Effect of acute stimulation on Fos expression in spinal neurons in the presence of persisting C-fiber activity.

Expression of the inducible transcription factor c-Fos has been examined in the lumbar spinal cord following noxious chemical stimulation (injection of 2% formalin) of the ankles or the ventral skin of the hindpaws of either normal rats, or monoarthritic rats during the chronic phase of the disease. In normal animals the basal expression of c-Fos was low. One day after induction of monoarthritis by an intra-articular injection of killed Mycobacterium butyricum (in complete Freund's adjuvant) there were numerous c-Fos labelled cells in the ipsilateral dorsal horn, and bilaterally in lamina VIII

Stimulation of mucin biosynthesis in rat gastric mucosa by FRG-8813 and its structural analogs.

Certain chemical properties, which may determine the stimulatory actions of the new histamine H2 receptor antagonist, FRG-8813 (2-(furfurylsulfinyl)-N-(4-[4-(piperidinomethyl)-2-pyridyl]o xy-(Z)- 2-butenyl)acetamide), on mucin biosynthesis, were identified by considering the derivation of this drug using an organ culture system of the rat stomach. [3H]Glucosamine and [35S]sulfate incorporation was stimulated in the corpus region by FRG-8813 and its structural analog, compound A (N-[4-[[4- (piperidinylmethyl)pyridyl]-2-oxy]-(Z)-2-butenyl]phthalimide). The chronotropic response to histamine in t

Regulation of melatonin production by catecholamines and adenosine in a photoreceptive pineal organ. An in vitro study in the pike and the trout.

The pineal organ of fish contains photoreceptor cells with structural and functional analogies to retinal photoreceptors. In these cells, the light/dark (LD) cycle influences the production of melatonin by controlling the activity of one of its synthetizing enzymes, serotonin N-acetyltransferase (NAT). The daily rhythm in NAT activity is generated endogenously in the pike but not in the trout pineal. We report here that in addition to the LD information, chemical factors are also involved in the control of melatonin production. Adenosine and two of its analogs stimulated or inhibited NAT activ

Regulation of ornithine decarboxylase activity by cyclic AMP in guinea pig lymphocytes: transcriptional and post-transcriptional control.

Induction of ornithine decarboxylase (L-ornithine carboxy-lyase, EC 4.1.1.17) in phytohemagglutinin-stimulated lymphocytes was suppressed by the administration of dibutyryl cyclic AMP and 3-isobutyl-1-methylxanthine. This inhibition of ornithine decarboxylase induction was overcome by removing the chemicals from the lymphocyte culture. However, the recovery of ornithine decarboxylase induction by washout of the chemicals did not occur when actinomycin D, cordycepin or alpha-amanitin was added to the cultures, suggesting that the synthesis of mRNA directed to ornithine decarboxylase was reduced

Estrogen-dependent effects of vagotomy and endogenous opioids on temporomandibular joint-responsive neurons in trigeminal subnucleus caudalis.

Temporomandibular joint (TMJ) disorders are common orofacial pain conditions influenced by multiple factors. This study examined how vagotomy and endogenous opioids affect TMJ-responsive neurons in the trigeminal subnucleus caudalis/cervical junction (Vc/C1-2) in female rats under different estrogen levels. Under low estrogen, cervical vagotomy enhanced TMJ unit responses to levels seen in high estrogen conditions but had no additional effect under high estrogen. Vagotomy did not change chemical stimulation (ATP) thresholds or spontaneous activity, suggesting a central neural mechanism. Respon

Intimacy of the neuroeffector junction and resistance to alpha-adrenoceptor-blockade of the neurogenic contractile response in vasa deferentia from guinea pig and rat.

The effect of phentolamine on the neurogenic contractile response in vasa deferentia from rat and guinea pig was studied during Wallerian degeneration. This response was also investigated after partial denervation (surgery or chemical sympathectomy by guanethidine treatment) in vasa deferentia from guinea pig. During Wallerian degeneration the response showed a gradual increase in sensitivity to phentolamine and was abolished in the late stages. The neurogenic contractile response of the partially denervated vas deferens was blocked by low concentrations of phentolamine. It is concluded that d

Compensatory increase of responses to nerve stimulation of the partially denervated rat urinary bladder.

