Drp1-dependent mitochondrial fission in cardiovascular disease.

Mitochondria are highly dynamic organelles undergoing cycles of fusion and fission to modulate their morphology, distribution, and function, which are referred as 'mitochondrial dynamics'. Dynamin-related protein 1 (Drp1) is known as the major pro-fission protein whose activity is tightly regulated to clear the damaged mitochondria via mitophagy, ensuring a strict control over the intricate process of cellular and organ dynamics in heart. Various posttranslational modifications (PTMs) of Drp1 have been identified including phosphorylation, SUMOylation, palmitoylation, ubiquitination, S-nitrosy…

Post-translational modifications of immune checkpoints: molecular mechanisms, tumor microenvironment remodeling, and therapeutic implications.

Immune checkpoints play pivotal roles in regulating immune responses and maintaining tolerance. In cancer, these molecules are hijacked to suppress antitumor immunity, resulting in therapeutic resistance to immune checkpoint blockade (ICB). Recent advances have highlighted the critical role of post-translational modifications (PTMs), including phosphorylation, ubiquitination, glycosylation, palmitoylation, UFMylation, acetylation, SUMOylation, methylation, and ISGylation, in modulating checkpoint stability, trafficking, and function across diverse immune and tumor cell types. These dynamic PTM…

C4orf3 Regulates HIF-1α Degradation Under Hypoxic Conditions and Contributes to the Malignant Phenotype in Small Cell Lung Cancer.

Hypoxia is a critical feature of the tumour microenvironment in small cell lung cancer (SCLC) and contributes to malignant progression throu … athway. Further analysis demonstrated that C4orf3 modulates HIF-1α stability through PIASy-mediated SUMOylation. Clinical relevance was supported by a positive association between C4orf3 and HIF-1α expression in resected SCLC tissues. These findings suggested that C4orf3 functions as a regulator of hypoxic adaptation in SCLC by maintaining HIF-1α stability an …

SIRT3 regulates PRDX3 acetylation to support mitochondrial peroxide detoxification and limit oxidative stress-associated ferroptosis vulnerability.

Oxidative stress disrupts mitochondrial redox homeostasis and contributes to ferroptosis-associated vulnerability, yet the molecular link between impaired mitochondrial peroxide detoxification and ferroptosis-associated vulnerability remains incompletely defined. Here, we identify PRDX3 as a candidate SIRT3-regulated effector of mitochondrial peroxide control. In AML12 cells, oxidative stress reduced mitochondrial SENP1, increased SIRT3 SUMOylation and elevated mitochondrial protein acetylation. Mitochondrial acetylome profiling identified PRDX3 K92 as a SIRT3-responsive acetylation site. Gene…

Decoding the PTM code of cGAS-STING in gastric cancer: from innate DNA sensing to precision combination therapy.

The cGAS-STING pathway represents a crucial component of innate immune responses and has been increasingly recognized as a pivotal regulatory mechanism governing gastric cancer progression and therapeutic resistance. This comprehensive review synthesizes current knowledge regarding the mechanisms through which Post-Translational Modifications (PTMs) modulate the activity, stability, and subcellular localization of cGAS and STING proteins, encompassing phosphorylation, ubiquitination, acetylation, SUMOylation, and palmitoylation. These PTMs function as pivotal regulators that maintain the delic…

High expression of SENP1 correlates with poor prognosis and immune evasion in gastric cancer.

Gastric cancer (GC) remains a highly prevalent malignancy with poor prognosis in advanced stages. SUMOylation, a key post-translational modification, regulates tumor progression and immune responses, yet the role of SUMO-specific protease 1 (SENP1) in GC remains unclear. Here, we show that SENP1 is significantly overexpressed in GC tissues and cell lines, as demonstrated by analyses of TCGA/GEO datasets and by immunohistochemistry, western blotting, and qRT-PCR. Higher SENP1 levels were associated with more advanced disease, undifferentiated histology, systemic dissemination, and worse surviva…

SENP2 alleviates CCl4-induced liver fibrosis by promoting activated hepatic stellate cell apoptosis and reversion.

