a group of cell adhesion molecules. This review summarizes the current knowledge about the possible functions of CNTN6 in the organism and its manifestations in animal models and human diseases. Histological, cellular, and molecular studies in rodents have shown the involvement of this protein in neurite guidance, neural network development, and oligodendrocytogenesis. The expression levels in the cerebellum, hippocampus, and visual cortex of rodents vary depending on the period of neurodevelopment.

Cell body clustering drives gap junction-mediated synchronous activity in command neurons.

The nervous system contains densely packed cell bodies, yet the role of neuronal cell body position in circuit function is poorly understood. Here we show that four Drosophila Moonwalker Descending Neurons (MDNs), command neurons for backward locomotion, must maintain cell body contact to allow gap junction-dependent synchronous activity necessary to initiate backward walking. MDNs express the transcription factor Hunchback, which drives expression of the Lar cell adhesion molecule; Hunchback, Lar, and its ligand Dlp promote MDN cell body clustering and backward walking. When clustered, the ga

Carcinoid tumors of the urinary bladder. Immunohistochemical study of 2 cases and review of the literature.

We report the clinical, histopathologic, and immunophenotypic features of 2 carcinoid tumors of the urinary bladder and review the literature. Both tumors were located in the bladder neck, presented with hematuria, were small, and appeared to be completely excised cystoscopically. The tumors were smooth-surfaced sessile polypoid nodules covered by urothelium. Both tumors had glandular architecture. One tumor had regular bland nuclei, and the other had focal moderate nuclear pleomorphism. Neuroendocrine differentiation was readily confirmed by the immunohistochemical markers chromogranin, synap

Perforin- and granulysin-mediated cytotoxicity and interleukin 15 play roles in neurocognitive impairment in patients with acute lymphoblastic leukaemia.

Acute lymphoblastic leukaemia (ALL) is an aggressive disease. The course of disease is regulated by pro-inflammatory agents, and malignant cell infiltration of tissues plays a deleterious role in disease progression, greatly impacting quality of life, especially in the cognitive domains. Our hypothesis is that significant serum concentrations of interleukin 15 (IL-15) are responsible for higher expression of adhesion molecules on endothelial cells of blood-brain barrier (BBB) which allow leukaemia cells and/or normal lymphocytes the infiltration into the brain. In brain tissue these cells coul

Persistent Expression of VCAM1 in Radial Glial Cells Is Required for the Embryonic Origin of Postnatal Neural Stem Cells.

During development, neural stem cells (NSCs) undergo transitions from neuroepithelial cells to radial glial cells (RGCs), and later, a subpopulation of slowly dividing RGCs gives rise to the quiescent adult NSCs that populate the ventricular-subventricular zone (V-SVZ). Here we show that VCAM1, a transmembrane protein previously found in quiescent adult NSCs, is expressed by a subpopulation of embryonic RGCs, in a temporal and region-specific manner. Loss of VCAM1 reduced the number of active embryonic RGCs by stimulating their premature neuronal differentiation while preventing quiescence in

CAMs and Igs: cell adhesion and the evolutionary origins of immunity.

The lymphoid system and cells of immunity are as morphologically well defined as those of any complex organ but in addition they show dynamic long-range interactions between the fluid tissues (lymphocytes, monocytes, etc.) and the solid, vascular and generative tissues and organs which they comprise. Given the observation that CAMs are present in epithelial components of lymphoid organs, it appears that, in their ontogeny, the organs of immunity will share a common principle of morphoregulation by CAMs with brains, feathers and other parts of the phenotype. As discussed here, this principle is

Grip and slip of L1-CAM on adhesive substrates direct growth cone haptotaxis.

Chemical cues presented on the adhesive substrate direct cell migration, a process termed haptotaxis. To migrate, cells must generate traction forces upon the substrate. However, how cells probe substrate-bound cues and generate directional forces for migration remains unclear. Here, we show that the cell adhesion molecule (CAM) L1-CAM is involved in laminin-induced haptotaxis of axonal growth cones. L1-CAM underwent grip and slip on the substrate. The ratio of the grip state was higher on laminin than on the control substrate polylysine; this was accompanied by an increase in the traction for

The optic nerve head in glaucoma: role of astrocytes in tissue remodeling.

Primary open angle glaucoma is a common eye disease characterized by loss of the axons of the retinal ganglion cells leading to progressive loss of vision. The site of damage to the axons is at the level of the lamina cribrosa in the optic nerve head. The mechanism of axonal loss is unknown but elevated intraocular pressure and age are the most common factors associated with the disease. Previous studies in human glaucoma and in experimental glaucoma in monkeys have established a relationship between chronic elevation of intraocular pressure and remodeling of the optic nerve head tissues known

A developmental study of soluble L1.

L1 is a neural cell adhesion molecule expressed by neurons and it is involved in cell interactions during axon elongation and fasciculation. L1 from rat brain consists of a membrane-inserted Mr 200,000 polypeptide from which two polypeptides of Mr 180,000 and Mr 140,000 can be derived. These latter polypeptides appear both as membrane-associated and as soluble molecules. In this report, both total and soluble L1 in rat brain have been quantified by an enzyme-linked immunosorbent assay. The amount of total L1 per gram brain varies with postnatal age showing a peak value at postnatal day 7. The

Substrate-bound human recombinant L1 selectively promotes neuronal attachment and outgrowth in the presence of astrocytes and fibroblasts.

Axonal pathfinding is a complex process that is mediated through cell-matrix and cell-cell interactions. A large number of studies have demonstrated that ECM and ECM-derived proteins and peptides are potent promoters of neurite outgrowth, however much less attention is given to the fact that these same ligands also elicit responses in a wide variety of non-neuronal cell types. We examined the use of a substrate-bound recombinant form of human L1, an integral membrane protein, as a ligand for bridging materials for repairing the CNS by studying its effectiveness in promoting specific responses
