Melatonin suppresses ILC2-driven airway hyperreactivity via glutathione-dependent metabolic reprogramming.

Allergic asthma is characterized by type 2 inflammation and overnight worsening of symptoms, yet dynamic fluxes in cellular metabolic profiles driving time-of-day variation remain poorly defined. Group 2 innate lymphoid cells (ILC2s) are central mediators of airway hyperreactivity. We identify melatonin as a previously unrecognized regulator of ILC2 metabolism and function. In murine models of allergic airway inflammation, melatonin reduced eosinophilia, type 2 cytokine production, and airway hyperreactivity without altering ILC2 abundance. Mechanistically, melatonin acted independently of can

Beyond energy storage: roles of glycogen metabolism in health and disease.

Beyond storing and supplying energy in the liver and muscles, glycogen also plays critical roles in cell differentiation, signaling, redox regulation, and stemness under various physiological and pathophysiological conditions. Such versatile functions have been revealed by various forms of glycogen storage diseases. Here, we outline the source of carbon flux in glycogen metabolism and discuss how glycogen metabolism guides CD8+ T-cell memory formation and maintenance. Likewise, we review how this affects macrophage polarization and inflammatory responses. Furthermore, we dissect how glycogen m

Catalase and glutathione peroxidase are equally active in detoxification of hydrogen peroxide in human erythrocytes.

Genetic deficiencies of glucose-6-phosphate dehydrogenase (G6PD) and NADPH predispose affected erythrocytes to destruction from peroxides. Conversely, genetic deficiencies of catalase do not predispose affected erythrocytes to peroxide-induced destruction. These observations have served to strengthen the assumption that the NADPH/glutathione/glutathione peroxidase pathway is the principal means for disposal of H2O2 in human erythrocytes. Recently, however, mammalian catalase was found to have tightly bound NADPH and to require NADPH for the prevention and reversal of inactivation by its toxic

Bridging sugar metabolism and signaling with hormones in shoot branching regulation.

Shoot branching, a major determinant of plant architecture and productivity, has long been attributed to the coordinated action of auxin, cytokinins (CKs), and strigolactones (SLs). Recent work shows that sugars act not only as metabolic resources but also as early developmental signals that regulate axillary bud outgrowth, the initial step in shoot branching. Sugar availability can rapidly trigger bud outgrowth, often preceding major hormonal changes. Mechanistically, sugars antagonize auxin and SL signaling while acting synergistically with CKs. These effects involve sugar signaling pathways

Engineering energy-efficient Saccharomyces cerevisiae for methanol and CO2 assimilation.

Methanol is a promising one-carbon (C1) feedstock for microbial bioconversion; however, engineered Saccharomyces cerevisiae often faces energetic constrains during its assimilation. Here, we develop SC-AOX25, an energy-efficient methylotrophic S. cerevisiae, through engineering of heterologous methanol-formaldehyde-formate (MFF) oxidation pathways coupled with adaptive laboratory evolution. SC-AOX25 efficiently generates adenosine triphosphate (ATP) and nicotinamide adenine dinucleotide (NADH) during methanol metabolism while co-assimilating methanol-derived intermediates (formaldehyde, format

Black Sea picocyanobacteria exploit ancestral traits for energy and chlorophyll production in dark anoxia.

Picocyanobacteria, typically associated with oxygenated waters, also thrive in oxygen minimum zones (OMZs) and anoxic environments. The extent to which retention of diverse ancestral anaerobic traits facilitate their metabolic flexibility with regards to transitioning between oxic and anoxic habitats is poorly understood. Here, using Black Sea Cyanobium (BSA11S), we performed 13C-glucose and 15N-ammonium incubations coupled with nanoscale ion mass spectrometry (NanoSIMS) and transcriptomics to characterize the growth mode and gene expression during transition from light-to-dark and oxic-to-ano

GATA1-deficient human pluripotent stem cells generate neutrophils with improved antifungal immunity that is mediated by the integrin CD18.