The rat urinary bladder was deprived of half of its innervation by removing the pelvic ganglion on one side. The motor response of such a partially denervated bladder to stimulation of the pelvic nerve on the other side were examined 1 week, 1 month and 2 months postoperatively. On all three occasions the increase in pressure of the operated bladder was larger than that of the control bladder; the enlargement was most marked 2 months after operation. The responses were further enlarged by eserine, and markedly reduced by atropine. The present results combined with those of previous investigati

[Evoked potentials in the optic tectum of the carp brain upon electric stimulation of the facial nerve].

The evoked potentials in the carp tectum opticum in response to stimulation of the afferent pathway for chemical nonolfactory reception, the facial nerve, are positive deflections of 5-25 ms latency which do not reverse during microelectrode submersion. According to the evoked potential amplitude and latent period 7 active zones were distingnished in the ipsilateral tectum differing from each other either in one or in both parameters. The dependence of the response evoked by facial nerve stimulation in the tectum opticum on the impulsation from the medulla is proved by the difference in latenc

[Research progress on the pathogenesis of inflammatory external root resorption].

Inflammatory external root resorption (IERR) refers to the pathological process of dissolving the hard tissue on the outer surface of the tooth root by the body's own immune system under the stimulation of various physical and chemical factors such as infection, stress, trauma and orthodontic treatment. Severe IERR can lead to endodontic and periodontal diseases, and even the loss of teeth. Therefore, understanding the etiology and the pathogenic mechanism of IERR are of importance in its prevention and treatment. This article will review the etiology and the regulation mechanisms of IERR.

Enkephalin modulation of neural transmission in the cat stellate ganglion: pharmacological actions of exogenous opiates.

Neural ganglionic transmission was studied in vivo in the cat, using closed chest anesthetized preparations. The right stellate ganglion and its branches were exposed retropleurally and prepared for electrical stimulation of pre- and postganglionic nerve fibers. The axillary artery was cannulated allowing direct administration of drugs in the arterial blood supplying the ganglion. Stimulation of postjunctional receptors could thus be obtained by local administration of selective agents. Local administration of nicotinic, muscarinic or histaminergic agents increased heart rate and blood pressur

Ginger relieves intestinal hypersensitivity of diarrhea predominant irritable bowel syndrome by inhibiting proinflammatory reaction.

BACKGROUND: Ginger or ginger extracts have been used in traditional medicine relieve pain caused by diarrhea predominant irritable bowel syndrome (IBS-D), but few data exists about its effectiveness. This present study was to validate the effect of ginger on visceral pain, and to further explore the possible underlying mechanism by which ginger is used to relieve IBS-D intestinal hypersensitivity. METHODS: First, the IBS-D rat model was established by chemical stimulation and acute and chronic pressure stimulation. Then, different dose of ginger were administrated to IBS-D rats and evaluate t

Olfactory responses of the antennal trichoid sensilla to chemical repellents in the mosquito, Culex quinquefasciatus.

Insect repellents are widely used to protect against insect bites and thus prevent allergic reaction and the spread of disease. To gain insight into the mosquito's response to chemicals repellents, we investigated the interaction between the olfactory system of the mosquito Culex quinquefasciatus Say and chemical repellents using single sensillum recording. The interactions of 50 repellent chemicals with olfactory receptor neurons were measured in six different types of mosquito sensilla: long sharp trichoid (LST), short sharp trichoid (SST), short blunt trichoid I (SBT-I), short blunt trichoi

Clinical expression of endocrine disruptors in children.

