An 124I-radiolabeled exosome-based treatment platform for enhanced boron neutron capture therapy: precise delivery and real-time PET tracking of boron.

Boron neutron capture therapy (BNCT) is a promising targeted radiotherapy modality, whose efficacy critically depends on the precise spatiotemporal synchronization of sufficient boron accumulation in tumor cells with neutron irradiation. To achieve this coordination, we developed an integrated treatment platform based on exosome-delivered 10B-containing carbon dots (BCDs), which allows tracking via positron emission tomography (PET) imaging. We first established a screening system focusing on various key parameters, including blood-brain barrier (BBB) permeability, glioma-targeting specificity

Comparative biological evaluation of [18F]fluciclovine, [18F]FBPA-Fr, and [18F]FET for boron neutron capture therapy: Preclinical validation and clinical feasibility.

PURPOSE: The PET tracer 4‑borono-2-[18F]-fluoro-phenylalanine fructose ([18F]FBPA-Fr) is commonly used to estimate the biodistribution of L-4-dihydroxy-borylphenylalanine fructose (BPA-Fr). However, its limited regulatory approval has prompted the exploration of alternative imaging agents. In this study, we evaluated the feasibility of [18F]fluciclovine and [18F]fluoroethyl-tyrosine ([18F]FET) PET for pre-BNCT assessment, with direct comparison to [18F]FBPA-Fr. METHODS: In vitro cellular uptake studies were performed using two different cancer cell lines. In vivo microPET/MR imaging and biodi

Lyophilized tumour cell-loaded 10B-doped carbon dots for simultaneous boron neutron capture therapy and enhancement of antitumor immunity of prostate cancer.

10B agents are critical components in the application of boron neutron capture therapy (BNCT) for tumor treatment. The development of a multi-functional 10B agent plays an important role in the application of BNCT. This study presents a pioneering lyophilized-tumour-cell-based approach for the enhanced accumulation of 10B-doped carbon dots (10B-CDs) within tumour tissue, irradiating thermal neutrons to treat prostate cancer. In this platform, lyophilized RM-1 cells function as both tumor cell antigens and delivery vehicles for 10B-CDs. This design facilitates the co-delivery of tumor antigens …

Internal radiation dose estimates in organs of Wistar rats exposed to sprayed neutron-activated 31SiO2 microparticles: first results of international multicenter study.

Neutron-activated 31Si is an almost pure beta emitter and is one of the short-lived radionuclides, including beta-gamma emitter 56Mn, which were created in a form of residual radioactivity in the early period after the atomic bombing of Hiroshima and Nagasaki. The features of the biological effects of internal irradiation by these radionuclides are a subject of scientific discussions and research. The publication presents data on internal radiation doses in experimental Wistar rats that were exposed to sprayed neutron-activated microparticles of 31SiO2. Doses of internal radiation could be con

The structure of aqueous solutions of hexafluoro-iso-propanol studied by neutron diffraction with hydrogen/deuterium isotope substitution and empirical potential structure refinement modeling.

Neutron diffraction measurements of H/D isotopic substitution are made at room temperature for seven H/D substituted hexafluoro-iso-propanol (HFIP; 1,1,1,3,3,3-hexafluoro-2-propanol)-water mixtures at 0.1, 0.2, and 0.4 HFIP mole fraction (xHFIP). The eight partial structure factors except for the H(CH)-H(CH) pair obtained are subjected to an empirical potential structure refinement (EPSR) method to derive all site-site pair correlation functions. It is found that with increasing HFIP concentration the ice-like network of water disappears between xHFIP = 0.1 and 0.2, followed by the formation o

Relative Biological Effectiveness and Non-Poissonian Distribution of Dicentric Chromosome Aberrations following Californium-252 Neutron Exposures of Human Peripheral Blood Lymphocytes.

Cells exposed to fast neutrons often exhibit a non-Poisson distribution of chromosome aberrations due to the high ionization density of the secondary reaction products. However, it is unknown whether lymphocytes exposed to californium-252 (252Cf) spectrum neutrons, of mean energy 2.1 MeV, demonstrate this same dispersion effect at low doses. Furthermore, there is no consensus regarding the relative biological effectiveness (RBE) of 252Cf neutrons. Dicentric and ring chromosome formations were assessed in human peripheral blood lymphocytes irradiated at doses of 12-135 mGy. The number of aberra…

Induction of chromosome aberrations in G0 human lymphocytes by low doses of ionizing radiations of different quality.

Human peripheral blood lymphocytes from two donors were exposed to low doses (0.05 to 2.0 Gy) of gamma rays, X rays, or fast neutrons of different energies. Chromosome aberrations were analyzed in metaphase of first-division cells after a culture time of 45-46 hr. At this time, less than 5% of the cells were found in second division. Different dose-response relationships were fitted to the data by using a maximum likelihood method; best fits for radiation-induced dicentric aberrations were obtained with the linear-quadratic law for all radiations. The linear component of this equation predomin…

Boron neutron capture therapy for recurrent pediatric sialoblastoma: the first clinical experience and dosimetric considerations for skeletal safety with 4-year follow-up.

Sialoblastoma is a rare, aggressive pediatric salivary gland tumor often refractory to conventional therapies, which carry significant risks of growth impairment in children. We report the 52-month follow-up of a 6-year-old girl with recurrent sialoblastoma treated with two sessions of boron neutron capture therapy (BNCT). She achieved a 23-month complete response after the first session and a sustained partial response following the second. Dosimetric analysis using a conservative α/β ratio of 2.0 Gy for late-responding tissues showed that the equivalent dose in 2-Gy fractions to the mandible

Comparison of dose distribution with and without reflecting heterogeneous boron distribution using 18F-BPA positron emission tomography in boron neutron capture therapy.

This study investigated the dosimetric impact of implementing heterogeneous boron distribution into dose evaluations on tumors in BNCT. The study involved 27 patients who underwent 18F-BPA PET/CT scans. Dose evaluations were performed using various methods reflecting boron concentrations on CT images. The conventional dose evaluations, which reflected uniform boron concentration in blood of 25 ppm and a constant tumor-to-blood (T/B) ratio of 3.5, were compared with individual dose evaluations, which reflected the blood boron concentration and T/B ratio calculated from 18F-BPA in each patient.

Rational surface modification of gadolinium borate nanoparticles enhancing colloidal stability in physiological media for potential neutron capture therapy and magnetic resonance imaging.

Colloidal stability of nanomaterials in physiological media is an indispensable property for their biomedical applications. However, gadolinium borate (GdBO3) nanoparticles that hold promise as a theranostic agent for neutron capture therapy (NCT) and magnetic resonance imaging (MRI) of cancer tend to precipitate in phosphate buffered saline (PBS) owing to formation of insoluble gadolinium phosphate. To address this issue, in this work 10B-enriched GdBO3 nanoparticles were prepared and coated with mesoporous silica (mSiO2) of ~ 40 nm in thickness and subsequently grafted with hydrophilic polyg
