Three otherwise healthy relatives of patients with congenital adrenal hyperplasia (CAH) due to 21-hydroxylase deficiency and salt-wasting presented with clinical and/or biochemical findings, which exceeded those usually seen in heterozygotes: Two females (1 mother and 1 prepubertal sister of a patient with CAH) had marked hypertrichosis and hirsutism and excreted pregnanetriolone in their urine. The mother had increased basal plasma 17alpha-OH-progesterone (296 ng/100 ml), which increased to 7170 ng/100 ml after ACTH as in homozygotes of CAH. One adult male (brother of a patient with CAH) was

Neutral steroids in urine were determined quantitatively with gaschromatography on capillary columns in a case of benign hydatidiform mole associated with bilateral theca-lutein cysts. A remarkable finding was the very high levels of 17-hydroxypregnanolone and pregnanetriol, which continued to rise until the 15th day after molar evacuation.

The pregnane X receptor (PXR, NR1I2) is a ligand-activated nuclear receptor that regulates xenobiotic and endobiotic metabolism and has been associated with diverse physiological and pathological processes. However, the biological consequences of PXR modulation are highly context-dependent, and the translational relevance of many reported effects remains incompletely defined. The remarkable structural plasticity of the PXR ligand-binding pocket (LBP), together with its context-dependent effects in disease progression, poses substantial challenges for the development to potent and highly select

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a common metabolic disorder with rising global incidence without effective treatment. Emerging evidence highlights bile acids (BAs) as pivotal signalling molecules that modulate genes involved in their own metabolism and lipid homeostasis in hepatic and extrahepatic tissues, thereby influencing MASLD pathogenesis. These regulatory effects are primarily driven by the activation of nuclear receptors, including the farnesoid X receptor (FXR), vitamin D receptor (VDR), pregnane X receptor (PXR), and by the membrane-bound G protein

Emerging evidence suggests that Parkinson's disease (PD) extends beyond the brain and involves early disturbances along the gut-brain axis. Among the metabolites shaping this communication, bacterial products derived from tryptophan, particularly indole compounds, are gaining attention as key biochemical links between intestinal dysbiosis and neurodegeneration. Multi-omics studies consistently show a reduction in commensal bacteria capable of producing beneficial indoles and an enrichment of Enterobacteriaceae that redirect tryptophan catabolism toward toxic intermediates. This shift disrupts

BACKGROUND: As lung cancer treatment has progressed, there has been an increase in awareness of the short- and long-term adverse effects of targeted cancer therapies of tyrosine kinase inhibitors, particularly cardiovascular toxicities.
METHODS: The current study assessed the potential drug-drug interactions using interaction checkers (IBM Micromedex and Drugs.com). Molecular docking was employed to further investigate the involvement of human ether-à-go-go-related gene (hERG) and pregnane X receptor (PXR) proteins to elucidate their potential interactions and their underlying mechanisms.

Long-chain metabolites produced through hepatic and microbiota-associated ω-oxidation of vitamin E are increasingly recognized as bioactive regulators of lipid metabolism and inflammatory pathways. These properties suggest interesting opportunities in drug development and garcinoic acid (GA) - a δ-tocotrienol-derived natural product and a chemically accessible analogue of these metabolites - is a useful probe for investigating their molecular and pharmacological properties. GA has been identified as an agonist of pregnane X receptor, a modulator of peroxisome proliferator-activated receptor γ,

Rifampicin (RIF), a cornerstone drug in tuberculosis treatment, is associated with hepatotoxicity, which represents a significant adverse effect that frequently causes discontinuation of therapy. However, a comprehensive evaluation of the mechanisms underlying RIF-induced hepatotoxicity remains limited, and the identification of highly effective, low-toxicity therapeutic interventions is urgently needed. In this study, we employed a RIF-induced mouse hepatotoxicity model to systematically investigate the cellular and molecular events associated with RIF-induced liver injury. By integrating sin

