[Study progress of special AT-rich sequence binding protein 2].

SATB2 is a transcription factor, which plays an important role in transcriptional regulation and chromatin recombinant by combining with matrix attachment regions. SATB2 is a key regulatory molecule in the progress of osteoblast differentiation and bone matrix formation. Mutations in this gene are associated with congenital craniofacial malformation. In addition, SATB2 is involved in the development of central nervous system, especially the corpus callosum and the pons. At the same time, SATB2 may participate in the process of tumor formation. In malignant tumors, such as breast cancer, the ex

A population approach to eplerenone pharmacokinetics and saturable protein binding.

Eplerenone deviates from linear pharmacokinetics at doses above the therapeutic dose range. In addition, saturable protein binding of eplerenone is observed in in vitro plasma protein binding studies. The purpose of the present study was to clarify the factors contributing to the nonlinear pharmacokinetics of eplerenone. Plasma concentration data for eplerenone and its metabolite SC-70303, to which eplerenone is reversibly converted, obtained from four phase I studies were analyzed using NONMEM. A population pharmacokinetic model incorporating protein binding and the reversible relationship be

Signatures of protein-DNA recognition in free DNA binding sites.

One obstacle to achieving complete understanding of the principles underlying sequence-dependent recognition of DNA is the paucity of structural data for DNA recognition sequences in their free (unbound) state. Here, we carried out crystallization screening of 50 DNA duplexes containing cognate protein binding sites and obtained new crystal structures of free DNA binding sites for three distinct modes of DNA recognition: anti-parallel beta strands (MetR), helix-turn-helix motif + hinge helices (PurR), and zinc fingers (Zif268). Structural changes between free and protein-bound DNA are manifest

Caspase-cleaved TAR DNA-binding protein-43 in Pick's disease.

The hyperphosphorylation and proteolytic modification of the TAR DNA binding protein-43 (TDP-43) is a key finding in a number of neurodegenerative diseases including frontotemporal dementia with ubiquitin-positive inclusions (FTLD-U), amyotrophic lateral sclerosis (ALS), and most recently Alzheimer's disease (AD). To examine whether proteolytic modifications of TDP-43 is a relevant finding in Pick's disease, we utilized a novel site-directed caspase-cleavage antibody based upon a known caspase-3 cleavage consensus site within TDP-43 at position 219. Application of this antibody, termed TDP cas

Methyl-CpG-binding protein 2 polymorphisms and vulnerability to autism.

The methyl-binding protein gene, MECP2, is a candidate for involvement in autism through its implication as a major causative factor in Rett syndrome that has similarities to autism. Rare mutations in MECP2 have also been identified in autistic individuals. We have examined the possible broader involvement of MECP2 as a predisposing factor in the disorder. Analysis of polymorphic markers spanning the gene and comprising both microsatellites and single nucleotide polymorphisms (SNPs) by the transmission disequilibrium test in two collections of families (219 in total), one in the USA and one in

Local expression of lipopolysaccharide-binding protein in human gingival tissues.

BACKGROUND: Lipopolysaccharide-binding protein (LBP) functions as a crucial molecule in innate host defense responses to bacterial challenge through neutralization of bacterial lipopolysaccharide (LPS) and activation of cellular responses. OBJECTIVES: This study was to investigate the expression profile and levels of LBP in gingival tissues and their associations with periodontal health and disease. METHODS: Gingival biopsies were collected from 44 chronic periodontitis patients, including periodontal pocket tissues (PoTs) and the adjacent healthy gingival tissues (HT-Ps), as well as from 15

Myosin binding protein C: structural abnormalities in familial hypertrophic cardiomyopathy.

The muscle protein myosin binding protein C (MyBPC) is a large multi-domain protein whose role in the sarcomere is complex and not yet fully understood. Mutations in MyBPC are strongly associated with the heart disease familial hypertrophic cardiomyopathy (FHC) and these experiments of nature have provided some insight into the intricate workings of this protein in the heart. While some regions of the MyBPC molecule have been assigned a function in the regulation of muscle contraction, the interaction of other regions with various parts of the myosin molecule and the sarcomeric proteins, actin

Babesia divergens: cloning of a Ran binding protein 1 homologue.

