The irreversible human platelet aggregation induced by threshold concentrations of platelet-activating factor (PAF) in citrated plasma was reversed by aspirin (100 mumol/L, with 10 minutes' preincubation). The aspirin-sensitive amplification was linked to thromboxane generation, although thromboxane could be successfully replaced by cyclic endoperoxides, as suggested by the lack of effect of dazoxiben, a selective thromboxane-synthase inhibitor. The inhibitory effect of aspirin could be overcome by using 10 times higher PAF concentrations or, even more effectively, by combining threshold conce

We have studied the pathways that lead to arrest and firm adhesion of rolling PMN on activated, surface-adherent platelets. Stable arrest and adhesion strengthening of PMN on thrombin-stimulated, surface-adherent platelets in flow required distinct Ca2+- and Mg2+-dependent regions of Mac-1 (alphaMbeta2), and involved interactions of Mac-1 with fibrinogen, which was bound to platelets via alphaIIbbeta3. Mac-1 also bound to other unidentified ligands on platelets, which were not intracellular adhesion molecule-2 (ICAM-2), heparin, or heparan-sulfate proteoglycans. This was shown by inhibition wi

Platelets play a crucial role in atherosclerosis and thrombus formation, and their activation is regulated by reactive oxygen species (ROS). … tential novel target for antithrombotic interventions. Endogenous ROS participate in the cascade of platelet activation and thrombus stabilization by modifying key signaling proteins and regulating ion channel function. Phospholipase C (PLC)-evoked rising intracellular Ca2+-levels are a prerequisite for platelet activation with subsequent thrombosis. The protein-tyrosine kinase Janus kinase 3 (JAK3) regulates phosphatidylinositol 4,5-bisphosphate (PI(4,5)P2)-levels in platelets, thus significantly mediating activation-dependent platelet function and arterial thrombosis.

The concept of "rebalanced hemostasis" has improved our understanding of the coexistence of bleeding and thrombosis in cirrhosis. This balance, including changes of platelet function, is easily disrupted by inflammation, through bacterial gut translocation. The modulation of platelet activation through non-absorbable antibiotics remains unexplored. A cross-sectional analysis between patients with decompensated cirrhosis (Child-Pugh, CP, classes B and C) versus age-matched controls without cirrhosis and cardiometabolic diseases was performed to characterize the platelet phenotype and assessed s…

Background/Objectives: The recommendations for antithrombotic therapy after transcatheter edge-to-edge mitral valve repair (TEER) are empirical, and the benefit of antiplatelet (APT) or anticoagulation therapy (ACT) remains undetermined. The study sought to investigate the degree and the timing of coagulation and platelet marker activation after TEER. Methods: This was a prospective study including 46 patients undergoing TEER. The markers of coagulation activation, namely prothrombin fragment 1 + 2 (F1 + 2) and thrombin-antithrombin III (TAT), and the markers of platelet activation, namely sol…

This letter comments on the recently published article in the World Journal of Gastroenterology, in which the authors demonstrated a strong link between lymphangiogenesis and the process of platelet adherence, aggregation, and activation by employing a rat model of liver cirrhosis caused by bile duct ligation (BDL). The authors applied both gain and loss of function approach by using platelet-rich plasma and vascular endothelial growth factor 3 receptor inhibitor MAZ-51 to activate and inhibit angiogenetic signaling in BDL rat model, respectively, to verify the crucial function of lymphangioge

Antibodies to β2-glycoprotein I (β2GPI) cause thrombosis in antiphospholipid syndrome; however, the role of β2GPI in coagulation in vivo is … thrombin-induced activation of Apoh-/- platelets. These studies demonstrate that β2GPI may promote platelet activation by enhancing the ability of PAR3 to present thrombin to PAR4.

Excessive platelet activation plays a pivotal role in the development of cardiovascular disease. Taxifolin is a natural dihydroflavonol compound with antioxidant properties. However, its mechanism of action on platelets and thrombosis remains unclear. Therefore, this study aimed to investigate the effects of taxifolin on platelet function and thrombus formation and identify its potential targets. The results revealed that taxifolin inhibited platelet aggregation induced by various agonists, such as ADP, collagen, thrombin, U46619, and convulxin. Moreover, it also limited platelet adhesion to c…

Coagulopathy and traumatic brain injury (TBI) are complexly intertwined. In isolated TBI, coagulopathy may contribute to hemorrhagic lesion development, progression, or recurrence, as it may lead to a particular pattern of coagulopathy called TBI-induced coagulopathy (TBI-IC). We performed a retrospective and descriptive evaluation of 63 patients admitted to the Emergency Clinical Hospital Bucharest with the diagnosis of moderate/severe brain injury. In addition to demographic data, all included patients had a complete paraclinical evaluation that included rotational thromboelastometric (ROTEM

BACKGROUND: Newborns are at high risk of sepsis. At present there is no definitive "rule in" blood test for sepsis at the point of clinical concern. A positive blood culture remains the gold standard test for neonatal sepsis, however laboratory markers that correlate prospectively with culture positive sepsis could aid clinicians in making decisions regarding administration of empiric antibiotic therapies.
METHODS: This multi-site, prospective observational study will take place in two neonatal intensive care units (National Maternity Hospital and Rotunda Hospital, Dublin). Neonates born at l

In order to improve the safety of COVID-19 vaccines, there is an urgent need to unravel the pathogenesis of vaccineinduced immune thrombotic thrombocytopenia (VITT), a severe complication of recombinant adenoviral vector vaccines used to prevent COVID-19, and likely due to anti-platelet factor 4 (PF4) IgG antibodies. In this study, we demonstrated that 1E12, a chimeric anti-PF4 antibody with a human Fc fragment, fully mimics the effects of human VITT antibodies, as it activates platelets to a similar level in the presence of platelet factor 4 (PF4). Incubated with neutrophils, platelets and PF

BACKGROUND: Impaired glucose tolerance (IGT) is associated with increased cardiovascular morbidity and mortality. Enhanced thromboxane (TX)-dependent platelet activation plays a pivotal role in atherothrombosis and characterizes type 2 diabetes mellitus (DM). Whether this also pertains to IGT is currently unknown. We investigated whether TXA2 -dependent platelet activation, as reflected by 11-dehydro-TXB2 (TXM) urinary excretion, is comparably abnormal in IGT as in DM, is persistent over long-term follow-up, changes as a function of metabolic disease progression, and is influenced by food inta…
