Association of Sodium Thiosulfate With Risk of Ototoxic Effects From Platinum-Based Chemotherapy: A Systematic Review and Meta-analysis.

IMPORTANCE: Platinum-induced ototoxic effects are a significant issue because platinum-based chemotherapy is one of the most commonly used therapeutic medications. Sodium thiosulfate (STS) is considered a potential otoprotectant for the prevention of platinum-induced ototoxic effects that functions by binding the platinum-based agent, but its administration raises concerns regarding the substantial attenuation of the antineoplastic outcome associated with platinum. OBJECTIVE: To evaluate the association between concurrent STS and reduced risk of ototoxic effects among patients undergoing plat

Hydroquinone colorimetric sensing based on platinum deposited on CdS nanorods as peroxidase mimics.

Pt deposited on CdS nanorods (Pt/CdS) have been prepared via the UV light photoreduction method. The Pt/CdS nanocomposites possess highly significant peroxidase-like activity with the assistance of the colorless substrate 3,3,5,5-tetramethylbenzidine (TMB). In the presence of peroxidase mimic Pt/CdS, TMB is quickly oxidized into a typical blue product (oxTMB, which has an obvious absorption at 652 nm) by H2O2 only in 3 min, which is easily detected visually. The catalytic activity of Pt/CdS originates from the accelerated electron transfer between the reactants. Combining the peroxidase-like a

Limited efficacy of platinum-based adjuvant treatment on the outcome of borderline ovarian tumors.

Adjuvant treatment of borderline ovarian tumors (BOT) remains highly debatable. This article evaluates the benefits of platinum-based adjuvant treatment in patients with BOT. The PubMed and Cochrane Library databases were systematically searched for articles using the terms ((Borderline) OR (low malignant potential) AND (ovarian)) AND ((tumor) OR (cancer)) AND ((follow-up) OR (survival) OR (treatment) OR (chemotherapy) OR (adjuvant treatment)). We identified 31 articles including 4965 patients. Together, 592 patients presented non-invasive-, 244 invasive- and 77 unspecified implants. Central p

Preparation of a DNA aptamer-Pt complex and its use in the colorimetric sensing of thrombin and anti-thrombin antibodies.

DNA aptamers carrying Pt nanoparticles were prepared by the reaction of DNA aptamers (without functionalization with biotin, thiol, or other reactive groups) with K 2[PtCl 4] in solution at 60-90 degrees C. The DNA-Pt complexes possessed peroxidase enzymatic activity while retaining the specific binding ability of the aptamers. The enzymatic reaction of these complexes obeyed Michaelis-Menten kinetics. K M for the DNA-Pt complex was found to be on the same order as K M for hemin and hemin-DNA complex but 1 or 2 orders of magnitude higher than that of horseradish peroxidase. The rate of the rea

In vivo measurement of platinum in the kidneys using X-ray fluorescence.

A noninvasive in vivo method has been developed and optimized for measuring platinum concentrations in the kidneys of patients receiving chemotherapy. The method is based on polarizing the X-ray beam from an orthovoltage radiotherapy treatment unit, the Pantak DXT-300, and using the beam to produce emission of the characteristic platinum X-rays from the kidney. The platinum is derived from platinum-based chemotherapy drugs, such as cisplatin and its analogues (carboplatin and iproplatin), used to treat cancer patients. The clinical motivation for measuring the platinum concentration in both th

Hearing loss related to ototoxicity in children with cancer.

Children undergoing treatment for cancer often receive agents that put them at risk for ototoxicity. Platinum-based chemotherapy, aminoglycoside antibiotics, loop diuretics, and radiotherapy are all capable of inducing inner ear damage, which may result in significant sensorineural hearing loss. Frequent audiological monitoring is necessary for the early detection of changes in hearing thresholds. Age-appropriate modification of audiological testing is essential to obtain accurate results and provide maximum comfort for pediatric patients. When hearing loss is identified promptly, consideratio

Mechanism of cisplatin nephrotoxicity.

cis-Diamminedichloroplatinum II (cisplatin) is widely used in cancer treatments. Renal dysfunction is the major toxic effect of this drug. Micropuncture studies suggest that cisplatin reduces single-nephron glomerular filtration rate (GFR) and causes a significant backleak of inulin across the renal tubule. Pathological alterations are localized to the S3 segment of the proximal tubule situated in the outer stripe of the outer medulla. Renal clearance studies in humans demonstrate that the free platinum clearance exceeds the GFR, which suggests that cisplatin or a platinum metabolite is active

Dual signal-enhanced immunochromatographic test strip based on Au@PtNPs: From sensitive detection of thiamethoxam to multiplex pesticide screening in vegetables.

Immunochromatographic test strip (ICTS) is a rapid analytical technique widely used in environmental and food detection owing to its merits of simple operation and short analysis time. Herein, three-dimensional nanoflower-structured gold‑platinum nanoparticles (Au@PtNPs) were synthesized via a seed-growth method. Compared with conventional gold nanoparticles (AuNPs), Au@PtNPs exhibited stronger signal intensity, excellent catalytic performance, and efficient antibody binding efficiency. Colorimetric Au@PtNPs-ICTS and catalytic colorimetric Au@PtNPs-ICTS were developed for the sensitive detecti

Engineering Pt-decorated g-C3N4 nanosheets with Fe3+ and chitosan for amplified photodynamic ROS production and hydrogen-mediated ATP depletion.