SUMOylation and deSUMOylation, a dynamic process, is proved to be involved in various fibrotic diseases. Here, we found SENP2, one of deSUMOylation protease family member, was decreased in CCl4-induced mice fibrotic liver tissues, primary HSCs and restored after spontaneously recovery. In addition, HSC-T6 cells with TGF-β1 treatment resulted in a significant reduction of SENP2. Ectopic expression of SENP2 hindered cells activation and proliferation induced by TGF-β1 while knockdown of SENP2 showed an opposite effect. Importantly, SENP2 promoted apoptosis of HSC-T6 cells activated by TGF-β1. Fu…

Post-Translational Modification Crosstalk in Gastric Cancer: Molecular Switches, Signaling Networks, and Therapeutic Opportunities, with Comparative Insights from Colorectal Cancer.

Gastric cancer remains a leading cause of cancer-related death, and its malignant behavior is driven in large part by dysregulated post-translational modifications (PTMs). Yet the prevailing tendency to study individual modifications in isolation has obscured a defining feature of PTM biology: modifications frequently interact, competing for the same residue or cooperating across residues to form regulatory switches that dictate protein fate. This review synthesizes the PTM crosstalk framework in gastric cancer around three principal axes-phosphorylation-ubiquitination relays, SUMOylation-ubiq…

Development of the first-in-class RNF4 PROTAC degrader as potential therapeutics for hepatocellular carcinoma.

Ring Finger Protein 4 (RNF4) recognizes poly-SUMOylated proteins via its SUMO-Interacting Motifs (SIMs) and subsequently ubiquitinates them, thus effecting some key regulatory proteins involved in cancer development and progression. Our previous study found RNF4 was a potential target for HCC interruption. However, none of its inhibitor has been developed so far. Targeting RNF4 for degradation, rather than mere binding, emerged as a promising alternative strategy. To this end, we designed, synthesized and evaluated 28 PROTACs targeting RNF4 based on a reported covalent binder. Among them, RD12

ATM, KAP1 and the Epstein-Barr virus polymerase processivity factor direct traffic at the intersection of transcription and replication.

The timing of transcription and replication must be carefully regulated for heavily-transcribed genomes of double-stranded DNA viruses: transcription of immediate early/early genes must decline as replication ramps up from the same genome-ensuring efficient and timely replication of viral genomes followed by their packaging by structural proteins. To understand how the prototypic DNA virus Epstein-Barr virus tackles the logistical challenge of switching from transcription to DNA replication, we examined the proteome at viral replication forks. Specifically, to transition from transcription, th

Point-of-Care Peptide Hormone Production Enabled by Cell-Free Protein Synthesis.

In resource-limited settings, it can be difficult to safely deliver sensitive biologic medicines to patients due to cold chain and infrastructure constraints. Point-of-care drug manufacturing could circumvent these challenges since medicines could be produced locally and used on-demand. Toward this vision, we combine cell-free protein synthesis (CFPS) and a 2-in-1 affinity purification and enzymatic cleavage scheme to develop a platform for point-of-care drug manufacturing. As a model, we use this platform to synthesize a panel of peptide hormones, an important class of medications that can be

CBX4 stabilizes NEUROD1 through SUMO-ubiquitin crosstalk to delay β-cell senescence.

Aging-associated β-cell senescence is a key driver of type 2 diabetes mellitus (T2DM). However, its associated molecular mechanisms remain p … rectly with the transcription factor NEUROD1 via its SUMO-interacting motif 1 (SIM1). CBX4 promotes SUMOylation of NEUROD1 at lysine 38 and drives its assembly into nuclear condensates via phase separation. These processes collectively antagonize NEDD4-mediated ubiquitination at the same site, stabilizing NEUROD1. The preserved NEUROD1 enhances Ins2 transcrip …
