Neutralization Capacity of Tissue Alterations Caused by the Venoms of the Most Dangerous Scorpions in North Africa Using a Selective Antivenom.

In North Africa, scorpion stings pose an urgent public health problem, particularly for children with high morbidity and mortality rates. The main species implicated are the Androctonus mauretanicus (Am), Androctonus australis hector (Aah), and Buthus occitanus (Bo). Immunotherapy is the specific therapeutic approach aimed at directly neutralizing toxins, despite their severity and rapid diffusion. In the present study, we evaluate, histologically and immunohistologically, the neutralization potency of the selective antivenom produced against, among other species, the Am, Aah, and Bo at the le

Why does the number of dangerous species of scorpions increase? The particular case of the genus Leiurus Ehrenberg (Buthidae) in Africa.

The aim of this contribution is to bring some precise information on the reasons why the number of noxious scorpion species is constantly gr … ssible and too technical for non-experts. The exposed example could be taken from several groups of scorpions possessing infamous species, but the choice went to the genus Leiurus Ehrenberg, 1828 distributed from North Africa to the Middle East. Maybe this contribution will help to explain why so numerous cases of species misidentification are regularly pre …

An in vitro comparative study upon the toxic properties of the venoms from Hemiscorpius lepturus, Androctonus crassicauda and Mesobuthus eupeus scorpions.

The aim of the present study was to compare the toxic effects of the venoms from Hemiscorpius lepturus (H. lepturus), Androctonus crassicauda (A. crassicauda) and Mesobuthus eupeus (M. eupeus). For this purpose, three in vitro models were employed to compare the toxic effects of various concentrations of the venoms from these three scorpions, namely: hemolytic potential using human RBCs, phospholipase activity using Saubouraud's dextrose agar (SDA) supplemented with 2% egg yolk and lactate dehydrogenase (LDH) enzyme releasing effect using K562 leukemia cell line. In addition, the neutralizing …

First epidemiological reports of scorpion stings by Tityus confluens Borelli, 1899 (Scorpiones: Buthidae) in southern Brazil: an upcoming medically important species in Paraná.

Scorpion envenomation is a growing public health concern in Brazil. The species Tityus confluens Borelli, 1899 has historically been overlooked as a major threat, although its recent presence in Paraná raises new concerns. This study documents for the first time 30 epidemiological reports of T. confluens stings in Paraná, primarily in Foz do Iguaçu (86.7 %) between 2012 and 2024. Most cases involved males (66.7 %), with victims exhibiting local symptoms such as pain (100 %), edema (60 %), and less commonly other manifestations as erythema and tingling (3.3 %). The most commonly stung body part

Venom diversity in the Neotropical scorpion genus Tityus: Implications for antivenom design emerging from molecular and immunochemical analyses across endemic areas of scorpionism.

Scorpions of the Neotropical genus Tityus are responsible for most severe envenomations in the Caribbean, South America, and Lower Central America (LCA). Although Tityus is taxonomically complex, contains high toxin polymorphism, and produces variable clinical manifestations, treatment is limited to antivenoms produced against species with restricted distributions. In this study, we explored the compositional and antigenic diversity of Tityus venoms to provide improved guidelines for the use of available antivenoms at a broader geographic scale. We used immunoblotting, competitive ELISA, and i…

Structural underpinnings of human Slo1 inhibition by scorpion and fungal toxins.

Slo1 channels regulate key electrochemical signaling events in a variety of excitable and nonexcitable cells. Here, we have investigated the mechanisms by which distinct small molecules inhibit human Slo1 (hSlo1) channel activity using single-particle cryo-EM, liposome flux, and toxin-binding assays. We find that unlike classical permeation blockers like scorpion toxins, indole diterpene (ID) class of fungal mycotoxins, paxilline and penitrem A, are ensconced in a binding pocket, deep within the putatively closed hSlo1 pore and sterically restrict its opening. Binding of paxilline to its gatin

Scorpionism in the Amazon: Evidence, Challenges and Actions for Controlling a Neglected Disease.

Scorpion sting envenomations impose an increasing burden in the Amazon and result in a considerable impact on public health. The genus Tityus accounts for nearly all clinically relevant cases, with T. metuendus, T. silvestris and T. obscurus being responsible for most of envenomations in the region. In the state of Amazonas, Manaus and Apuí account for most of cases reported, with an evident expansion to other regions, including the Solimões River region and the Upper Negro River region, where most of the exposed population is found in Indigenous and riverine communities. The increasing burden

Properties and Pharmacology of Scorpion Toxins and Their Biotechnological Potential in Agriculture and Medicine.

Scorpion venoms contain a wide range of toxins that interact with a variety of target molecules (ion channels, receptors and enzymes) associated with synaptic transmission, action potential propagation, cardiac function, hemostasis and other physiological systems. Scorpion toxins are also active towards bacteria, viruses, fungi and parasites. Such interactions make scorpion toxins useful lead molecules for developing compounds with biotechnological and therapeutic applications, and as tools for cell biology. In addition, scorpion toxins act as insectotoxins, with promising applications as inse

Short-Chained Linear Scorpion Peptides: A Pool for Novel Antimicrobials.

Scorpion venom peptides are generally classified into two main groups: the disulfide bridged peptides (DBPs), which usually target membrane-associated ion channels, and the non-disulfide bridged peptides (NDBPs), a smaller group with multifunctional properties. In the past decade, these peptides have gained interest because most of them display functions that include antimicrobial, anticancer, haemolytic, and anti-inflammatory activities. Our current study focuses on the short (9-19 amino acids) antimicrobial linear scorpion peptides. Most of these peptides display a net positive charge of 1 o
