Injury to retinal ganglion cell (RGC) axons leads to selective loss of RGCs and vision. Previous studies have shown that exogenous neurotrophic factors promote RGC survival. We investigated the neuroprotective effects of oncostatin M (OSM), a member of the IL-6 family of cytokines, on pattern electroretinogram (PERG) and RGC survival after optic nerve crush (ON-crush) in the mouse. BALB/C mice received ON-crush in the left eyes for either 4-second or 1-second duration (4-s or 1-s). Fluoro-gold retrograde labeling was used to identify RGCs. RGC function was assessed by PERG measurement. OSM or

Intrinsically photosensitive retinal ganglion cells (ipRGCs) respond directly to light and are responsible of the synchronization of the circadian rhythm with the photic stimulus and for the pupillary light reflex. To quantify the total population of rat-ipRGCs and to assess their spatial distribution we have developed an automated routine and used neighbour maps. Moreover, in all analysed retinas we have studied the general population of RGCs - identified by their Brn3a expression - and the population of ipRGCs - identified by melanopsin immunodetection - thus allowing the co-analysis of thei…

Valproate (VPA) is an anticonvulsant and mood-stabilizing drug. It is a broad-spectrum histone deacetylase inhibitor with neuroprotective effects. We investigated whether VPA reduces retinal neuronal death induced by optic nerve crush (ONC). To evaluate further VPA-mediated neuroprotection on retinal ganglion cells (RGCs), another histone deacetylase (HDAC) inhibitor, sodium butyrate (SB) was compared with VPA. Adult male Wistar rats were subjected to ONC injury. VPA and SB were administered subcutaneously 1 day prior to ONC until sacrifice 14 days later. RGC density was counted using hematoxy…

In vertebrate retina, melatonin regulates various physiological functions. In this work we investigated the mechanisms underlying melatonin-induced potentiation of glycine currents in rat retinal ganglion cells (RGCs). Immunofluorescence double labelling showed that rat RGCs were solely immunoreactive to melatonin MT(2) receptors. Melatonin potentiated glycine currents of RGCs, which was reversed by the MT(2) receptor antagonist 4-P-PDOT. The melatonin effect was blocked by intracellular dialysis of GDP-beta-S. Either preincubation with pertussis toxin or application of the phosphatidylcholine…

PURPOSE: Methylprednisolone (MP) is commonly used to treat traumatic optic neuropathy and optic neuritis, but its benefit in terms of neuronal survival remains controversial. The aim of this study was to investigate the effects of MP on retinal ganglion cell (RGC) survival and visual function after ischemia in rats.
METHODS: Ocular ischemia was induced by elevated intraocular pressure. Rats were treated with NaCl, 1 mg/kg/d, or 30 mg/kg/d intraperitoneal MP over 3 days. RGCs were labeled retrogradely 4 days after ischemia and were quantified 6 days later. Post-ischemic retinal function was as

Generally, nerve injury of adult mammalian CNS neurons leads to a retrograde neuronal degeneration and cell death. The retinal ganglion cells (RGCs) of rat fail to regenerate and become apoptotic after optic nerve injury. In contrast, goldfish RGCs can survive and regrow their axons after injury. Focusing on this different response of RGCs in both species to optic nerve injury, we compared cell death and cell survival signals in the rat and goldfish RGCs after optic nerve injury. In goldfish retina, levels of phospho-Akt (p-Akt) and phospho-Bad (p-Bad) first rapidly increased at 3-5 days after…

Ganglion cells were fluorescently labeled, dissociated from 7- to 11-day-old rodent retinas, and placed in tissue culture. Whole-cell recordings with patch electrodes were obtained from solitary cells lacking processes, which permitted a high-quality space clamp. Both GABA (1-200 microM) and glycine (10-300 microM) produced large increases in membrane conductance in virtually every ganglion cell tested, including ganglion cells from different size classes in both rats and mice. Taurine evoked responses similar to those of glycine, but considerably greater concentrations of taurine (150-300 mic

Behavioral perimetry methods were used to assess the monocular visual fields of 12 cats that had received intraocular injections of antibodies against large (alpha/Y) retinal ganglion cells. The antibodies produced defects in head and eye orientation responses to stimuli presented in the nasal visual field of the treated eye; responses to stimuli in the temporal visual hemifield were normal. Similar results were seen in cats that received antibody injections at 4 weeks of age or as adults. In the context of previous results, these findings suggest that a loss of Y cells in the lateral genicula…

Thyroid eye disease (TED) is characterized by progressive orbital inflammation. In severe cases, it can lead to dysthyroid optic neuropathy (DON), involving death of retinal ganglion cells and permanent vision loss. Imaging with optical coherence tomography (OCT) has enabled noninvasive examination of the structural impact of the disease, including of the retinal nerve fiber layer (RNFL), the ganglion cell layer (GCL), and the inner plexiform layer (IPL). In this study, we systematically review 20 studies (897 patients, 1430 eyes) to evaluate changes in retinal thickness in patients with TED, …

Ischemia-reperfusion (I/R) injury represents a prevalent etiology of permanent vision loss across multiple ocular pathologies. Apoptotic death and inflammation of retinal ganglion cells (RGCs) induced by I/R injury constitutes a major contributor to visual dysfunction. Nevertheless, the molecular mechanisms that initiate the apoptotic and inflammation cascade in RGCs remain largely uncharacterized. Recent investigations have demonstrated that PRD1-BF1-RIZ1 homeodomain protein 16 (PRDM16) attenuates renal cell death in acute kidney injury. However, its specific function and regulatory mechanism…

BACKGROUND: As the structural framework and conduit for axonal material transport, microtubule stability plays a crucial role in the repair of optic nerve injury. Tau maintains the integrity of these transport tracks by promoting microtubule bundling. However, injury-induced aberrant phosphorylation of Tau markedly impairs its microtubule-binding capacity, leading to microtubule disassembly and eventual neurodegeneration. This study investigated Tau-mediated regulation of microtubule homeostasis and explored its potential application in axonal repair and regeneration following optic nerve inju
