The RAD4 gene of Saccharomyces cerevisiae, which is essential for the nucleotide excision repair, was isolated from a yeast genomic library and the expression of this gene has been investigated. RAD4 mRNA was approximately 2.3 kb and di not contain intervening sequence, as determined by S1 nuclease mapping and Northern blot analysis. Transcription start site was located at 48 bp upstream of the ATG initiation codon. RAD4 gene is not induced by UV light damages as indicated by the absence of change in mRNA level after UV exposure in wild type yeast cells. The size of Rad4 protein overexpressed …

The exposure to ionizing radiation of radiosensitive mutants of diploid yeast Saccharomyces cerevisiae deficient in double-strand break repair results in formation of morphologically unstable colonies. Some characteristics of this process were studied. The results obtained are consistent with the hypothesis on relationship between DNA double-strand breaks or their repair with the formation of unstable clones of diploid yeast cells.

Induced mitotic chromosome loss was assayed using diploid yeast strain S. cerevisiae D61.M. The test relies upon the uncovering and expression of multiple recessive markers reflecting the presumptive loss of the chromosome VII homologue carrying the corresponding wild-type alleles. An interlaboratory study was performed in which 12 chemicals were tested under code in 2 laboratories. The results generated by the Berkeley and the Darmstadt laboratories were in close agreement. The solvents benzonitrile and methyl ethyl ketone induced significantly elevated chromosome loss levels. However, a trea

We introduced CEN6 DNA via integrative transformation into the right arm of chromosome II in a haploid Saccharomyces cerevisiae strain thus creating a dicentric chromosome. The majority of the transformed cells did not grow into colonies as concluded from control transformations with mutated CEN6 DNA. Five percent of the initial transformants with the wild-type centromere gave rise to well growing cells. We analysed the probable fate of the dicentric chromosome in two transformants by electrophoretic separation of chromosome sized DNA and by hybridizations with chromosome II DNA probes. We fou…

Saccharomyces cerevisiae strain XV185-14C for reversion studies was used to investigate the genetic activity of alpha-benzene hexachloride dibutyl phthalate and trichloroethylene. The results indicate that none of the three compounds was genetically active when yeast cells were treated in phosphate buffer (pH 7.0) in the absence of metabolic conversion. However, in the presence of the 9000 g supernatant of mice liver homogenate, NADP, glucose-6-phosphate, phosphate buffer (PH 7.4), MgCl2, KCl, the components which were used for the metabolic conversion, trichloroethylene porved to be a powerfu…

A mutant has been isolated which carries a nuclear mutation capable of suppressing certain aspects of the phenotype imposed by a specific mitochondrial mutation. The mitochondrial mutation [tso-r] confers cold sensitivity to growth on nonfermentable substrates and resistance to oligomycin. When both the mitochondrial and nuclear mutations are present in the same cell the cell is phenotypically cold resistant but retains a high level of oligomycin resistance. The extent of cold sensitivity suppression is dependent upon other unspecified nuclear genes. The molecular basis for the suppression may

Modifications of a commercial 2,450-megahertz microwave oven were made so that 6 ml of microbial suspension could be exposed to the microwav … off. Continuous application of microwaves to suspensions of 10(8) to 10(9)Streptococcus faecalis or Saccharomyces cerevisiae per ml appeared to produce no lethal effects other than those produced by heat. Respiration rates of microwave-exposed Scerevisiae were directly related to decreases in viable count produced by increased microwave exposure times.

