Morphological analysis of dentine formation in young rat molars during the recovery phase with calcium alone or combined with xylitol following a low-calcium dietary regimen.

The effects of dietary calcium deficiency and subsequent replenishment of the diet with calcium alone or with xylitol were studied. Thirty 3 … roup and did not differ from the Ca-deficient period. These results indicate that in rat molars the odontoblasts can accelerate the rate of dentine formation when calcium is restored to the diet, at least during primary dentinogenesis. This effect was not seen when 5% xylitol was added to the replenishment diet.

Early cellular events in an actinomycin D-created dentin niche in the rat incisor.

Administration of actinomycin D at a dose level of 0.375 microgram/g resulted in the selective disruption of developing odontoblasts at a critical stage of morphogenesis. A dentin niche was formed which was later repaired by cellular reparative dentin. The cellular changes which resulted in dentin niche formation were studied histologically and ultrastructurally in serial longitudinal and transverse sections from tissues obtained 10 h to 80 h following injection of the drug. Five stages were identified: initial destruction (10-20 h), rapid destruction (30-40 h), debris removal (50-60 h), proli…

Effect of bacterial products on inflammatory reactions in the dental pulp.

Culture filtrates (extracellular components) and material obtained from disintegrated cells (intracellular components) of cultured plaque ba … ual animals and differences in thickness of the remaining dentin. The controls showed damage to the odontoblasts but little or no neutrophil infiltration. The findings confirm that products of bacteria applied to exposed dentin initiate inflammatory reactions in the dental pulp.

Novel strategy for dental caries by physiologic dentin regeneration with CPNE7 peptide.

OBJECTIVE: CPNE7-derived functional peptide (CPNE7-DP) has been introduced as a bioactive therapeutics for dentin diseases. CPNE7-DP regener … ena could be the consequence of the modulation of autophagy by CPNE7-DP, which reactivates inflamed odontoblasts. CONCLUSIONS: Overall, CPNE7-DP, which repairs caries through physiological dentin regeneration, might help overcoming the limitations of current restorative caries treatments.

[Cephalic neural crests and disorders of craniofacial morphogenesis. Neurocristopathies (author's transl)].

To establish the pathogenicity, and attempt to define a classification of craniofacial malformations is a difficult task, both in animals and humans. Mention is often made of the classification developed by Paul Tessier, which according to this author "is neither based on a theory nor on an embryonic definition. It is the by-product of anatomical findings during clinical examination and operative dissections" (31). The authors propose an embryologic classification of facial malformations, based on disorders of embryonic development of cells derived from the neural crests (NC) of cephalic origi

Pulp condition of successfully formocresol-treated primary molars.

The aim of the study was to assess the state of the pulp of primary teeth successfully treated by formocresol pulpotomy. Nineteen teeth, all of them with clinically and radiographically successful treatments, were extracted 3-24 months after treatment. The pulpal condition had changed in all the teeth. Inflammatory reaction or necrosis was observed in all teeth. Dentinal resorption followed by apposition of hard tissue were common findings. In the teeth with vital tissue only in the apical area, the tissue had minor or no signs of inflammation. Microorganisms could be demonstrated in the necro

Compound Odontoma Associated with Multiple Odontogenic Abnormalities in the Mandible of a Child: A Case Report and Review of Literature.

UNLABELLED: Odontomas have been described as a dental abnormality that develops due to the overgrowth of certain components of the dental lamina, which eventually leads to the formation of a poorly structured calcified mass. The occurrence has been attributed to local trauma, infection, congenital defects, odontoblastic hyperactivity, or changes in the genetic material. Clinically, odontomas are largely asymptomatic and are diagnosed during routine examinations and procedures. However, they might frequently be associated with delayed eruption, impacted teeth, or missing teeth, warranting early

Dentin dysplasia: single-tooth involvement?

Dentin dysplasia is a genetic defect of dentin formation inherited as an autosomal dominant trait. It is characterized by normal enamel but atypical dentin formation with abnormal pulpal morphology. Once thought to be a single entity, dentin dysplasia has now been divided into type I (radicular) and II (coronal). Type I is by far the more common. Both types include multiple/generalized involvement of primary and permanent dentition. Combinations of both types have also been described in the literature. Four distinct forms of dentin dysplasia type I and 1 form of dentin dysplasia type II are id

Histopathological evaluation of pulpotomy with Er,Cr:YSGG laser vs formocresol.