Graphitic carbon nitride (g-C3N4) has attracted considerable attention as a multifunctional platform for cancer therapy, including photodynamic therapy (PDT) and hydrogen (H2)-based therapeutic strategies, owing to its intrinsic photocatalytic activity. Nevertheless, its clinical translation is hindered by rapid photogenerated electron-hole recombination and tumor-associated defense mechanisms, such as elevated glutathione (GSH) levels and hypoxic microenvironments. In this work, we report the development of chitosan-modified g-C₃N₄ nanosheets (OCN/Pt-CTS) as a synergistic anticancer nanoplatf

Self-Adaptive Sn-Fe Electron Pair Centers in Pt/SnFe2O4 for Anti-Tumor Synergistic Therapy.

Despite the multiple advantages of high tissue penetration depth, selectivity, and noninvasiveness of photothermal-assisted antitumor synergistic therapy, developing photothermal agents with multifunctional characteristics, desirable tumor therapeutic effects, and biological stability remains a key challenge. Herein, an electronic structure-regulated strategy is proposed to activate self-adaptive Sn-Fe electron pair centers (Sn2+/Sn4+ and Fe2+/Fe3+) through loading the electron donor Pt (Pt/SFO-1, -2, -3, -4). Experimental and theoretical calculations indicate that abundant dynamic active cent

Colorimetric determination of Pb2+ ions based on surface leaching of Au@Pt nanoparticles as peroxidase mimic.

We report the first use of metallic nanozyme as colorimetric probe for Pb2+ determination. The method is based on the surface leaching of Au@PtNP nanozyme by Pb2+-S2O32- ions, accompanied by a decreased catalytic activity of the metallic nanozyme. To construct this colorimetric determination, the Pt deposition onto the AuNPs was carefully investigated and other experimental factors including kind of substrate and buffer were optimized. With increasing Pb2+ concentration, the catalytic activity of the Au@PtNPs decreased gradually. As a result, the blue color at 650 nm from the oxidation of 3,3'

Activation of T cells by dendritic cells exposed to a reference sensitizer: Towards a promising model to assess the allergenic potential of chemicals.

BACKGROUND: Low molecular weight chemicals constitute one of the major causes of occupational allergies. European legislation on chemicals recommends limiting the use of in vivo models for assessing the sensitizing potential of chemicals, and encourages the development of integrated alternative methods. An in vitro mouse model of bone marrow-derived dendritic cells (BMDCs) that showed good accuracy (75%) and sensitivity (69%) has previously been developed to assess the sensitizing potential of chemicals. OBJECTIVE: To assess the ability of BMDCs to activate T cells (TCs) in vitro. METHODS: B

VPA does not enhance platinum binding to DNA in cisplatin-resistant neuroblastoma cancer cells.

Neuroblastoma represents a malignancy of the sympathetic nervous system characteristic by biological heterogeneity. Thus, chemotherapy exhibits only low effectivity in curing high-risk forms. Previous studies revealed the cytotoxic potential of valproate on neuroblastoma cells. Nevertheless, these studies omitted effects of hypoxia, despite its undeniable tumorigenic role. In this study, we addressed the question whether valproate promotes binding of platinum-based anti-cancer drugs (cisplatin, carboplatin and oxaliplatin) to DNA and role of hypoxia, cellular antioxidant capacity and cisplatin

Molecular mechanisms for synergistic effect of proteasome inhibitors with platinum-based therapy in solid tumors.

The successful development of the proteasome inhibitor bortezomib as an anticancer drug has improved survival in patients with multiple myeloma. With the emergence of the newly US Food and Drug Administration-approved proteasome inhibitor carfilzomib, ongoing trials are investigating this compound and other proteasome inhibitors either alone or in combination with other chemotherapy drugs. However, in solid tumors, the efficacy of proteasome inhibitors has not lived up to expectations. Results regarding the potential clinical efficacy of bortezomib combined with other agents in the treatment o

Magnetic properties of Fe(x)Co(1-x) nanochains on Pt(1 1 1) surfaces.

The magnetic properties of FexCo1-x nanochains on Pt(1 1 1) were studied using the first-principles real-space linear muffin-tin orbital-atomic sphere approximation (RS-LMTO-ASA) method within the density functional theory. The relative amounts of Fe and Co atoms in a chosen nanochain were varied and several possible arrangements of the atomic species were taken into account. The results of the exchange interaction demonstrates ferromagnetic coupling for the nanowires. Our calculations of Fe and Co average magnetic moments reveal a large enhancement of both spin and orbital moments compared to

Guanidine platinum(II) complexes: synthesis, in vitro antitumor activity, and DNA interactions.

The novel guanidine compounds trans-[Pt(NH2Me)2{NH=C(NHMe)NR}2](Cl)2 (R=NEt2 [7], NC5H10 [8]) (trans-7,8) were synthesized by the nucleophilic addition of methylamine to dialkylcyanamide ligands of the push-pull nitrile complexes trans-[PtCl2(RCN)2] (R=NEt2, NC5H10). Detection of mercury ions based on mercury-induced switching of enzyme-like activity of platinum/gold nanoparticles.

In this study, bimetallic platinum/gold nanoparticles (Pt/Au NPs) were found to exhibit peroxidase-like activity, and the deposition of mercury was found to switch the enzymatic activity to a catalase-like activity. Based on this phenomenon, we developed a new method for detecting mercury ions through their deposition on bimetallic Pt/Au NPs to switch the catalytic activity of Pt/Au NPs. Pt/Au NPs could be easily prepared through reduction of Au(3+) and Pt(4+) by sodium citrate in a one-pot synthesis. The peroxidase catalytic activity of the Pt/Au NPs was controlled by varying the ratios of Pt
