Cytokine profiles in adult mink infected with Aleutian mink disease parvovirus.

The aim of this study was to examine the levels of gamma interferon (IFN-gamma)-, interleukin 4 (IL-4)-, and IL-8-producing cells in peripheral blood mononuclear cells from mink infected with the Aleutian mink disease parvovirus (ADV). As expected, ADV-infected mink developed high plasma gamma globulin values (hypergammaglobulinemia) and enhanced quantities of CD8-positive (CD8(+)) cells in the blood during the infection. We quantified the percentages of IFN-gamma- and IL-4-positive lymphocytes and IL-8-positive monocytes up to week 38 after virus challenge. The results clearly indicated marke…

Long term follow up of serostatus after maternofetal parvovirus B19 infection.

BACKGROUND: Maternofetal parvovirus B19 infection may result in fetal hydrops or abortion. Chronic infection has been associated with long term complications (polyarthritis, persistent aplastic anaemia, hepatitis). In pregnancy maternal immunosuppression caused by a TH2 dominant response to viral antigens has been observed. There is little information on long term reactivity to intrauterine infection. AIMS: To assess the serological status in children and their mothers after maternofetal parvovirus B19 infection and development of fetal hydrops. METHODS: A total of 18 children and their moth…

Detection of human parvovirus B19 infection in first-trimester fetal loss.

OBJECTIVE: To investigate the frequency of parvovirus B19 infection in first-trimester fetal loss, as measured by B19 DNA polymerase chain reaction in placental tissue in a prospective descriptive study from a nonendemic area. METHODS: Placental tissues from first-trimester fetal losses were examined for presence of B19 DNA by polymerase chain reaction in a prospective study during 30 months. For comparison, placental tissues from second-trimester fetal losses, as well as from full-term normal pregnancies, were also studied. RESULTS: B19 DNA was detected by polymerase chain reaction in one o…

Thrombotic microangiopathy associated with parvovirus B 19 infection after renal transplantation.

Human parvovirus B19 is considered an etiologic agent of aplastic anemia in immunosuppressed patients. Microscopic vasculitis, with or without renal involvement, has recently been attributed to this viral infection in immunocompetent patients. This study describes four cases of thrombotic renal graft microangiopathy presumably secondary to B19 infection. Twelve to 50 days after transplantation, four patients presented a renal graft dysfunction with creatinine rising to 360 to 1088 micromol/L and requiring hemodialysis in three cases. Renal involvement appeared after a systemic illness characte…

[Human parvovirus B19 infection in a child suffering from chronic arthritis].

The pathogenicity of the human parvovirus B19 has been disclosed in the last two decades. Different syndromes (erythema infectiosum or hydrops fetalis) can be produced by this virus as well as aplastic crisis in patients with haemolytic anaemia. It is a common infection both in adults and in children. The frequency of joint symptoms in erythema infectiosum is about 8% in childhood but tenfold of it in the adults. However, the arthropathy could be the only symptom without constitutional complains or rash. The joint problems used to be solved after some weeks in most cases but occasionally they …

Detection of human parvovirus B19 DNA by nested polymerase chain reaction.

We describe the development of a nested polymerase chain reaction (PCR) technique used to detect human parvovirus B19 DNA. It was performed in a two-step reaction, first with a pair of outer primers, then with a pair of inner (ie, nested) primers. Primer sequences were selected in the VP1 gene, corresponding to the capsid protein, of human parvovirus B19. To establish the nested PCR assay, the plasmid pGEM-1 containing almost the entire coding sequence of a human parvovirus B19 isolate was used. The nested PCR could detect up to 1.8 x 10(-3) ag of B19 DNA, equivalent to 10(-4) genomes, by elec…

Maternal serum biochemical markers in pregnancies with fetal parvovirus B19 infection.

Hydrops fetalis and fetal death caused by fetal parvovirus B19 infection have been reported to be associated with elevated maternal serum alpha-fetoprotein (AFP), based on a total of six cases. It has been suggested that the absence of AFP elevation may be reassuring. Maternal serum levels of the Down syndrome screening markers unconjugated oestriol and human chorionic gonadotropin in cases of fetal parvovirus infection have not been previously reported. We report four cases of hydrops fetalis and fetal death caused by fetal parvovirus infection, each with unremarkable second-trimester levels …

Acute parvovirus B19 infection mimicking myelodysplastic syndrome of the bone marrow.

A 36-year-old, previously healthy woman was referred to our institution with pancytopenia and splenomegaly for suspected acute leukemia. Bone marrow aspiration showed marked dysplastic changes, excess of blasts, and only spurious red blood cell precursors. Action was taken to prepare allogeneic bone marrow transplantation from an HLA identical sibling for myelodysplastic syndrome. Repeat cytological examination of the bone marrow revealed striking hyperplasia of the red cell line with presence of abnormal giant proerythroblasts. Acute parvovirus B19 infection was suspected and confirmed by det…

Successful intrauterine treatment of fetal hydrops caused by parvovirus B19 infection.

