Inactivated rotavirus vaccines: a priority for accelerated vaccine development.

Live oral rotavirus vaccines have proven to be generally safe and effective to prevent severe dehydrating diarrhea among children in high and some middle income countries. However, concerns linger about rare but severe adverse events, such as intussusception and their efficacy against the full range of rotavirus serotypes. More importantly, live oral vaccines have been less immunogenic and results of trials to assess their efficacy in poor children of both Africa and Asia will not be available for 2-3 years. This review describes the rationale for developing an inactivated rotavirus vaccine (I

Detection of rotaviruses and intestinal lesions in broiler chicks from flocks with runting and stunting syndrome (RSS).

The intestinal tract and intestinal contents were collected from 34 stunted, 5-to-14-day-old broiler chicks from eight flocks with runting and stunting syndrome (RSS) in Northern Germany to investigate intestinal lesions and the presence of enteric pathogens with a special focus on rotaviruses (RVs). Seven chicks from a healthy flock were used as controls. Severe villous atrophy was seen in chicks from six flocks with RSS but not in the control flock. Lesions were often "regionally" distributed in the middle-to-distal small intestine. Transmission electron microscopy (TEM), polyacrylamide-gel

Human bocavirus infection in children hospitalized with acute gastroenteritis in China.

BACKGROUND: Human bocavirus (HBoV) was first identified in children with acute respiratory-tract infections, but recent studies have revealed that HBoV is also frequently detected in fecal specimens from children with gastroenteritis. OBJECTIVES: To investigate the prevalence of HBoV in children hospitalized with gastroenteritis in different areas of China. STUDY DESIGN: Employing ELISA, RT-PCR or PCR, we evaluated 1216 fecal samples for common diarrheal agents from children aged less than 5-year-old hospitalized with acute gastroenteritis. MEGA software was used to construct phylogenetic tr

[Epidemiology of viral nosocomial infections in pediatrics].

Nosocomial viral infections account for at least 5% of the total of NI and reach 23% in pediatric wards. The nosocomial infection (NI) incidence rate varies from 0.59 to 0.72 per 100 patients in pediatric wards. Many viruses have been associated with NI in pediatric wards. Rotavirus and respiratory syncytial virus (RSV) are the most frequent. Other viruses frequently identified are: calicivirus, adenovirus, astrovirus, influenza et para-influenza, rhinovirus and coronavirus. Asymptomatic infections occur frequently. The period of communicability varies and depends on the virus. It often begins

[Trend analysis of rotavirus diarrhea incidence among children aged 5 years and under in Shanghai City from 2015 to 2023].

Objective: To analyze the temporal trend of reported incidence of rotavirus diarrhea among children aged 5 years and under in Shanghai from 2015 to 2023, and to identify the effects of age, period, and birth cohort. Methods: Confirmed cases of other infectious diarrhea, a legally notifiable Class C infectious disease, reported between July 1, 2015, and June 30, 2023, among residents of Shanghai were selected. Rotavirus-positive cases aged 5 years and under were further identified, and the reported incidence was calculated. A Bayesian age-period-cohort (BAPC) model was established to assess the

The efficiency of rotavirus A spread to extraintestinal tissues is not determined by the levels of its replication in the gut.

Rotavirus A (RVA) spreads to multiple extraintestinal organs; however, it is not well understood what viral or host characteristics regulate the efficiency of this spread. We conducted this study to determine whether more efficient intestinal RVA replication leads to a higher rate of its extraintestinal spread. We also examined the distribution of known RVA glycan receptors in different tissues to better understand their potential role in facilitating viral dissemination to extraintestinal sites. We inoculated germ-free pigs with porcine OSU G5P[7] characterized by remarkably robust in vitro/i

Self-controlled risk interval study of rotavirus vaccine safety: Findings and implications.

BACKGROUND: The self-controlled case series (SCCS) is often used to monitor vaccine safety. The evaluation of intussusception after the rotavirus vaccine is complicated because the baseline rate varies with age. Time-varying baseline risk adjustments with data from unexposed cohorts are utilised. Self-controlled risk interval (SCRI), with a shorter observation period, can also mitigate the problem by studying a control period close to the risk period. OBJECTIVE: An Indian rotavirus vaccine has previously been studied using SCCS. The risk of intussusception in the high-risk windows (21 days af

Interferon-λ3 alleviates intestinal epithelium injury induced by porcine rotavirus in mice.

Interferons are a group of glycoproteins that are expressed in various cell types in their inflammatory responses to infections. In this study, we explored the protective effects of porcine interferon-λ3 (PIFN-λ3) on intestinal inflammation and injury in mice induced by porcine rotavirus (PRV). BALB/c mice were administrated by PIFN-λ3 or phosphate buffer solution (PBS) for three days prior to PRV infection. We show that PRV infection caused acute inflammatory responses in mice, as indicated by increases in serum concentrations of inflammatory cytokines such as the interlukin-1β (IL-1β), inter

Cost-utility Analysis of Rotavirus Vaccines Including the Latest Evidence and Data as of June 2020 in Japan.

