Echinacea is commonly used as a dietary supplement owing to its putative anti-inflammatory, antiviral, and antibacterial activities. Unfortunately, determining risk posed to human health is challenging. A novel combination of chemical and biological characterization was employed for selection of a representative extract sample used for in vivo investigation of the immunomodulatory properties of Echinacea (E.) purpurea. In vitro cytokine profiling showed the extract could stimulate pro-inflammatory cytokine production in the absence of detectable endotoxin. An in-depth in vivo evaluation of the

Catalases (CATs) play important roles in plant growth, development and defense responses. Previous studies have shown that CATs exhibit different or even opposite effects on plant immunity in different plant-pathogen interactions, but little is known about the mechanisms. In this study, Nicotiana tabacum plants with overexpression or knockout of CAT genes, tobacco mosaic virus (TMV) and cucumber mosaic virus (CMV) were employed to investigate the role of CAT in compatible plant-virus interactions. The results showed that there were dynamic changes in the effect of CAT on N. tabacum defense res…

The plant cell wall is a dynamic matrix integrating mechanical, biochemical and immune signals during plant-microbe interactions. Although fungal exopolysaccharides (EPS) modulate these interactions, their structures and roles in cell wall remodelling remain unclear. Here we characterize the EPS secreted by Hymenoscyphus fraxineus, the causal agent of ash dieback, and demonstrate its biological activity in Fraxinus excelsior and Arabidopsis thaliana. Biochemical analyses revealed that the EPS is enriched in mixed linkage β-glucans (MLG). EPS exposure triggered substantial remodelling of cell w

Microneedle patches (MNPs), initially developed for minimally invasive drug and vaccine delivery, combine engineering precision with biological function to achieve controlled, painless transport. Beyond medicine, MNPs are increasingly viewed as sustainable interfaces for agriculture and food systems under pressure to increase productivity and ensure safety. This review traces MNP evolution from brittle silicon and metals to biodegradable polymers, dissolvable hydrogels, and stimuli-responsive or bioelectronic platforms. Fabrication techniques utilizing MEMS, micromolding, and 3D printing are e

Microbial pathogens pose a serious challenge to food security under climate change. Microbiomes can filter out incoming pathogens. However, the specific microbial taxa and mechanisms that indicate the response of the microbiome and plant health to the invasion of pathogens remain largely unknown. Here, we combine multiple field surveys and experiments with DNA and RNA sequencing using tobacco (Nicotiana tabacum) and the bacterial wilt pathogen Ralstonia solanacearum as the model system, and f ind that Pseudomonadaceae constitute an important root-associated component against pathogen invasions

The exponential spread of Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) emphasizes the immediate need for effective antiviral drugs and vaccines that could control and prevent the spread of this pandemic. Several new and repurposed drugs are being tested for their effectiveness in the treatment regime, and the development of vaccines is underway. The availability of genome sequence information of the virus and the identification of potential targets to neutralize and eradicate the infection have enabled the search for novel as well as existing molecules to perform the desired fu

Common scab, a globally important potato disease, is caused by infection of tubers with pathogenic Streptomyces spp. Previously, disease-resistant potato somaclones were obtained through cell selections against the pathogen's toxin, known to be essential for disease. Further testing revealed that these clones had broad-spectrum resistance to diverse tuber-invading pathogens, and that resistance was restricted to tuber tissues. The mechanism of enhanced disease resistance was not known. Tuber periderm tissues from disease-resistant clones and their susceptible parent were examined histologicall

Some plants respond to infection by pathogens with both localized and systemic resistance responses. These prevent the spread of the disease-causing organism and reduce the severity of a subsequent infection. Recent evidence suggests that systemic increases in the host's salicylic acid levels act as a signal for the activation of at least some of these induced defenses.

SlXTH3 promotes tomato lateral root development and cell wall remodeling, which facilitate early root colonization by Ralstonia solanacearum and increase susceptibility to bacterial wilt. Ralstonia solanacearum is a highly destructive soil-borne bacterial pathogen whose infection typically initiates in the root system and is accompanied by remodeling of host root development and defense responses. The results of this study showed that, during the early infection of tomato roots, R. solanacearum preferentially colonized lateral root emergence sites and promoted lateral root development, thereby

