Regulatory Mechanisms and Synergistic Enhancement of the Diiron YtfE Protein in Nitric Oxide Reduction.

The diiron-containing YtfE protein in Escherichia coli is pivotal in counteracting nitrosative stress, a critical barrier to bacterial viability. This study delves into the biochemical complexity governing YtfE's conversion of nitric oxide (NO) to nitrous oxide, a key process for alleviating nitrosative stress. Through site-directed mutagenesis, we explored YtfE's molecular structure, with a particular focus on two internal transport tunnels important for its activity. Our findings illuminate Tunnel 1 as the primary conduit for substrate transport, regulated by conformational shifts within the

Differential effects of synaptic and extrasynaptic NMDA receptors on Aβ-induced nitric oxide production in cerebrocortical neurons.

Oligomerized amyloid-β (Aβ) peptide is thought to contribute to synaptic damage, resulting in dysfunctional neuronal networks in patients with Alzheimer's disease. It has been previously suggested that Aβ may be detrimental to neuronal health, at least in part, by triggering oxidative/nitrosative stress. However, the mechanisms underlying this process remain to be elucidated. Here, using rat primary cerebrocortical cultures, we demonstrate that Aβ1-42 oligomers trigger a dramatic increase in intracellular nitric oxide (NO) concentration via a process mediated by activation of NMDA-type glutama

Luteolin ameliorates cisplatin-induced nephrotoxicity in mice through inhibition of platinum accumulation, inflammation and apoptosis in the kidney.

The aim of this study was to investigate the effects of flavone luteolin against cisplatin (CP)-induced kidney injury in mice. Luteolin at doses of 10mg/kg was administered intraperitoneally (ip) once daily for 3 days following single CP (10 or 20mg/kg) ip injection. Mice were sacrificed 24h after the last dose of luteolin. The CP treatment significantly increased serum creatinine and blood urea nitrogen and induced pathohistological changes in the kidneys. Renal oxidative/nitrosative stress was evidenced by decreased glutathione (GSH) levels and increased 3-nitrotyrosine (3-NT) and 4-hydroxyn

CYP2E1 potentiates toxicity in obesity and after chronic ethanol treatment.

CYP2E1 activates several hepatotoxins and contributes to alcoholic liver damage. In this report, we review our studies on whether induction of CYP2E1 can potentiate liver injury in obesity. Acetone- or pyrazole-induced severe liver injury in obese mice as compared to obese controls and lean mice. Severe liver injury was associated with elevated oxidative and nitrosative stress and could be blunted by inhibitors of CYP2E1 and inducible nitric oxide synthase (iNOS). S-Adenosyl-L-methionine (SAM) lowered the elevated oxidative and nitrosative stress, steatosis, liver injury and mitochondrial dysf

Effects of oxygen on the reactivity of nitrogen oxide species including peroxynitrite.

This paper describes the O(2)-dependent control of the reactivity of nitrogen oxide species for the production of biologically important nitrated and nitrosated compounds. In this study, the effects of O(2) on the reactivity of NO, NO(2), and ONOO(-)/ONOOH for nitration of tyrosine (Tyr) and nitrosation of glutathione (GSH) and morpholine (MOR) were examined. NO produced S-nitrosoglutathione (GSNO) and N-nitrosomorpholine (NMOR) through the formation of N(2)O(3) under aerobic conditions, and NO(2) produced 3-nitrotyrosine (3-NO(2)Tyr), GSNO, and NMOR. Transnitrosation from GSNO to MOR was obse…

Differential cytostatic effects of NO donors and NO producing cells.

A 3-h exposure to NO donors (spermine-NO, DETA-NO, or SNAP), or to NOS II-expressing cells (activated macrophages or EMT6 cells) reversibly inhibited DNA synthesis in K562 tumor cells. In GSH-depleted K562 cells, cytostasis remained reversible when induced by DETA-NO or NOS II activity, but became irreversible after exposure to spermine-NO or SNAP. Only SNAP and spermine-NO efficiently inhibited GAPDH, an enzyme with a critical thiol, in GSH-depleted cells. Thus, the irreversible cytostasis induced in GSH-depleted cells by spermine-NO or SNAP can be tentatively attributed to S-nitrosating or o

A novel type of hemoglobin confers host-derived nitric oxide resistance to the opportunistic pathogen Acinetobacter baumannii.

Acinetobacter baumannii is an opportunistic Gram-negative pathogen responsible for various infections, such as those of the bloodstream and lungs, which often resist antibiotic therapy. In the course of an infection, the human innate immune system's phagocytic cells are activated producing nitric oxide (NO) that cause bacterial injury. While the antimicrobial effects of nitrosative stress and the bacterial resistance mechanisms are well-characterized for several pathogens, the adaptations of Acinetobacter spp. to NO have not been studied. In this work, we define the transcriptional response of

A sequence-activatable dual-locked fluorescent probe for simultaneous detection of hypochlorous acid and peroxynitrite during drug-induced liver injury.

Drug-induced liver injury (DILI) is a crucial factor that poses a significant threat to human health. DILI process leads to the changes of reactive oxygen species and reactive nitrogen species content in cells, which leads to oxidative and nitrosative stress in cells. However, the high reactivity of hypochlorous acid (HOCl) and peroxynitrite (ONOO⁻), combined with a lack of in situ imaging techniques, has hindered a detailed understanding of their roles in DILI. Therefore, this paper reports a novel sequence-activatable dual-locked molecular probe HA-P3 for the identification and imaging of tw

Essential Oils Downregulate Pro-Inflammatory Cytokines and Nitric Oxide-Mediated Oxidative Stress in Alloxan-Induced Diabetogenic Rats.

