Potential chemo preventive and anti-inflammatory constituents from Centaurea pumilio: bio-guided isolation.

The natural alternatives offer cost-effective solutions compared to chemically synthesised drugs, with the added advantage of minimal side effects. Centaurea pumilio L., a rare and endangered species within the Centaurea genus belonging to the Asteraceae family, has received limited attention regarding its chemical composition. Aerial parts of C. pumilio were extracted with methanol (TE-1) and then fractionated into four fractions as follows (DCM-2, WF-3, MW-4 and MF-5). In a rigorous assessment of their anti-inflammatory potential, all samples underwent scrutiny utilising LPS-activated RAW264

Isolation and characterization of a novel glycoside from the inner bark of Sclerocarya birrea: insights into antibiotic and anticancer activity.

Phytochemical analysis of the dichloromethane-methanol (1:1) extract from Sclerocarya birrea inner bark led to the identification of three compounds: a novel glycoside, 3α-sorbithoxyglucose (1), and two known compounds, retusasterol (2) and β-sitosterol-3-O-D-glycoside (3), isolated for the first time from this plant. Structural elucidation via NMR confirmed their identities. Cytotoxicity studies against MCF7 breast cancer and DU145, PC3, LNCaP prostate cancer cell lines revealed that compounds 1 and 2 lacked inhibitory effects on cell proliferation, while retusasterol exhibited cytotoxicity w

Sterol Composition in the Lichens Lobaria pulmonaria and Lobaria retigera: Does Photobiont Matter?

The lipid composition of the mycobint and photobiont symbiotic partners of lichenized ascomycetes varies greatly. The aim of this study was to compare the profile of the major sterols in two closely related lichens from the genus Lobaria with different photobionts. The three-component lichen Lobaria pulmonaria has two photobionts. While the main photobiont is the chlorophycean alga Symbiochloris reticulata, this lichen contains small amounts of the cyanobacterium Nostoc. By contrast, the cyanobacterium Nostoc is the main photobiont in Lobaria retigera. Relatively loosely bound sterols were ext

Antioxidant and anti-inflammatory agents in chronic liver diseases: Molecular mechanisms and therapy.

Chronic liver disease (CLD) is a continuous process that causes a reduction of liver function lasting more than six months. CLD includes alcoholic liver disease (ALD), non-alcoholic fatty liver disease (NAFLD), chronic viral infection, and autoimmune hepatitis, which can lead to liver fibrosis, cirrhosis, and cancer. Liver inflammation and oxidative stress are commonly associated with the development and progression of CLD. Molecular signaling pathways such as AMP-activated protein kinase (AMPK), C-Jun N-terminal kinase, and peroxisome proliferator-activated receptors (PPARs) are implicated in

Oxidative Stress and Mitochondrial Complex I Dysfunction Correlate with Neurodegeneration in an α-Synucleinopathy Animal Model.

The α-synucleinopathies constitute a subset of neurodegenerative disorders, of which Parkinson's disease (PD) is the most common worldwide, characterized by the accumulation of misfolded α-synuclein in the cytoplasm of neurons, which spreads in a prion-like manner to anatomically interconnected brain areas. However, it is not clear how α-synucleinopathy triggers neurodegeneration. We recently developed a rat model through a single intranigral administration of the neurotoxic β-sitosterol β-D-glucoside (BSSG), which produces α-synucleinopathy. In this model, we aimed to evaluate the temporal pa

Effect of acute alterations in small bowel transit time upon the biliary excretion rate of bile acids.

The rate of biliary bile acid excretion is a product of bile acid pool size and enterohepatic cycling frequency. Cycling frequency, in turn, is determined by the time necessary for bile acid to travel from the ampulla of Vater to its small bowel absorptive site, from its absorptive site to the liver, and from the liver to the duodenum. In 10 subjects receiving constant intraduodenal infusion of an isocaloric formula containing the nonabsorbable markers polyethylene glycol and beta-sitosterol, the authors altered small intestinal transit time and measured the effect upon biliary bile acid excre

Oxidative Stability and Quality Deterioration of Ganoderma lucidum Spore Oil Under Accelerated Storage Conditions.

Prior studies on Ganoderma lucidum spore oil (GLSO) rarely systematically explored its storage oxidative deterioration mechanism via multi-marker combined characterization. This study innovatively integrated the Schaal oven accelerated storage test and dual-NMR detection (LF-NMR and 1H NMR) to evaluate the oxidative stability of GLSO over 60 d. Dynamic changes in routine oxidation indices, glycerol core aldehydes (GCAs), phytosterols and their triterpenoid biosynthetic precursors, fatty acids and volatile compounds were synchronously tracked. Results revealed complex fluctuating accumulation o
