Transcriptome and metabolome analyses reveal that GA3ox regulates the dwarf trait in mango (Mangifera indica L.).

BACKGROUND: Mango is a tropical fruit with high economic value. The selection of suitable dwarf mango varieties is an important aspect of mango breeding. However, the mechanisms that regulate mango dwarfing remain unclear. RESULTS: In this study, we compared the transcriptomes and metabolomes of mango varieties Guiqi (a dwarfed variety) and Jinhuang (an arborized variety). A total of 4,954 differentially expressed genes and 317 differentially abundant metabolites were identified between the two varieties, revealing the molecular mechanism of the gibberellin 3β-hydroxylase gene GA3ox in regula

Low temperature modulates spike development and yield via ABA pathway at post-reproductive transition stage in wheat.

Wheat (Triticum aestivum L.) yield is very sensitive to environmental temperature. Here, we integrated field sowing experiments and controlled-temperature treatments to explore how low temperature affects spike differentiation and yield formation in wheat. Early sowing allowed the wheat to undergo the reproductive transition before the winter, supporting prolonged spikelet differentiation under low-temperature conditions. By contrast, late sowing markedly shortened the reproductive growth period, reducing effective tiller number per plant and grain number per spike. Exposure to low temperature

Lysine catabolism, amino acid transport, and systemic acquired resistance: what is the link?

Lysine is an essential amino acid for human nutrition, which is generally low in cereal diets. Its biosynthesis via the aspartate-pathway and catabolism is controlled by complex feedback mechanisms. Recently, aspartate-derived amino acids were found to be elevated during pathogen infection in Arabidopsis and a lysine catabolite, pipecolic acid, was identified as critical regulator of systemic acquired resistance. Pipecolic acid is mobile in plants, functions as an intensifier of defense responses and mediates systemic acquired resistance establishment via signal amplification. The altered path

TOPlus: An algorithm integrating prior genetic and environmental information for multi-trait synergic selection in maize hybrid breeding.

In maize hybrid breeding, synergic multi-trait selection of elite hybrids in specific target environments remains a major challenge. Enviromic data and functional gene knowledge are rapidly increasing; however, they have not been effectively integrated into crop breeding decisions. Here, we present TOPlus, a multi-trait hybrid prioritization framework comprising predictive and selective modules. The predictive module uses environmental information to improve phenotype prediction for untested genotypes and environments, whereas the selective module incorporates functional gene priors and multip

Plant chimeras: The good, the bad, and the 'Bizzaria'.

Chimeras - organisms that are composed of cells of more than one genotype - captured the human imagination long before they were formally described and used in the laboratory. These organisms owe their namesake to a fire-breathing monster from Greek mythology that has the head of a lion, the body of a goat, and the tail of a serpent. The first description of a non-fictional chimera dates back to the middle of the seventeenth century when the Florentine gardener Pietro Nati discovered an adventitious shoot growing from the graft junction between sour orange (Citrus aurantium) and citron (Citrus

Automatic Scoring of Rhizoctonia Crown and Root Rot Affected Sugar Beet Fields from Orthorectified UAV Images Using Machine Learning.

Rhizoctonia crown and root rot (RCRR), caused by Rhizoctonia solani, can cause severe yield and quality losses in sugar beet. The most common strategy to control the disease is the development of resistant varieties. In the breeding process, field experiments with artificial inoculation are carried out to evaluate the performance of genotypes and varieties. The phenotyping process in breeding trials requires constant monitoring and scoring by skilled experts. This work is time demanding and shows bias and heterogeneity according to the experience and capacity of each individual person. Optical

Tobamoviruses: Advances in Molecular Biology, Host Interactions and Integrated Disease Management.

Tobamoviruses (viruses in the genus Tobamovirus, family Virgaviridae) lead to major yield losses in economically important crops around the world. In this review, we go beyond the canonical gene expression framework by integrating recent discoveries of reverse open reading frames (rORFs) on the negative-strand RNA. These rORFs have only been experimentally validated in cucumber green mottle mosaic virus (CGMMV), with predicted sequence-conserved homologs across a subset of the genus, including TMV, ToBRFV, and PMMoV. However, they are not universally present in all tobamoviruses. We systematic

Analysis of the Mechanisms of Rice oszfp30 Mutant in Improving Forage-Quality-Related Traits Based on Transcriptome and Metabolome Analyses.

BACKGROUND/OBJECTIVES: Using straw as feed is crucial for addressing resource scarcity and waste-based environmental pollution and alleviating the contradiction between forage production and food security. However, rice straw has disadvantages such as a high lignin, cellulose, and hemicellulose content, as well as a dense cell wall structure, which greatly limit its utilization as feed. Improving the forage quality of rice straw through genetic breeding is an important means of enhancing its utilization rate. METHODS: To explore the molecular basis underlying the improved nutritional and cell

A robust biotechnology induces artificial genomic duplication via transient RNAi-mediated suppression of OSD1 in rice.

Ploidy manipulation is a crucial strategy for generating germplasm in crop breeding. However, artificial genomic duplication, often induced by colchicine treatment, is associated with toxicity and unpredictability. Although mutations in OSD1 have shown promise for inducing genomic duplication, the instability of ploidy across generations limits their practical application. In this study, we developed a Plant Polyploidization via Gene Interference (PPGI) system that utilizes transient RNAi-mediated suppression of OSD1 to efficiently induce artificial genomic duplication, demonstrating obvious p

Underpinning regions associated with nitrogen use efficiency in Brassica juncea L. Czern: using associative transcriptomics approach.