PURPOSE OF REVIEW: Health status is the result of complex interaction between individual factors, general environmental factors and specific factors as nutrition or the presence of chemicals. Aim of this review is to point out the more recent knowledge covering the role of the endocrine disrupting chemical (EDC) on pediatric population wellbeing. RECENT FINDINGS: Prenatal, postnatal life and puberty are the three main temporal windows of susceptibility when EDCs may act. The mechanism is independent from dose or duration of exposition, sex, age or combination of chemicals and may also be tran

Development of a recombinant human ovarian (BG1) cell line containing estrogen receptor α and β for improved detection of estrogenic/antiestrogenic chemicals.

Estrogenic endocrine-disrupting chemicals are found in environmental and biological samples, commercial and consumer products, food, and numerous other sources. Given their ubiquitous nature and potential for adverse effects, a critical need exists for rapidly detecting these chemicals. The authors developed an estrogen-responsive recombinant human ovarian (BG1Luc4E2) cell line recently accepted by the US Environmental Protection Agency (USEPA) and Organisation for Economic Co-operation and Development (OECD) as a bioanalytical method to detect estrogen receptor (ER) agonists/antagonists. Optogenetic Manipulation of Neuronal Activity to Modulate Behavior in Freely Moving Mice.

Optogenetic modulation of neuronal circuits in freely moving mice affects acute and long-term behavior. This method is able to perform manipulations of single neurons and region-specific transmitter release, up to whole neuronal circuitries in the central nervous system, and allows the direct measurement of behavioral outcomes. Neurons express optogenetic tools via an injection of viral vectors carrying the DNA of choice, such as Channelrhodopsin2 (ChR2). Light is brought into specific brain regions via chronic optical implants that terminate directly above the target region. After two weeks o

Methamphetamine plus scopolamine potentiates behavioral sensitization and conditioning.

The effects of repeated methamphetamine (4.0 mg/kg) plus scopolamine (0.5 mg/kg) treatment on behavioral sensitization and drug conditioning in rats were compared with the effects of repeated methamphetamine treatment. Behavioral sensitization induced by repeated methamphetamine plus scopolamine treatment was more vigorous than that induced by repeated methamphetamine treatment. Repeated methamphetamine plus scopolamine treatment produced sensitized responses, not only to methamphetamine plus scopolamine and methamphetamine but also, to a lesser extent, to scopolamine. Novel psychoactive substances (designer drugs): overview and pharmacology of modulators of monoamine signaling.

Novel psychoactive substances are newly used designer drugs ("internet drugs", "research chemicals", "legal highs") potentially posing similar health risks to classic illicit substances. Chemically, many novel psychoactive substances can be classified as phenethylamines, amphetamines, synthetic cathinones, piperazines, pipradrols/piperidines, aminoindanes benzofurans, and tryptamines. Pharmacologically, these substances interact with various monoaminergic targets. Typically, stimulants inhibit the transport of dopamine and noradrenaline (pipradrols, pyrovalerone cathinones) or induce the relea

Disruption of thyroid hormone functions by low dose exposure of tributyltin: an in vitro and in vivo approach.

Triorganotins, such as tributyltin chloride (TBTCl), are environmental contaminants that are commonly found in the antifouling paints used in ships and other vessels. The importance of TBTCl as an endocrine-disrupting chemical (EDC) in different animal models is well known; however, its adverse effects on the thyroid gland are less understood. Hence, in the present study, we aimed to evaluate the thyroid-disrupting effects of this chemical using both in vitro and in vivo approaches. We used HepG2 hepatocarcinoma cells for the in vitro studies, as they are a thyroid hormone receptor (TR)-positi

A comparison of statistical approaches to the derivation of EC3 values from local lymph node assay dose responses.

Effective risk assessment and management of allergic contact dermatitis require three key factors: adequate hazard identification, measurement of the relative potency of identified hazards and an understanding of the nature, extent and duration of exposure. Suitable methods for hazard identification, such as the murine local lymph node assay (LLNA) and the guinea-pig maximization test, are well established and conditions of human exposure normally can be well anticipated. Thus, the need is for a robust and quantitative method for the estimation of relative skin sensitizing potency. One possibl
