In Situ Anabolic Activity of Periodontal Pathogens Porphyromonas gingivalis and Filifactor alocis in Chronic Periodontitis.

Porphyromonas gingivalis and Filifactor alocis are fastidious anaerobic bacteria strongly associated with chronic forms of periodontitis. Our understanding of the growth activities of these microorganisms in situ is very limited. Previous studies have shown that copy numbers of ribosomal-RNA precursor (pre-rRNA) of specific pathogen species relative to genomic-DNA (gDNA) of the same species (P:G ratios) are greater in actively growing bacterial cells than in resting cells. The method, so-called steady-state pre-rRNA-analysis, represents a novel culture-independent approach to study bacteria. T…

Concentration of antibodies against Porphyromonas gingivalis is increased before the onset of symptoms of rheumatoid arthritis.

BACKGROUND: The periodontal pathogen Porphyromonas gingivalis is hypothesized to be important in rheumatoid arthritis (RA) aetiology by inducing production of anti-citrullinated protein antibodies (ACPA). We have shown that ACPA precede RA onset by years, and that anti-P. gingivalis antibody levels are elevated in RA patients. The aim of this study was to investigate whether anti-P. gingivalis antibodies pre-date symptom onset and ACPA production. METHODS: A case-control study (251 cases, 198 controls) was performed within the Biobank of Northern Sweden. Cases had donated blood samples (n = 4…

Involvement of an Skp-Like Protein, PGN_0300, in the Type IX Secretion System of Porphyromonas gingivalis.

The oral Gram-negative anaerobic bacterium Porphyromonas gingivalis is an important pathogen involved in chronic periodontitis. Among its virulence factors, the major extracellular proteinases, Arg-gingipain and Lys-gingipain, are of interest given their abilities to degrade host proteins and process other virulence factors. Gingipains possess C-terminal domains (CTDs) and are translocated to the cell surface or into the extracellular milieu by the type IX secretion system (T9SS). Gingipains contribute to the colonial pigmentation of the bacterium on blood agar. In this study, Omp17, the PGN_0…

Activation of RANKL-induced osteoclasts and memory T lymphocytes by Porphyromonas gingivalis is serotype dependant.

AIM: Destructive periodontitis is associated with a Th1-Th17 immune response and activation of RANKL-induced osteoclasts. In addition, Porphyromonas gingivalis K1 and K2 serotypes induce a strong Th1-Th17 response. This study aimed to investigate whether these P. gingivalis serotypes induce higher osteoclasts activation, by increased Th17-associated RANKL production, and an antigen-specific memory T-lymphocyte response. MATERIAL AND METHODS: The RANKL production and TRAP(+) osteoclast induction were quantified on naïve T lymphocytes stimulated with dendritic cells primed with the P. gingivali…

Protective role of the PG1036-PG1037-PG1038 operon in oxidative stress in Porphyromonas gingivalis W83.

As an anaerobe, Porphyromonas gingivalis is significantly affected by the harsh inflammatory environment of the periodontal pocket during initial colonization and active periodontal disease. We reported previously that the repair of oxidative stress-induced DNA damage involving 8-oxo-7,8-dihydroguanine (8-oxoG) may occur by an undescribed mechanism in P. gingivalis. DNA affinity fractionation identified PG1037, a conserved hypothetical protein, among other proteins, that were bound to the 8-oxoG lesion. PG1037 is part of the uvrA-PG1037-pcrA operon in P. gingivalis which is known to be upregul…

Involvement of a periodontal pathogen, Porphyromonas gingivalis on the pathogenesis of non-alcoholic fatty liver disease.

BACKGROUND: Non-alcoholic fatty liver disease (NAFLD) is a hepatic manifestation of metabolic syndrome that is closely associated with multiple factors such as obesity, hyperlipidemia and type 2 diabetes mellitus. However, other risk factors for the development of NAFLD are unclear. With the association between periodontal disease and the development of systemic diseases receiving increasing attention recently, we conducted this study to investigate the relationship between NAFLD and infection with Porphyromonas gingivalis (P. gingivalis), a major causative agent of periodontitis. METHODS: Th…

Mutualistic biofilm communities develop with Porphyromonas gingivalis and initial, early, and late colonizers of enamel.

Porphyromonas gingivalis is present in dental plaque as early as 4 h after tooth cleaning, but it is also associated with periodontal disease, a late-developing event in the microbial successions that characterize daily plaque development. We report here that P. gingivalis ATCC 33277 is remarkable in its ability to interact with a variety of initial, early, middle, and late colonizers growing solely on saliva. Integration of P. gingivalis into multispecies communities was investigated by using two in vitro biofilm models. In flow cells, bacterial growth was quantified using fluorescently conju…

Recognition of the carbohydrate modifications to the RgpA protease of Porphyromonas gingivalis by periodontal patient serum IgG.

Periodontal infections by Porphyromonas gingivalis are associated with a sustained systemic IgG antibody response and elevations in local antibody synthesis to this organism. One of the targets of this response is a protease, RgpAcat, which is an important virulence determinant of this organism. Recently, we demonstrated that this molecule is glycosylated and that the glycan chains are immunologically related to P. gingivalis lipopolysaccharide (LPS) (Curtis et al., Infect Immun 1999;62:3816-3823). In the present study, we examined the role of these glycan additions in the immune recognition o…

Stimulation of splenocytes in C3H/HeJ mice with Porphyromonas gingivalis lipid A in comparison with enterobacterial lipid A.