Babesia divergens is an Apicomplexa transmitted to bovines by its acarian vector, the tick I. ricinus. Babesia divergens merozoites have an intraerythrocytic development in the blood of infected mammals. The nucleocytoplasmic transport system in this parasite is not yet characterized and no protein involvement in such transport has been described. In this report, we describe the cloning of a protein that shares important homologies with Ran binding protein 1. This protein in Eukaryote belongs to the nucleocytoplasmic transport system.

Lipopolysaccharide (LPS)-binding protein inhibits responses to cell-bound LPS.

Lipopolysaccharide (LPS)-binding protein (LBP) is an acute phase reactant that may play a dual role in vivo, both potentiating and decreasing cell responses to bacterial LPS. Whereas low concentrations of LBP potentiate cell stimulation by transferring LPS to CD14, high LBP concentrations inhibit cell responses to LPS. One inhibitory mechanism involves the ability of LBP to neutralize LPS by transferring it to plasma lipoproteins, whereas other inhibitory mechanisms, such as the one described here, do not require exogenous lipoproteins. Here we show that LBP can inhibit monocyte responses to L

Aberrant metal binding by prion protein in human prion disease.

Human prion diseases are characterized by the conversion of the normal prion protein (PrP(C)) into a pathogenic isomer (PrP(Sc)). Distinct PrP(Sc) conformers are associated with different subtypes of prion diseases. PrP(C) binds copper and has antioxidation activity. Changes in metal-ion occupancy can lead to significant decline of the antioxidation activity and changes in conformation of the protein. We studied the trace element status of brains from patients with sporadic Creutzfeldt-Jakob disease (sCJD). We found a decrease of up to 50% of copper and an increase in manganese of approximatel

The latent transforming growth factor beta binding protein (LTBP) family.

The transforming growth factor beta (TGFbeta) cytokines are a multi-functional family that exert a wide variety of effects on both normal and transformed mammalian cells. The secretion and activation of TGFbetas is regulated by their association with latency-associated proteins and latent TGFbeta binding proteins (LTBPs). Over the past few years, three members of the LTBP family have been identified, in addition to the protoype LTBP1 first sequenced in 1990. Three of the LTBP family are expressed in a variety of isoforms as a consequence of alternative splicing. This review summarizes the diff

Vacancy affinity capillary electrophoresis to study competitive protein-drug binding.

The suitability of the vacancy affinity capillary electrophoretic method (VACE) for study of displacement of a target drug from a protein by simultaneously administered drugs was investigated. As test system, the displacement of warfarin from bovine serum albumin (BSA) by furosemide and phenylbutazone was selected. It appears that the displacement can be observed well from the shift of the actual mobility of warfarin when a displacer drug is added. Also, the competitive action of the displacer drugs (affinity for BSA) is clearly visible. The VACE method seems to be attractive for rapid assessm

Fluoride activation of the Rho family GTP-binding protein Cdc42Hs.

Aluminum tetrafluoride (AlF4-) activation of heterotrimeric G-protein alpha-subunits is a well established aspect of the biochemistry of these proteins; however, until recently it has been thought that AlF4- does not mediate effects on the Ras superfamily of low molecular weight GTP-binding proteins. Recent work demonstrating aluminum fluoride-induced complex formation between Ras and its GTPase-activating proteins (RasGAP and NF1) has provided important insights into the mechanism of GAP-stimulated GTP hydrolysis. We have characterized the AlF4--induced complex formation between the GDP-bound

Tissue distribution of the receptor for plasma retinol-binding protein.

