Prevalence and screening of hemoglobinopathies and glucose-6-phosphate dehydrogenase deficiency in Yemeni blood donors.

OBJECTIVES: This study aimed to determine the prevalence of total blood cell abnormalities, hemoglobinopathies and G6PD deficiency and evaluate the efficacy of red blood cell (RBC) indices, mentzer index (MI) and naked-eye single tube red cell osmotic fragility (NESTROF) test as screening tools for diagnosis of β thalassemia trait among Yemeni blood donors. METHODS: A cross-sectional study was conducted with 106 volunteer blood donors who met the national standard criterion of blood donation. Various tests were performed, including complete blood count (CBC), serum ferritin, sickling test, G6…

Hemolytic anemia repressed hepcidin level without hepatocyte iron overload: lesson from Günther disease model.

Hemolysis occurring in hematologic diseases is often associated with an iron loading anemia. This iron overload is the result of a massive outflow of hemoglobin into the bloodstream, but the mechanism of hemoglobin handling has not been fully elucidated. Here, in a congenital erythropoietic porphyria mouse model, we evaluate the impact of hemolysis and regenerative anemia on hepcidin synthesis and iron metabolism. Hemolysis was confirmed by a complete drop in haptoglobin, hemopexin and increased plasma lactate dehydrogenase, an increased red blood cell distribution width and osmotic fragility,…

Factors affecting the hemolytic action of "lysolecithin" upon rabbit erythrocytes.

1. Lysolipid was prepared by the action of snake venom on egg yolk, and a study was made of the factors affecting its hemolytic action upon rabbit erythrocytes. 2. Lysis proceeded very rapidly at first, then ceased within a few minutes at room temperature. A given amount of lysin appeared to hemolyze a fixed number of cells, under specified conditions. 3. The more dilute erythrocyte suspensions required relatively more lysin per cell, for 50 per cent hemolysis of the suspension. There may be an equilibrium between the lysin dissolved in the medium and that adsorbed on the cells. 4. The degree

Anti-ischemic and membrane stabilizing activity of calmodulin inhibitors.

In the control perfused working rat hearts subjected to 25 min global ischemia, reperfusion resulted in a 50% recovery of the hemodynamic functions. A concentration-dependent improvement of this recovery and a reduction of the postischemic lactate dehydrogenase (LDH) release was caused by calmidazolium (CMZ), trifluoperazine (TFP), and chlorpromazine (CPZ) added prior to ischemia. The drugs were not effective when added only to the reperfusate. The concentrations of CMZ, TFP, and CPZ producing the half-maximal effects were 2.5 X 10(-9) M, 1.5 X 10(-7) M and 3 X 10(-7) M, respectively. Prolonga

Sickle-cell anaemia, sickle-cell thalassaemia, sickle-cell haemoglobin C disease, and asymptomatic haemoglobin C thalassaemia in one Ghanaian family.

A Ghanaian family is described in which a sickle-cell haemoglobin C man married to a sickle-cell thalassaemia woman produced 12 children (eight alive). Four children have sickle-cell anaemia, two sickle-cell haemoglobin C disease, one has sickle-cell thalassaemia, and one is asymptomatic haemoglobin C thalassaemia. It is emphasized that the contribution that adult sickle-cell disease patients make, through procreation, to the persistence of the S gene may be greater than is normally supposed, and that this contribution may soon outstrip that made by balanced polymorphism through falciparum mala

Pyrimidine-5'-Nucleotidase Deficiency: a New Homozygous NT5C3A Mutation (c.693+1G>A variant).

BACKGROUND: Erythrocytes have an average lifespan of 120 days, after which they are typically removed by macrophages in the reticuloendothel … ited macrocytic anemia and basophilic stippling on peripheral blood smear, with normal results from osmotic fragility tests and G6PD levels. Genetic testing revealed a homozygous c.693+1G>A variant in the NT5C3A gene, classified as possibly pathogenic based on ACMG criteria. This variant is linked to P5N deficiency, which aligns with the patient's clinical presenta …

Dietary supplementation with conjugated linoleic acid induces anemia with combined features of megaloblastic and hemolytic anemia.