The infection with parvovirus B19 during pregnancy causes in 1/3 of the cases an aplastic crisis in the fetus and consecutively generalized fetal hydrops as a result of severe anemia. Some cases of fetal hydrops and intrauterine death are reported. In our two cases, the fetal therapy by intrauterine intravasal transfusion was successful and the children developed normal. The techniques and indications for fetal blood sampling and the method of intrauterine intravasal transfusion are explained.

[Association of non-immunologically-induced hydrops fetalis with Parvovirus B19 infection].

Five weeks after the onset of clinical symptoms of erythema infectiosum, a 24 year old pregnant woman, gravida I, para 0, suffered a hydrops fetalis which resulted in fetal loss. The maternal infection with the human parvovirus B19 could be confirmed serologically with ELISA and immunoblot by detecting specific IgM antibodies against B19. In an amniotic sample of the fetus Parvovirus B19 DNA could be detected by hybridisation. Fetal loss was obviously due to a parvovirus B19 infection of the mother and the fetus.

A human parvovirus-like virus inhibits haematopoietic colony formation in vitro.

Viruses have been shown to cause bone marrow aplasia in animals and have been implicated in bone marrow failure in man; however, until recently, a specific link between human viral infection and bone marrow failure has not been proven. In 1975 Cossart and colleagues found a serum parvovirus-like virus (SPLV, sometimes referred to as B19) in human serum. Antibody to this virus is present in the sera of 30-45% of healthy adults (Y. E. Cossart, P. P. Mortimer, unpublished observations). However, evidence for a direct link came from work by Pattison et al. who found five children with transient ap…

Insights into parvovirus B19 clinical and virological dynamics among blood donors.

Coinciding with the exceptional European resurgence of human parvovirus B19 (B19V), between 2023 and 2024, twelve B19V DNA-positive blood donations out of 27.737 were detected by the plasma fractionating industry via nucleic acid testing (NAT) and notified to the blood collection centers of Trieste and Gorizia (Italy). In order to prevent potential B19V transmission, the implicated donors were recalled for clinical and virological follow-up for one year. None of the subjects had reported symptoms at the time of donation, though six subjects subsequently recalled either mild, transient symptoms…

Parvovirus B19 Intrauterine Infection and Cerebral Cortical Dysplasia: A Case Report.

BACKGROUND: Intrauterine parvovirus B19 infection usually causes fetal anemia, hydrops, and stillbirth. Less commonly, central nervous system injuries have also been described. CASE PRESENTATION: We report an atypical case of severe brain injury with diffuse malformation of cortical development and severely immature gyration in a preterm infant with profound fetal anemia and hydrops due to congenital parvovirus B19 infection. The fetus was treated with multiple intrauterine transfusions. Postnatal magnetic resonance imaging of the brain showed extensive encephalomalacia with severe volume los…

Oral glutamine supplementation in dogs infected with canine parvovirus type 2.

This study aimed to assess the impact of oral glutamine supplementation in dogs infected with Canine Parvovirus Type 2 (CPV-2). The dogs were randomly divided into two groups: Group 1 (n = 10) received glutamine supplementation (500 mg/kg/day), and ad libitum feeding; Group 2 (control) (n = 10) received a placebo and ad libitum feeding. Group 1 dogs were significantly younger (mean 3.5 ± 0.5 months) than those in Group 2 (mean 6.3 ± 3.0 months; p = 0.036). Data from the two groups were compared at the hospital admission and discharge. No significant differences were observed between groups in …

Intra-Host Evolution During Relapsing Parvovirus B19 Infection in Immunocompromised Patients.

BACKGROUND: Parvovirus B19 (B19V) can cause severe relapsing episodes of pure red cell aplasia in immunocompromised individuals, which are commonly treated with intravenous immunoglobulins (IVIGs). Few data are available on B19V intra-host evolution and the role of humoral immune selection. Here, we report the dynamics of genomic mutations and subsequent protein changes during relapsing infection. METHODS: Longitudinal plasma samples from immunocompromised patients with relapsing B19V infection in the period 2011-2019 were analyzed using whole-genome sequencing to evaluate intra-host evolutio…

Human parvovirus B19 vertical infection and hydrops fetalis. A case report.

Non-immune hydrops fetalis represents a diagnostic challenge in high-risk pregnant women. Vertical infection with human parvovirus B19 (B19V) is a possible cause. National guidelines propose maternal serologic screening (IgG/IgM), which may be insufficient in some situations. We report a case of vertical B19V infection with difficulties in prenatal diagnosis. Preterm newborn, normal weight (2950 g), born to a 30-year-old mother with anemia and hydrops fetalis (week 17). Cardiac, chromosomal, isoimmunization-Rh, and usual infectious causes (TORCH) were ruled out. Maternal serology for B19V show…