BACKGROUND: In several health economic analyses of rotavirus vaccines in Japan, all were not cost-effective from the healthcare payer perspective (HPP) but generally cost-effective from the societal perspective (SP). However, few studies have incorporated clinically significant factors of vaccine herd immunity, convulsions with gastroenteritis, encephalopathies, nosocomial infections, death, and intussusception as a vaccine side effect. A cost-utility analysis incorporating these were conducted. METHODS: We used Bakir's decision-tree model and data in Japan with 94% coverage rate, 5-year time

Treading a HOSTile path: Mapping the dynamic landscape of host cell-rotavirus interactions to explore novel host-directed curative dimensions.

Viruses are intracellular pathogens and are dependent on host cellular resources to carry out their cycles of perpetuation. Obtaining an integrative view of host-virus interaction is of utmost importance to understand the complex and dynamic interplay between viral components and host machineries. Besides its obvious scholarly significance, a comprehensive host-virus interaction profile also provides a platform where from host determinants of pro-viral and antiviral importance can be identified and further be subjected to therapeutic intervention. Therefore, adjunct to conventional methods of

Incidence of intussusception before and after the introduction of rotavirus vaccine in Korea.

BACKGROUND: Recent studies have reported that after the introduction of rotavirus vaccine the incidence of intussusception did not change among infants, or slightly increased at the age immediately after the first dose. The rotavirus vaccines were introduced in Korea for private market use in 2007-2008. We investigated the incidence of intussusception before (2002-2006) and after (2009-2015) the vaccine introduction in Korea. METHODS: We conducted an interrupted time series study that used data from the Korean National Health Insurance database to identify infants (<12 months of age) who were

Rotavirus vaccines for the developing world.

PURPOSE OF REVIEW: The authors discuss the most relevant information in the field of rotavirus vaccines published from October 2007 to June 2009; new information on the virus, host response and disease burden that relate to our understanding of vaccine mechanisms and impact are discussed. The review will focus on the role of the vaccines for the developing world but this does not preclude the relevance of these vaccines for children living in the industrialized world. RECENT FINDINGS: Immune mechanisms involved in rotavirus-associated immunity potentially relevant for vaccine-associated immun

Viral gastroenteritis.

There are many viruses infecting the human gut: some are found to cause acute gastroenteritis regularly (although not always) like rotaviruses, enteric adenoviruses, small round structured viruses and astroviruses; others enter the host via the gut and most often cause systemic infection (entero-viruses, parvoviruses); and others are not regularly associated with human disease (reoviruses, viruses of the Coronaviridae family). The human gut can also be infected directly by HIV and, as a consequence of immuno-suppression, by viruses of the Herpesviridae family. Most remarkable during the last o

A broad assessment of rotavirus vaccine safety in infants in Korea: Insights from a data-driven signal detection approach.

In light of the widespread use of rotavirus vaccines, there is a pressing need to perform thorough, large-scale surveillance to actively monitor safety. This study aimed to identify potential adverse events following rotavirus vaccination in infants. Using a nationwide linked database of the national immunization registry and health insurance claims data, we identified infants vaccinated with the first dose of rotavirus vaccine between January 2016 and October 2022. The self-controlled tree-temporal scan statistics method analyzed all incident diagnoses recorded within 56 days post-vaccination

Rotavirus-Associated Myositis in a 12-Year-Old Child.

Rotavirus infection is a common cause of pediatric gastroenteritis, often self-limited but occasionally associated with extraintestinal complications. This report describes a 12-year-old vaccinated girl who presented with acute gastroenteritis and an incidental elevation of serum creatine kinase (CK). Despite the absence of muscle pain or weakness during hospitalization, laboratory findings indicated skeletal muscle involvement, with elevated CK and aldolase levels, and a predominance of CK-MM isoenzyme. Myocardial involvement and rhabdomyolysis were ruled out. The patient recovered with suppo

Viral agents of acute gastroenteritis in German children: prevalence and molecular diversity.

Acute gastroenteritis is a major source of morbidity and mortality among young children in developed and developing countries. Enteropathogenic viruses are regarded as particularly relevant causative agents. Between February 2001 and January 2002, fecal specimens were obtained from German children admitted to hospital with acute gastroenteritis and examined for rotaviruses, Noroviruses, enteric adenoviruses, and astroviruses using (RT-)PCR methods. Of the 59% (129/217) samples positive for > or =1 viral agent, 79% (102/129) carried rotavirus, whereas Norovirus was detected in 35% (45/129), ent