The tissue distribution of the retinol-binding-protein receptor has been studied by using a cell-free binding assay. High binding activity was found in placenta, retina pigment epithelial cells, bone marrow and kidneys. Specific binding activity was also found in the small intestines, spleen and liver, and to a lesser extent in lung. Scatchard analysis revealed that the difference in binding activity was due to variations in receptor level and not affinity changes. When the kidneys were separated into cortex and medulla we found that almost all the specific binding activity present in kidneys

Binding and endocytosis of 39 kDa protein by MDBK cells.

A 39 kDa protein copurifies with the low density lipoprotein receptor-related protein (LRP) and regulates ligand interactions with LRP. In our recent studies on the clearance of the 39 kDa protein in vivo, we demonstrated that once the liver LRP receptors were saturated, the kidney became the major organ responsible for the 39 kDa protein clearance (Warshawsky et al., 1993, J. Clin. Invest., 92:937-944). The current study was undertaken in order to investigate the potential binding and cellular processing of the 39 kDa protein by kidney-derived MDBK cells. Herein we demonstrate specific, high-

Novel reticular calcium binding protein is purified on taipoxin columns.

We identified, by affinity chromatography, two putative binding proteins for the presynaptic snake venom toxin taipoxin. We have previously characterized one of these proteins [neuronal pentraxin (NP)] as a neuronally secreted protein with homology to acute-phase proteins. Here we report the identification of the second protein as a 49-kDa lumenal calcium binding protein that we have named taipoxin-associated calcium binding protein 49 (TCBP-49). This protein contains six EF-hand putative calcium binding domains and the carboxyl-terminal sequence His-Asp-Glu-Leu (HDEL), identical to the yeast

Immunocytochemical demonstration of interphotoreceptor retinoid-binding protein in cerebellar medulloblastoma.

Previously, immunoreactive rod-opsin and S-antigen (arrestin), two highly characteristic markers of retinal photoreceptors and pinealocytes, were shown to be present in certain medulloblastoma cells. It, thus, has been suggested that such cells differentiate along the photoreceptor lineage. This is corroborated in the present immunocytochemical investigation using antibodies against another photoreceptor-cell marker, the interphotoreceptor retinoid-binding protein (IRBP). As shown in preparations of human retina and pineal organ, IRBP can be successfully demonstrated in formalin-fixed and para

Protein binding of xanthine derivatives to guinea pig serum albumin.

Binding of the bronchodilators N3-alkylxanthine and N3-alkyl-N1-methylxanthine derivatives to guinea pig serum albumin was investigated in vitro using the ultrafiltration method. A marked difference in the binding parameters of xanthine derivatives was observed, and binding was shown to be concentration dependent. Significant relations were observed among their binding parameter, dissociation constant (Kd), and hydrophobicity (log PC). The extent of binding of xanthine derivatives was increased both when a N3-methyl group was replaced by a longer alkyl chain and when a N3-alkylxanthine molecul

Calcium binding protein immunoreactivity in a reptilian thalamic reticular nucleus.

Immunoreactivity to antisera directed against a variety of calcium binding proteins was investigated in the thalamic reticular nucleus of one species of reptiles, Caiman crocodilus. Cells and fibers were immunoreactive for parvalbumin whereas fibers but not neurons were immunoreactive for calbindin and calcitonin gene related peptide. These data support the observations that Caiman thalamic reticular nucleus contains at least two neuronal populations. One group, whose cells are immunoreactive for parvalbumin, forms at least part of the projection to the dorsal thalamus. The other group, which

High prostaglandin-E1 binding to serum protein in allergic subjects.

Protein binding to both salicylate and 3H-labelled prostaglandin-E1 ([3H]PGE1) was examined in the sera of 22 allergic and 16 normal individuals. Protein binding to salicylate (P less than 0.001) and to [3H]PGE1 (P less than 0.01) was significantly greater in the allergic than in the normal group. The nature of the binding sites of salicylate and PGE1 was investigated with two fluorescent probes, dansylamide and dansylsarcosine as specific marker ligands for established Sites I and II, found to be specific for anionic drugs. The serum protein of 11 allergic subjects showed a higher binding at