Neutrophils are critical for host defense against fungi. However, the short life span and lack of genetic tractability of primary human neutrophils has limited in vitro analysis of neutrophil-fungal interactions. Human induced pluripotent stem cell (iPSC)-derived neutrophils (iNeutrophils) provide a genetically tractable system to study host defense responses of human neutrophils. Here, we show that deletion of the transcription factor GATA1 from human iPSCs results in iNeutrophils with improved antifungal activity against Aspergillus fumigatus. GATA1-knockout (KO) iNeutrophils have increased

Hybrid Methylotrophic Pathway in Serratia marcescens for Sustainable Terpenoid Biosynthesis.

Terpenoids are valuable chemicals used across industries. Methanol, a nonsugar-based feedstock, offers an eco-friendly approach to terpenoid production. In this study, a hybrid methylotrophic pathway was engineered in Serratia marcescens (S. marcescens) HBQA7, which is tolerant to both methanol and terpenoid compounds. The pathway utilizes bacterial methanol dehydrogenase (Mdh) and yeast dihydroxyacetone synthase (Das) to produce monoterpenes and sesquiterpenes from methanol and xylose. 13C labeling experiments identified the optimal enzyme pair: Mdh from Acinetobacter gerneri and Das from Pic

Unveiling the mechanisms of metolachlor biodegradation and physiological adaptations in Penicillium oxalicum MetF1.

Environmental risk posed by metolachlor to farmland has garnered significant concern. AlthoughPenicillium oxalicumMetF1 exhibits metolachlor degradation potential, underlying biochemical mechanisms remain unclear. To address this, a comprehensive investigation integrating physicochemical properties, phenotypic characterization, transcriptomic profiling, and metabolomic analysis was conducted. Results demonstrate that MetF1 significantly accelerated metolachlor degradation during the logarithmic growth phase, with only trace metabolites detected. Scanning electron microscopy coupled with energy

Bacillus velezensis GV1 polysaccharides enhance immune function in cyclophosphamide-induced immunosuppressed mice via gut microbiota modulation and metabolic pathway regulation.

In this study, we investigated the immunomodulatory effects of Bacillus velezensis GV1 polysaccharides (BPS) on gut microbial regulation in a cyclophosphamide (CTX)-induced immunosuppressed mouse model. BPS treatment significantly restored immune function, as evidenced by improvements in spleen and thymus indices and increased cytokine expression levels, including IL-6, TNF-α, IFN-γ, and IL-2. Additionally, BPS administration effectively alleviated the CTX-induced histopathological damage to the colon, enhanced tissue repair, and maintained epithelial integrity. The beneficial effects of BPS w

Function and therapeutic potential of transketolase enzymes in parasitic protists.

Transketolase (TKT), the rate-limiting enzyme in the non-oxidative branch of the pentose phosphate pathway (PPP), catalyzes the reversible transfer of glycolaldehyde moieties between sugar phosphates. Here, we present wide-ranging methods to study TKT function in a clinically-relevant and globally-prevalent model intracellular pathogen, Toxoplasma gondii. The experimental framework utilizes heterologous expression of recombinant TgTKT proteins in a bacterial system, anti-TgTKT antibody production, CRISPR/Cas9-assisted genome editing to generate TgTKT-null mutants, and in-depth phenotyping, inc

Integrated approach for cellulosic ethanol and succinic acid production: Gamma valerolactone-based pretreatment and co-fermentation of peanut shells.

The inability to utilize pentose poses as a significant limitation to the production of cellulosic ethanol. To attain efficient raw material conversion and mitigate carbon dioxide emissions during cellulosic ethanol synthesis, a integrated approach focused on the co-processing of ethanol and succinic acid (SA) from peanut shells was proposed. The results demonstrated that the GVL system, containing 30 % water and catalyzed by dilute sulfuric acid, exhibited remarkable efficiency in pretreatment, boosting glucose yield sixfold relative to the untreated raw material. Under optimal conditions of

Enhanced Biosynthesis of d-Allulose from a d-Xylose-Methanol Mixture and Its Self-Inductive Detoxification by Using Antisense RNAs in Escherichia coli.

d-Allulose, a C-3 epimer of d-fructose, has great market potential in food, healthcare, and medicine due to its excellent biochemical and physiological properties. Microbial fermentation for d-allulose production is being developed, which contributes to cost savings and environmental protection. A novel metabolic pathway for the biosynthesis of d-allulose from a d-xylose-methanol mixture has shown potential for industrial application. In this study, an artificial antisense RNA (asRNA) was introduced into engineered Escherichia coli to diminish the flow of pentose phosphate (PP) pathway, while

The regulon of Brucella abortus two-component system BvrR/BvrS reveals the coordination of metabolic pathways required for intracellular life.