The purpose of this study was to histologically investigate whether pulpotomy with Er,Cr:YSGG laser is an acceptable alternative for formocresol. Pulpotomy of 48 dog's primary canine teeth was performed with formocresol or Er,Cr:YSGG laser. Histological evaluations on hematoxylin and eosin-stained pulp tissues were made by an optical microscope 7 or 60 days later. Statistical analysis was performed with Fisher's exact test, Mann-Whitney U test, and Student's t test. Seven days after pulpotomy, samples treated with laser had significantly favorable histological features in the following measure

Pulpal tissue reaction to formocresol vs. ferric sulfate in pulpotomized rat teeth.

The purpose of this study was to assess histologically the pulpal healing process after pulpotomies in rat teeth were done using two different agents: 20% dilution of formocresol (FMC) and ferric sulfate (FS). Two criteria were used: degree of inflammation and extent of pulpal involvement. Zinc oxide-eugenol paste (ZOE) or polycarboxylate cement (PCX) were placed to seal the cavity preparations. The maxillary first molars of 120 Sprague-Dawley rats were treated in this study and were divided into four groups: 1) FMC + ZOE, 2) FS + PCX, 3) FMC + PCX, and 4) FS + ZOE. After treatment the animals

Healing of primate dental pulps capped with Teflon.

The pulps of Rhesus monkey teeth were exposed and capped with three materials: Teflon, a commercial hard-set calcium hydroxide (Ca(OH)2) material, and Ca(OH)2 plus saline. Experimental test periods were 3, 10, and 21 days, and 5 and 8 weeks. After treatment, the teeth were removed and processed for routine histologic evaluation. Teeth treated with the two Ca(OH)2 materials showed resolution of the inflammatory response and hard tissue formation at the exposure site as early as 10 days postoperatively, with consistent healing at 21 days and longer. Teflon had a similar soft tissue healing patte

Ultrastructural distribution of sulfated glycosaminoglycans in epithelial-mesenchymal interface of developing rat tooth germs.

We investigated the ultrastructural distribution of sulfated glycosaminoglycans in the epithelial-mesenchymal interface of tooth germs by use of the high-iron diamine thiocarbohydrazide silver proteinate (HID-TCH-SP) staining and enzymatic digestion method. At an early stage in odontoblast differentiation, HID-TCH-SP stain deposits were sparsely distributed in the basement membrane and in the intercellular spaces. Subsequently, as formation of the initial predentin matrix began, HID-TCH-SP stain deposits were densely distributed in the interfibrillar spaces and the basement membrane. Testicula

Lipoteichoic acid up-regulates VEGF expression in macrophages and pulp cells.

Vascular endothelial growth factor (VEGF) is a potent inducer of angiogenesis, vascular permeability, and edema. Up-regulation of VEGF expression in the dental pulp may result in increased intra-pulpal pressure, and contribute to pain and irreversible tissue damage. Lipoteichoic acid (LTA) is an amphiphilic molecule from Gram-positive bacteria that has been associated with the pathogenesis of pulpitis. To investigate if LTA regulates expression of VEGF, we exposed mouse odontoblast-like cells (MDPC-23), undifferentiated pulp cells (OD-21), fibroblasts, or macrophages to streptococcal LTA, and

Dynamics of the pulpo-dentin complex.

Dentin has a relatively high water content due to its tubular structure. Once dentin is exposed, this intratubular water is free to move in response to thermal, osmotic, evaporative, or tactile stimuli. Fluid shifts across dentin are thought to cause sufficient shear forces on odontoblasts, nerve endings, nearby fibroblasts, and blood vessels to cause significant mechanical irritation, disruption, or damage, depending on the magnitude of the fluid shift. Even in the absence of fluid shifts, the water-filled tubules provide diffusion channels for noxious (i.e., bacterial products) substances wh…

The granular layer of Tomes in experimental caries in rats.

In examination of ground sections of human third maxillary molar teeth, the granular layer of Tomes was shown to consist of expansion of dentinal tubules. The microradiographic density was decreased well inside the layer. It was postulated that the findings were expressions of dentin resorption. The theory was tested experimentally in rats fed a cariogenic diet with low calcium and phosphorus. The morphology of mandibular molar teeth was studied by electron probe microanalysis and by microradiography. It was concluded that the primary event in cariogenesis was an expansion of peripheral dentin

Histochemical evaluation of thiamine pyrophosphatase activity during first molar odontogenesis of the neonatal hairless mouse.

Localization of thiamine pyrophosphatase activity has been evaluated in the developing first molar of the neonatal hairless mouse. Postnatal animals from parturition to five days of age were decapitated and the severed heads frozen and sectioned in a frontal plane on a cryostat. 14 micron thick sections were fixed and subsequently incubated for thiamine pyrophosphatase activity according to the method of GOLDFISCHER et al. (1971). The tissue was visualized, dehydrated, cleared and mounted. Light microscopy was utilized in evaluating thiamine pyrophosphatase activity. Thiamine pyrophosphatase a