PURPOSE: Rifampicin is known to induce clindamycin metabolism via CYP3A4/5, but the magnitude of this interaction varies greatly from one patient to another. Because this metabolic induction results from rifampicin activation of the nuclear pregnane X receptor (PXR), the genetic polymorphisms of its NR1I2 gene and that of CYP3A4/5 might predict the risk of clindamycin underdosage when combined with rifampicin. We therefore conducted a study to determine the possible influence of NR1I2 and CYP3A4/5 gene polymorphisms on baseline and rifampicin-induced clindamycin clearance.
METHODS: Eighty-fiv

Cytochrome P450 enzymes (CYPs) are versatile biocatalysts involved in the biosynthesis, activation, and detoxification of a wide range of endogenous and exogenous compounds. While the CYP107 family is primarily associated with macrolide antibiotic biosynthesis, some members, such as OleP (CYP107D1) and CYP107X1, have also been shown to catalyze steroid hydroxylation, albeit with limited substrate scope. In this study, we identified CYP107E41 from Streptoalloteichus sp. KCCM40925 as a novel member of the CYP107 family with selective hydroxylation activity toward both androstane and pregnane ste

Previous studies have focused on advantages and disadvantages of antenatal dexamethasone therapy (ADT) on offspring development, but the effects on the mother have yet to be reported. In this study, the clinical cohort study found that ADT caused maternal susceptibility to cholestatic liver injury, which was manifested by elevated plasma total bile acid (TBA) levels, altered bile acid metabolic profiles and changed gut microbiota (mainly characterized by increased Bacteroides), accompanied by fetal dysplasia associated with high bile acids. Animal experiments confirmed that prenatal dexamethas

Octahydro-tetramethyl-naphthalenyl-ethanone (OTNE) is a synthetic fragrance ingredient. OTNE was evaluated in repeated-dose toxicological studies. Target organs via oral and dermal routes were the liver and skin/liver, respectively. Effects were observed on the thyroid and thyroid hormones, suggesting hypothalamic-pituitary-thyroid axis perturbation. We investigated the molecular initiating event(s) (MIEs), key events (KEs), and adverse outcomes of OTNE-induced thyroid perturbation within an adverse outcome pathway (AOP). Data were generated using new approach methodologies (NAMs) on human, mo

Phenobarbital (PB), a widely used antiepileptic drug, is known to upregulate the expression of numerous drug-metabolizing enzymes and transporters in the liver primarily via activation of the constitutive androstane receptor (CAR, NR1I3). The solute carrier family 13 member 5 (SLC13A5), a sodium-coupled citrate transporter, plays an important role in intracellular citrate homeostasis that is associated with a number of metabolic syndromes and neurological disorders. Here, we show that PB markedly elevates the expression of SLC13A5 through a pregnane X receptor (PXR)-dependent but CAR-independe

In crude oil contaminant plumes, the dissolved organic carbon (DOC) is mainly hydrocarbon degradation intermediates only partly quantified by the diesel range total petroleum hydrocarbon (TPHd) method. To understand potential biological effects of degradation intermediates, we tested three fractions of DOC: (1) solid-phase extract (HLB); (2) dichloromethane (DCM-total) extract used in TPHd; and (3) DCM extract with hydrocarbons isolated by silica gel cleanup (DCM-SGC). Bioactivity of extracts from five wells spanning a range of DOC was tested using an in vitro multiplex reporter system that ev

During middle age, women are less susceptible to nonalcoholic steatohepatitis (NASH) than men. Thus, we investigated the underlying molecular mechanisms behind these sexual differences using an established rat model of NASH. Mature female and male stroke-prone spontaneously hypertensive 5/Dmcr rats were fed control or high-fat-cholesterol (HFC) diets for 2, 8, and 14 weeks. Although HFC-induced hepatic fibrosis was markedly less severe in females than in males, only minor gender differences were observed in expression levels of cytochrome P450 enzymes (CYP)7A1, CYP8B1 CYP27A1, and CYP7B1, and