BACKGROUND: Breeding crops for enhanced understanding of apparent nitrogen use efficiency (NUE) requires understanding of phenotypic variations and underlying genetics. Improving NUE in Indian mustard is considered important to reduce nitrogen (N) fertilizer use without sacrificing crop yield. In the present study, 260 genotypes of Brassica juncea were field evaluated for seed yield and NUE-related traits at three N levels: N0 (no added nitrogen), N50 (50 kg/ha nitrogen), and N100 (100 kg/ha nitrogen) for two years. RESULTS: N response in plants varied depending on the dose of nitrogen applie

Improving the genetic potential of okra (Abelmoschus esculentus L.) germplasm to tolerate salinity stress.

Okra (Abelmoschus esculentus L.) is the most consumed vegetable worldwide with the potential for diverse ecological adaptation. However, increasing salinization and changing climatic conditions are posing serious threats to the growth, yield, and quality of okra. Therefore, to mitigate increasing soil salinization and ensure sustainable okra production under rapidly changing climatic conditions, evaluation of new okra germplasm to develop salt tolerant cultivars is direly needed. The present study was designed to evaluate the genetic resources of okra genotypes for salt tolerance at growth and

Enhanced Soybean Immunity to the Soybean Mosaic Virus Through RNA Interference Targeting the CP Gene.

The soybean mosaic virus (SMV), a significant viral pathogen impacting soybean cultivation, leads to substantial yield losses and diminishes seed quality. In a prior study, we developed a targeted silencing vector using RNA interference (RNAi) technology targeting the CP gene, which codes for the viral coat proteins in the SMV genome. This vector was delivered into soybean plants through Agrobacterium-mediated transformation. In our current research, we utilized ongoing molecular characterization and resistance screening to identify four genetically pure lines that display moderate to high res

SPINet: structure-enhanced point cloud instance segmentation network for multi-plant rice phenotyping.

Accurate 3D phenotyping of rice in multi-plant scenarios is important for evaluating yield-related traits and supporting high-throughput breeding. In particular, grain-level instance segmentation from point clouds provides a direct basis for quantifying panicle structure and grain number. However, this task remains challenging because densely grown rice plants often exhibit severe inter-plant occlusion, organ adhesion, and highly similar local structures, making it difficult to simultaneously capture global plant organization and fine-grained grain geometry. To address these challenges, we pro

Sunlight-sensitive carbon dots for plant immunity priming and pathogen defence.

Global food production faces persistent threats from environmental challenges and pathogenic attacks, leading to significant yield losses. Conventional strategies to combat pathogens, such as fungicides and disease-resistant breeding, are limited by environmental contamination and emergence of pathogen resistance. Herein, we engineered sunlight-sensitive and biodegradable carbon dots (CDs) capable of generating reactive oxygen species (ROS), offering a novel and sustainable approach for plant protection. Our study demonstrates that CDs function as dual-purpose materials: priming plant immune r

A multi-race blast resistance locus identified and validated in US elite rice via association and biparental mapping.

Rice blast, caused by Magnaporthe oryzae (syn. Pyricularia oryzae), remains a major disease in US rice (Oryza sativa L.) production, highlighting the need for additional resistance loci and continued development of blast-resistant rice varieties. We evaluated 258 US elite long-grain breeding lines for resistance to four M. oryzae strains representing blast races IB-1, IB-17, IB-49, and IE1K and conducted a genome-wide association study using 543,136 single-nucleotide polymorphisms. Thirteen significant loci were detected, including three multi-race resistance loci. The strongest locus, qBlast4

Induction of dwarf and early flowering phenotypes in Tricyrtis sp. by ectopic expression of LEAFY from Arabidopsis thaliana.

LEAFY (LFY), which encodes a plant-specific transcription factor, plays an important role in the transition from vegetative to reproductive development. Ectopic expression of LFY has been reported to induce dwarfism and early flowering in some model plants. In order to examine the possibility of using LFY for molecular breeding of ornamental plants, we produced and characterized transgenic plants ectopically expressing LFY from Arabidopsis thaliana (AtLFY) in the liliaceous ornamental plant Tricyrtis sp. Nine independent transgenic plants have been obtained, and all of them exhibited dwarf phe

Ghd7‑triggered transcriptional regulation for synergistically improving rice eating quality and yield.

Improving both eating quality and yield remains a major challenge in rice breeding. Here, we identify Ghd7 as the causal gene underlying the grain amylose content QTL qAC7 and reveal Ghd7-triggered transcriptional regulation that balances starch and protein to coordinate yield and quality. Ghd7 transcriptionally activates PCF3 and exhibits dominant epistasis over it. A natural TA/T variant in PCF3 determines its DNA-binding affinity: the PCF3‑T allele strongly represses OsNF-YC4, increasing starch and reducing protein in grain, whereas PCF3‑TA fails to exert this repressive effect. Bulked segregant analysis reveals candidate genes responsible for dwarf formation in woody oilseed crop castor bean.

Plant dwarfism is a desirable agronomic trait in non-timber trees, but little is known about the physiological and molecular mechanism underlying dwarfism in woody plants. Castor bean (Ricinus communis) is a typical woody oilseed crop. We performed cytological observations within xylem, phloem and cambia tissues, revealing that divergent cell growth in all tissues might play a role in the dwarf phenotype in cultivated castor bean. Based on bulked segregant analyses for a F2 population generated from the crossing of a tall and a dwarf accession, we identified two QTLs associated with plant heig