Faba bean (Vicia faba L.) is a high-protein legume limited by its sensory profile and antinutritional factors like RFOs (Raffinose Family Oligosaccharides). The impact of solid-state fermentation (SSF) with Saccharomyces cerevisiae on properties of starch-(SF) and protein-rich (PF) faba bean flours was investigated. Proximate composition, amino acid profile, digestible carbohydrates, organic acid contents, and techno-functional properties were determined. Sensory properties of SF- and PF-based crepes were also assessed. SSF reduced digestible carbohydrates by 10% (SF) and 22% (PF). RFOs concen…

The nutritional deficiency of honey, particularly in yeast assimilable nitrogen (YAN), represents a major limitation for mead production, often resulting in sluggish fermentation and accumulation of undesirable metabolites. This study evaluated the jabuticaba (Plinia jaboticaba) aqueous extract as a natural nutritional adjunct for mead fermentation. Musts containing 0 (control), 10, 20, and 30% (v/v) extract were fermented with Saccharomyces cerevisiae M05 and characterized using kinetic modeling, physicochemical analysis, and volatile profiling by GC-FID. The supplementation increased YAN fro…

Mismatch repair (MMR) is a highly conserved DNA repair pathway that recognizes mispairs that occur spontaneously during DNA replication and coordinates their repair. In Saccharomyces cerevisiae, Msh2-Msh3 and Msh2-Msh6 initiate MMR by recognizing and binding insertion or deletion (in/del) loops up to ∼17 nucleotides (nt.) and base-base mispairs, respectively; the 2 complexes have overlapping specificity for small (1-2 nt.) in/dels. The DNA-binding specificity for the 2 complexes resides in their respective mispair binding domains (MBDs) and has distinct DNA-binding modes. Msh2-Msh3 also plays …

Yeast sugar-induced cell death (SICD) is a phenomenon whereby cells lose viability in the presence of glucose and the absence of other nutrients to support their growth. Both stationary- and exponential-phase cells of Saccharomyces cerevisiae undergo SICD. The majority of studies on SICD are focused on S. cerevisiae; however, since glucose is a universal carbon source for yeast, we hypothesized that a similar response may be seen in other species of yeast. In this study, we sought to conduct a comparative analysis of SICD in S. cerevisiae and Candida spp. We first established that SICD is prev…

Saccharomyces cerevisiae's ability to withstand formic acid, the most impactful weak acid on yeast growth and fermentation commonly encountered in lignocellulosic bioethanol, is decisive towards industrial applications. Nevertheless, the mechanisms of formic acid resistance inS. cerevisiaeremain poorly understood. This study explored the transcriptional and metabolic responses of three strains - two resistant (YI30 and CESPLG05), and the sensitive DSM 70449 - to 4.0 g/L formic acid for defining the first model of formic acid resistance in S. cerevisiae. Resistance was linked to the upregulatio…

BACKGROUND: The 2-phenylethanol (2-PE) tolerance phenotype is crucial to the production of 2-PE, and Pdr1p mutation can significantly increase the tolerance of 2-PE in Saccharomyces cerevisiae. However, its underlying molecular mechanisms are still unclear, hindering the rational design of superior 2-PE tolerance performance.
RESULTS: Here, the physiology and biochemistry of the PDR1_862 and 5D strains were analyzed. At 3.5 g/L 2-PE, the ethanol concentration of PDR1_862 decreased by 21%, and the 2-PE production of PDR1_862 increased by 16% than those of 5D strain. Transcriptome analysis show…

Saccharomyces boulardii (S. boulardii) is a fungal probiotic used to treat digestive disorders. However, the mechanism(s) by which S. boulardii affects the small intestine remains unclear. Here, we aimed to explore the effects of S. boulardii on the small intestine and the underlying mechanisms in mice with loperamide-induced constipation. While S. boulardii administration did not fully reverse the alterations in loperamide-induced defecation parameters, it altered the small intestinal floral composition toward a community conducive to alleviate constipation. Moreover, S. boulardii up-regulate…

Quantum dots (QDs) have been highly sought after in the past few decades for their potential to be used in many biomedical applications. However, QDs' cytotoxicity is still a major concern that limits the incorporation of QDs into cutting-edge technologies. Thus, it is important to study and understand the mechanism by which QDs exert their toxicity. Although many studies have explored the cytotoxicity of quantum dots through the transcriptomic level and reactive species generation, the impact of quantum dots on the expression of cellular protein remains unclear. Using Saccharomyces cerevisiae…