Plant biostimulants represent a promising option to improve agricultural production and stress resistance while reducing the use of fertilizers and pesticides. Despite various evidence demonstrating the beneficial role of biostimulants in preventing the negative effects of abiotic stress on plants, the ability of biostimulants to bolster defense mechanisms has been brought to light only recently. In this work, the impact of a biostimulant based on Ecklonia maxima and yeast extracts (S/Y) on the response of tomato infected with Pseudomonas syringae pv. tomato DC3000 (Pst DC3000) was assessed. Distinct amino acid metabolic pathways constitute integral parts of the plant immune system. We have recently identified pipecolic acid (Pip), a lysine-derived non-protein amino acid, as a critical regulator of systemic acquired resistance (SAR) and basal immunity to bacterial infection in Arabidopsis thaliana. In Arabidopsis, Pip acts as an endogenous mediator of defense amplification and priming. For instance, Pip conditions plants for effective biosynthesis of the phenolic defense signal salicylic acid (SA), accumulation of the phytoalexin camalexin, and expression of defense-related genes.

• It has long been believed that plant species from the tropics have higher levels of traits associated with resistance to herbivores than do species from higher latitudes. A meta-analysis recently showed that the published literature does not support this theory. However, the idea has never been tested using data gathered with consistent methods from a wide range of latitudes. • We quantified the relationship between latitude and a broad range of chemical and physical traits across 301 species from 75 sites world-wide. • Six putative resistance traits, including tannins, the concentration of

Pyrroloquinoline quinone (PQQ) is a redox cofactor derived from prokaryotes that participates in various biological processes involving dehydrogenase enzymes. Previous field trials identified a PQQ-producing endophyte, Burkholderia seminalis 869T2, which enhances banana growth and reduces Fusarium wilt incidence from 24.5% to 3.4%. While more recent studies have confirmed its agricultural benefits across multiple plant species, the underlying molecular mechanisms remain unclear. Here, integrated omics and imaging mass spectrometry were employed to investigate the role of PQQ in planta. Our res

The magnesium chelatase subunit CHLI is well-characterized in chlorophyll biosynthesis, but its role in plant immunity remains unclear. In this study, we generated chimeric Fvchli mutant strawberries via CRISPR/Cas9 and obtained T1-T3 progeny with a wide range of editing efficiencies through self-pollination. Mutants with higher editing efficiency exhibited more leaf spots. Following inoculation with Xanthomonas fragariae, Fvchli mutants showed increased susceptibility, with both lesion area and disease incidence positively correlating with editing efficiency. We further demonstrated that stom…

The receptor-like kinase SOBIR1 (SUPPRESSOR OF BIR1) is a core component of immune receptor complexes mediating plant responses to extracellular immunogenic signals. While its role in pattern-triggered immunity is well defined in model species, its broader physiological functions remain largely unexplored. Here, we generated CRISPR-Cas9-derived double knockouts of two homeologous SOBIR1 genes in allotetraploid Nicotiana tabacum. The resulting Nt sobir1 double mutants failed to trigger elicitor-induced cell death in response to elicitins from Phytophthora and Pythium spp. but retained an attenu

BACKGROUND: Engineering root microbiomes holds great promise to enhance plant health. Enhanced plant resistance via breeding or genetic modification can promote recruiting beneficial microbes, but is challenging to achieve.
RESULTS: Here we showed that calcium (Ca) addition caused significant changes in tomato physiology, resulting in enhanced immunity towards the pathogen Ralstonia solanacearum along with increased levels of salicylic acid (SA), sugar content, and defense enzyme activities in roots. High Ca levels significantly altered the root microbiomes, enriching sixteen bacterial genera

Plants have evolved strong innate immunity mechanisms, but successful pathogens evade or suppress plant immunity via effectors delivered into the plant cell. Hyaloperonospora arabidopsidis (Hpa) causes downy mildew on Arabidopsis thaliana, and a genome sequence is available for isolate Emoy2. Here, we exploit the availability of genome sequences for Hpa and Arabidopsis to measure gene-expression changes in both Hpa and Arabidopsis simultaneously during infection. Using a high-throughput cDNA tag sequencing method, we reveal expression patterns of Hpa predicted effectors and Arabidopsis genes i…

BACKGROUND: Gut microbiota has been widely recognised as playing a critical role in maintaining immune imbalance and the development of rheumatoid arthritis (RA). As key roles mediating interkingdom crosstalk among plants, microbiomes and mammals, plant-derived exosome-like nanoparticles (ELNs) could use lipids and microRNA components to precisely modulate gene expression of gut microbiota, showing potential as a dietary intervention for RA treatment.
OBJECTIVE: We aimed to investigate gut microbiota-immune interactions inducing immune dysregulation in RA and explore potential applications of