INTRODUCTION: Hyperglycemia is associated with an elevated level of reactive nitrogen species (RNS) that leads to nitrosative stress and exacerbates the progression of diabetic complications. METHODS: Present study was aimed to evaluate the therapeutic effects of essential oils (EOs) on increased serum levels of nitric oxide (NO) in diabetogenic rats. Diabetogenic rats were treated with EOs separately and/or in combination at the dose of 100 mg/kg, orally for one month. Blood sampling was done at the 1st, 15th and 30th day of the treatment period to investigate the effect of treatment on biom

S-nitrosylated TDP-43 triggers aggregation, cell-to-cell spread, and neurotoxicity in hiPSCs and in vivo models of ALS/FTD.

Rare genetic mutations result in aggregation and spreading of cognate proteins in neurodegenerative disorders; however, in the absence of mutation (i.e., in the vast majority of "sporadic" cases), mechanisms for protein misfolding/aggregation remain largely unknown. Here, we show environmentally induced nitrosative stress triggers protein aggregation and cell-to-cell spread. In patient brains with amyotrophic lateral sclerosis (ALS)/frontotemporal dementia (FTD), aggregation of the RNA-binding protein TDP-43 constitutes a major component of aberrant cytoplasmic inclusions. We identify a pathol

Influence of cardiometabolic comorbidities on myocardial function, infarction, and cardioprotection: Role of cardiac redox signaling.

The morbidity and mortality from cardiovascular diseases (CVD) remain high. Metabolic diseases such as obesity, hyperlipidemia, diabetes mellitus (DM), non-alcoholic fatty liver disease (NAFLD) and non-alcoholic steatohepatitis (NASH) as well as hypertension are the most common comorbidities in patients with CVD. These comorbidities result in increased myocardial oxidative stress, mainly from increased activity of nicotinamide adenine dinucleotide phosphate oxidases, uncoupled endothelial nitric oxide synthase, mitochondria as well as downregulation of antioxidant defense systems. Oxidative an

Nephro-protective activity of wheatgrass juice against alcohol-induced oxidative damage in rats.

The present study is an attempt to check the protective effect of fresh wheatgrass juice (WJ) as a potential therapeutic agent against alcohol-induced oxidative/nitrosative stress leading to nephrotoxicity in rats. Administration of 20% ethanol (5 g/kg b.wt/day) for 60 days resulted in a significant rise in the plasma concentrations of urea and creatinine with decreased levels of uric acid. Besides, a significant decrease in plasma electrolyte/mineral levels along with decreased activity of Na+/K+-ATPase activity was recorded in alcohol administered rats. In alcohol administered rats augmented

S-adenosyl-L-methionine decreases the elevated hepatotoxicity induced by Fas agonistic antibody plus acute ethanol pretreatment in mice.

The current study was designed to investigate the effect and potential mechanism of exogenous administration of S-adenosyl-l-methionine (SAM) on the enhanced hepatotoxicity induced by the Fas agonistic Jo2 antibody plus acute ethanol pretreatment in C57BL/6 mice. Acute ethanol plus Jo2 treatment produces liver toxicity under conditions in which ethanol alone or Jo2 alone do not. SAM significantly attenuated this elevated hepatotoxicity in mice as manifested by a decrease of serum aminotransferases and morphological amelioration. Levels of SAM and activity of methionine adenosyltransferase were

A novel family of S-nitrosothiols: chemical synthesis and biological actions.

S-Nitrosothiols are a class of chemical compounds that decompose to release nitric oxide and show promise in the treatment of a variety of cardiovascular diseases. Some of these are present in vivo and others have been synthesized in vitro. However, those discovered or synthesized to date have very little tissue selectivity or specificity. We synthesized a number of novel S-nitrosated dipeptides of high purity and examined their effects on vasorelaxation using rat mesenteric arteries and on inhibition of platelet aggregation using platelets from healthyhuman subjects. For comparison, we also t

Effect of Peroxynitrite on Endothelial-Mesenchymal Transition Markers in Primary Endothelial Cell Cultures In Vitro.

We studied the effects of the nitrosative stress modulator peroxynitrite on changes in key markers of the endothelial-mesenchymal transition (EndMT) in primary endothelial cell cultures in vitro. Peroxynitrite exposure significantly increased the relative content of endothelial markers (platelet adhesion molecules (CD31) and von Willebrand factor) and reduced the levels of mesenchymal markers (fibronectin and vimentin). These findings suggest that peroxynitrite may inhibit EndMT in vitro.

A BODIPY-based fluorescent probe for rapid detection of NO in cells and zebrafish.

Nitric oxide (NO) is a small biological molecule that plays a critical role in numerous physiopathological processes throughout the human body. An extensive array of studies has elucidated a robust correlation between pathological conditions and endogenous levels of NO. In this paper, we synthesized a BODIPY-based fluorescent probe, dubbed BDD, which has excellent ability to accurately detect NO. The presence of secondary amines of BDD enables it to engage in an N-nitrosation reaction with NO. This chemical interaction effectively disrupts the initial photoinduced electron transfer (PET) proce…

Small-molecule-catalysed deamination enables transcriptome-wide profiling of N6-methyladenosine in RNA.

The deamination reaction is important to both fundamental organic chemistry and biochemistry. Traditional chemical methods of deamination rely on the use of aryldiazonium salts under harsh acidic conditions, which limits the application scope for most biological substrates. Here we present an N-nitrosation strategy for deamination under mild conditions that DNA and RNA biological macromolecules can tolerate. Cooperative catalysis combining a carbonyl organocatalyst with a Lewis acid catalyst facilitates the formation of a carbon-nitro intermediate from a primary amine, which, on rearrangement …