This study investigates whether dietary supplementation with conjugated linoleic acids (CLA) induces anemia in mice. Male Kunming White mice … enhanced phagocytic effect of Kupffer cells on RBCs. No intergroup differences were observed in RBC osmotic fragility, plasma iron levels, or renal erythropoietin (EPO) mRNA expression. These findings suggest CLA-induced anemia may result from nutritional deficiencies and/or enhanced phagocytic activity of the monocyte-macrophage system. We conclude that dietary CL …

A novel variant of SLC4A1 for hereditary spherocytosis in a Chinese family: a case report and systematic review.

BACKGROUND: The incidence of hereditary spherocytosis (HS) is approximately 1:2000 in the western population, while it is much lower in the … bilirubin in the patient and his father. Furthermore, small globular red blood cells with increased osmotic fragility were observed. In particular, the eosin-5'-maleimide binding test provided the strong evidence that band 3 protein was deleted in the erythrocyte membrane. Next-generation sequencing (NGS) and Sanger sequencing further demonstrated a heterozygous no …

Study on the protection of dextran on erythrocytes during drug loading.

In comparison to other carriers, erythrocytes (red blood cells, RBCs) hold the advantages of unmatched long circulation, biocompatibility an … ug loading amount and lower the phosphatidylserine eversion rate. Moreover, it maintained a similar osmotic fragility to NRBCs and exhibited no effect on the morphological structure of drug-loaded RBCs. Laser confocal results showed tight covering of dextran over RBCs, which could explain the protective effects. The addition of dextran increased the activity of dru …

Novel compound heterozygous mutations in the SPTA1 gene, causing hereditary spherocytosis in a neonate with Coombs‑negative hemolytic jaundice.

Hereditary spherocytosis (HS) is a common heterogeneous type of inherited hemolytic anemia characterized by jaundice and splenomegaly. Diagnosis of HS in neonates is considered unreliable, and is generally based on positive family history, spherocytes in peripheral smears, increased osmotic fragility, and jaundice. In the present study, routine laboratory tests, next‑generation sequencing, and Sanger sequencing were applied to diagnose a neonatal patient with Coombs‑negative hemolytic jaundice. The neonate had no family history of HS; however, spherocytes were observed in peripheral smears, an…

NFAT5 Controls the Integrity of Epidermis.

The skin protects the human body against dehydration and harmful challenges. Keratinocytes (KCs) are the most abundant epidermal cells, and it is anticipated that KC-mediated transport of Na+ ions creates a physiological barrier of high osmolality against the external environment. Here, we studied the role of NFAT5, a transcription factor whose activity is controlled by osmotic stress in KCs. Cultured KCs from adult mice were found to secrete more than 300 proteins, and upon NFAT5 ablation, the secretion of several matrix proteinases, including metalloproteinase-3 (Mmp3) and kallikrein-related

[Effect of dibazol, euphylline, No-Spa and galidor on thrombocyte osmotic resistance and blood clot retraction].

The osmotic resistance of thrombocytes and the blood clot retraction are shown to diminish in the presence of nospanum and holidor in the plasma and blood. A double logarithmic relationship between the values characterizing the action on the osmotic resistance of thrombocytes and the blood clot refraction, on the one hand, and concentrations of some of the study substances, on the other, was revealed. With parenteral introduction the fall of the osmotic resistance of thrombocytes and an increase in the amount of the retracted serum were noted to occur with a protracted subcutaneous injection o

Gilbert's syndrome in patients with gallbladder stones.

Patients with Gilbert's syndrome suffer from an abnormality which makes them jaundiced from time to time. A number also develop gallstones and come to cholecystectomy. If this condition has not been recognized these patients may subsequently run the risk of unnecessary operations on their bile ducts from the mistaken assumption that the intermittent episodes of jaundice which are a feature of the syndrome are due to a stone which has been left behind. Such a case history is reported here.In an attempt to determine how frequently these conditions coexist a prospective study was carried out on p