Porphyromonas gingivalis 381 lipid A induced strong mitogenic response in splenic B cells separated from LPS-non-responsive C3H/HeJ mice as well as LPS-responsive C3H/HeN mice, by using a magnetic cell sorting system. The lipid A also exhibited mitogenic activity in splenic T cells. P. gingivalis lipid A induced lower production of interleukin-1 beta (IL-1 beta) in splenic macrophage cultures and exhibited a comparable IL-6 producing activity in splenic B cells of C3H/HeN mice as compared to Escherichia coli-type synthetic lipid A (compound 506) and monophosphoryl lipid A from Salmonella minne…

Detection of Actinobacillus actinomycetemcomitans, Porphyromonas gingivalis, and Bacteroides forsythus in an A. actinomycetemcomitans-positive patient population.

A population of 33 subjects were selected on the basis that all had tested positive for A, actinomycetemcomitans at some time during the prior 7 years. Most subjects (31/33) belonged to families with a proband with confirmed localized juvenile periodontitis (JP); however, most subjects had no evidence of the typical lesions associated with JP. Two additional subjects with rapidly progressive periodontitis, known to be positive for A. actinomycetemcomitans, were also recruited. The patients with a history of JP had been treated, but were no longer enrolled in a regular maintenance program. With

Resistance to beta-lactam antibiotics and beta-lactamase production of Bacteroides, Porphyromonas and Prevotella strains.

Resistance to beta-lactam antibiotics of 183 clinical isolates belonging to Bacteroides, Porphyromonas and Prevotella was tested by a micro-broth dilution MIC method. Beta-lactamase production was screened by using nitrocefin sticks. Prevalence of resistance to different beta-lactam antibiotics was higher among the Bacteroides strains other than B. fragilis parallel with a beta-lactamase production (88% and 96%, respectively). Resistance was observed less frequently among Porphyromonas and Prevotella strains. Cefoxitin resistance was 11.5% and amoxicillin/clavulanic acid 3.5% among Bacteroides…

Porphyromonas gingivalis promotes oral squamous cell carcinoma progression via the IL-6/EZH2/Snai2 axis.

Oral squamous cell carcinoma (OSCC) is a common malignancy with poor prognosis. Chronic infection with Porphyromonas gingivalis (P.g.) has been implicated in OSCC progression, but the molecular mechanisms underlying P.g.-induced tumor invasion and metastasis remain incompletely understood. ELISA, flow cytometry, proliferation assay and migration assay, were conducted on OSCC primary cells and the HSC-2 cell line under P.g. infection or IL-6 stimulation. Zeste homologue 2 (EZH2) and Snai2 were modulated via overexpression, knockdown, and inhibitor treatments, to evaluate the role of EZH2 and Sn…

Porphyromonas gingivalis Outer Membrane Vesicles-Associated DNA Triggers Inflammation by Inducing IL-6 in Astrocytes.

Porphyromonas gingivalis (Pg), a key pathogen in periodontal disease, is suggested to be involved in the progression of Alzheimer's disease (AD); however, the molecular mechanism remains unclear. We previously reported that Pg-released outer membrane vesicles (OMVs) were detected in the brains of mice after intraperitoneal administration. We here investigated the effects of Pg OMVs on astrocytes, the most abundant glial cells in the central nervous system, which are involved in neuroinflammation. We demonstrated that Pg OMVs increased the expression of interleukin-6 (IL-6) mRNA, which is assoc…

Biochemical and functional characterization of Porphyromonas gingivalis HmuS protein reveals its participation in heme metabolism.

BACKGROUND: Porphyromonas gingivalis lacks the complete heme biosynthesis pathway and does not utilize siderophores to acquire iron. The exact mechanism of iron extraction from heme in this bacterium is unknown. This study aimed to characterize the P. gingivalis HmuSPg protein and to compare its properties with those of the Bacteroides fragilis homologous protein. METHODS: UV-visible spectroscopy was used to demonstrate heme binding and follow heme metabolism. Porphyrin levels in bacterial cells and pigment were determined using HPLC or LC-MS, and iron content using atomic absorption spectros…

Porphyromonas gingivalis Suppresses Interferon-Gamma Signaling in Macrophages Through a Contact-Dependent, Gingipain-Mediated Mechanism.

Interferon signaling serves as a crucial defense mechanism for host cells against intracellular pathogens. Interferon-gamma (IFN-γ) binding to macrophages impacts the expression of approximately 2000 genes, activating them to enhance intracellular bactericidal activity. While various immune evasion strategies of Porphyromonas gingivalis have been extensively studied, the specific mechanisms by which it suppresses IFN-γ-mediated macrophage activation remain insufficiently characterized. In this study, we elucidated the molecular mechanism by which P. gingivalis suppresses interferon signaling i…

Targeting Porphyromonas gingivalis virulence and viability with epimedokoreanin B prevents alveolar bone destruction in vivo.

Periodontal disease is a major oral infectious condition that leads to the degradation of alveolar bone and subsequent tooth loss. Porphyromonas gingivalis, a keystone pathogen in periodontitis, plays a critical role in disease initiation and progression. In a previous study, we demonstrated the anti-P. gingivalis activity of epimedokoreanin B, a prenylated flavonoid derived from Epimedium species. However, its in vivo efficacy against periodontitis remains unclear. Oral administration of epimedokoreanin B attenuated P. gingivalis-induced alveolar bone resorption in murine periodontitis models…