Brucella abortus is a facultative intracellular pathogen causing a severe zoonotic disease worldwide. The two-component regulatory system (TCS) BvrR/BvrS of B. abortus is conserved in members of the Alphaproteobacteria class. It is related to the expression of genes required for host interaction and intracellular survival. Here we report that bvrR and bvrS are part of an operon composed of 16 genes encoding functions related to nitrogen metabolism, DNA repair and recombination, cell cycle arrest, and stress response. Synteny of this genomic region within close Alphaproteobacteria members sugge

β-Adrenoceptor regulation of metabolism in U937 derived macrophages.

Macrophages have important roles in the immune system including clearing pathogens and wound healing. Metabolic phenotypes in macrophages have been associated with functional phenotypes, where pro-inflammatory macrophages have an increased rate of glycolysis and anti-inflammatory macrophages primarily use oxidative phosphorylation. β-adrenoceptor (βAR) signalling in macrophages has been implicated in disease states such as cancer, atherosclerosis and rheumatoid arthritis. The impact of βAR signalling on macrophage metabolism has not been defined. Using metabolomics and proteomics, we describe

Reactive Oxygen Species in Autoimmune Cells: Function, Differentiation, and Metabolism.

Accumulated reactive oxygen species (ROS) directly contribute to biomacromolecule damage and influence various inflammatory responses. Reactive oxygen species act as mediator between innate and adaptive immune cells, thereby influencing the antigen-presenting process that results in T cell activation. Evidence from patients with chronic granulomatous disease and mouse models support the function of ROS in preventing abnormal autoimmunity; for example, by supporting maintenance of macrophage efferocytosis and T helper 1/T helper 2 and T helper 17/ regulatory T cell balance. The failure of many

Stable Isotope-Resolved Metabolomics Studies on Corticosteroid-Induced PC12 Cells: A Strategy for Evaluating Glucose Catabolism in an in Vitro Model of Depression.

Depression is a common psychopathological state or mood disorder syndrome. The serious risks to human life and the inadequacy of the existing antidepressant drugs have driven us to understand the pathogenesis of depression from a new perspective. Our research group has found disturbances in glucose catabolism in both depression and nephrotic syndrome. What are the specific metabolic pathways and specificities of glucose catabolism disorders caused by depression? To address the above scientific questions, we creatively combined traditional metabolomics technology with stable isotope-resolved me

Hepatic Nfe2l2 Is Not an Essential Mediator of the Metabolic Phenotype Produced by Dietary Methionine Restriction.

The principal sensing of dietary methionine restriction (MR) occurs in the liver, where it activates multiple transcriptional programs that mediate various biological components of the response. Hepatic Fgf21 is a key target and essential endocrine mediator of the metabolic phenotype produced by dietary MR. The transcription factor, Nfe2l2, is also activated by MR and functions in tandem with hepatic Atf4 to transactivate multiple, antioxidative components of the integrated stress response. However, it is unclear whether the transcriptional responses linked to Nfe2l2 activation by dietary MR a

Metabolic conditioning of CD8+ effector T cells for adoptive cell therapy.

CD8+ effector T (TE) cell proliferation and cytokine production depends on enhanced glucose metabolism. However, circulating T cells continuously adapt to glucose fluctuations caused by diet and inter-organ metabolite exchange. Here we show that transient glucose restriction (TGR) in activated CD8+ TE cells metabolically primes effector functions and enhances tumour clearance in mice. Tumour-specific TGR CD8+ TE cells co-cultured with tumour spheroids in replete conditions display enhanced effector molecule expression, and adoptive transfer of these cells in a murine lymphoma model leads to gr
