Space exploration exposes astronauts to a variety of gravitational stresses. Exposure to a reduced gravity environment affects human anatomy and physiology. Countermeasures to restore homeostatic states within the human body have begun. The pathophysiological effects of exposure to microgravity, on the neurological system, are, however, still not clear. NASA has scheduled deep space exploration of extraterrestrial locations such as the Moon and Mars in the 2030s. Adverse health effects related to the human exposure to microgravity from previous, relatively shorter missions have been documented

One of the most important alterations that occur in man and experimental animals during spaceflight concerns the skeletal system, and entails important bone loss and degradation of mechanical properties. In the present work we investigate ex vivo the long-term effects of weightlessness (simulated microgravity) on bone tissue, by comparing the mesoscale structural properties of weight-bearing rat tibial epiphyseal cancellous structures of healthy animals (ground controls) with those of identical bone explants maintained ex vivo in the Rotary Cell Culture System (RCCS) bioreactor, used to model,…

High-molecular polymers apt to directly influence flow microstructure were tested as a fundamentally new method for correcting microhemodynamics in microgravity. Pressure in the mesenteric arterial microvessels was measured two weeks in rats adapted to the head-down suspension. Intravenous polyethylene oxide (Polyox WSR-301, end-concentration in the order of 2.10(-7) g/ml), reduced the microvascular pressure by 26%, whereas in the control pressure was reduced by only 15%. Systemic arterial pressure showed an equal drop in the groups (by 10 to 11%). These results suggest that the biomechanical

The extent to which the slow phase velocity (SPV) of nystagmus elicited by a vertical optokinetic stimulation with constant velocity could be modulated by sinusoidal angular motion in the vertical plane was investigated under normal gravity condition and during the microgravity period of parabolic flight. In normal gravity, when the angular head motion and the optokinetic stimulation were in the same direction, the peak SPV was slower than the velocity of the optokinetic display. When the head motion and the optokinetic stimulation were in opposite directions, the peak SPV was equal to the vel

The purpose of this study was to examine the effect of 30 d of 6 degrees headdown bedrest (BR) on the in vivo strength of skeletal muscle. Peak angle specific (0.78 rad below horizontal) torque of the knee extensor (KE) and flexor (KF) muscle groups of both limbs was assessed during unilateral efforts at four speeds (0.52, 1.74, 2.97 and 4.19 rad.s-1) during concentric and at three speeds (0.52, 1.74 and 2.97 rad.s-1) during eccentric actions. The average decrease (p less than 0.05) of peak angle specific torque directly post-BR for the KE across speeds of concentric and eccentric actions was

The microgravity environment of spaceflight produces rapid cardiovascular changes which are adaptive and appropriate in that setting, but are associated with significant deconditioning and orthostatic hypotension on return to Earth's gravity. The rapidity with which these space flight induced changes appear and disappear provides an ideal model for studying the underlying pathophysiological mechanisms of deconditioning and orthostatic hypotension, regardless of etiology. Since significant deconditioning is seen after flights of very short duration, muscle atrophy due to inactivity plays, at mo

Long-duration spaceflight presents unique physiological and nutritional challenges due to factors such as microgravity, radiation, confinement, and circadian rhythm disruption. These stressors reshape energy metabolism, bone and muscle homeostasis, immune responses, and gut microbiota composition, leading to impaired nutrient absorption and oxidative imbalance. Conventional Earth-based dietary models are inadequate to address such complex interactions, highlighting the need for precise nutritional strategies combined with sustainable in-situ food production systems. This review examines recent

Recent findings from studies involving astronauts and animal models indicate that microgravity increases immune cell activity and potentially alters the white and gray matter of the central nervous system (CNS). To further investigate the impact of microgravity on CNS cells, we established cultures of three-dimensional neural organoids containing isogenic microglia, the brain's resident immune cells, and sent them onboard the International Space Station. When using induced pluripotent stem cell (iPSC) lines from individuals affected by neuroinflammatory and neurodegenerative diseases such as m

OBJECTIVE: The purpose of this study is to gain a better understanding of the impact of microgravity on antibiotic resistance.
METHODS: K. pneumoniae original (KPO) strain was cultured under either simulated microgravity (SMG) conditions with background antibiotic exposure (SMGA) for the experimental strain or a normal gravity condition with background antibiotic exposure (NGA) for the control strain. The K. pneumoniae original (KPO) strain was also cultured under normal gravity (NG) as an additional control. Antibiotic susceptibility was evaluated prior to their incubation under SMGA, NGA, o

Interest in space habitation has grown dramatically with planning underway for the first human transit to Mars. Despite a robust history of domestic and international spaceflight research, understanding behavioral adaptation to the space environment for extended durations is scant. Here we report the first detailed behavioral analysis of mice flown in the NASA Rodent Habitat on the International Space Station (ISS). Following 4-day transit from Earth to ISS, video images were acquired on orbit from 16- and 32-week-old female mice. Spaceflown mice engaged in a full range of species-typical beha

Humans undergo several consistent and measurable changes of fluid distribution and regulation in the course of adapting to microgravity. Recently, a syndrome of objective findings has been described by Mader et al. associated with long-duration missions, including hyperopic shifts, mildly elevated intracranial pressure, papilledema, globe flattening, choroidal folds, and other anatomic findings. Experience with venous obstructive lesions leads the author to propose a primary obstructive process, unique to or exacerbated by microgravity, acting at the level of the proximal internal jugular vein

A variety of physiological changes are experienced by astronauts during both short- and long-duration space missions. These include space motion sickness, spatial disorientation, orthostatic hypotension, muscle atrophy, bone demineralization, increased cancer risk, and a compromised immune system. This review focuses on countermeasures used to moderate these changes, particularly exercise devices that have been used by National Aeronautics and Space Administration astronauts over the past six decades as countermeasures to muscle atrophy and bone loss. The use of these devices clearly has shown

Commercial suborbital flights, which include 3-5 minutes of 0 g between hyper-g launch and landing phases, will present suborbital passengers with a challenging sensorimotor experience. Based on the results of neurovestibular research in parabolic and orbital flight, and the anticipated wide range of fitness and experience levels of suborbital passengers, neurovestibular disturbances are likely to be problematic in this environment. Pre-flight adaptation protocols might alleviate some of these issues. Therefore, we describe a set of sensorimotor tests to evaluate passengers before suborbital f

We need to find novel ways of increasing exercise participation, particularly in those populations who find it difficult to participate in voluntary exercise. In recent years researchers have started to investigate the potential for using electrical stimulation to artificially stimulate a pattern of muscle activity that would induce a physiological response consistent with cardiovascular exercise. Work to date has indicated that this is best achieved by using a stimulation protocol that results in rapid rhythmical isometric contractions of the large leg muscle groups at sub tetanic frequencies

The review deals with the analysis of osteodensitometry data from the cosmonauts flown on Russian space station MIR and the International space station and suppositions about involvement of different levels of metabolism regulation in bone loss triggered by insufficient mechanic loading in microgravity attendant by redistribution of body liquids. It is surmised that the initial reactions are associated with the biomechanical factor and recruitment of local mechanisms, i.e. osteocyte osteolysis and inhibition of osteoblast histogenesis. Regulation on the level of tissues and organs is responsib

Autonomic neural functions are important to regulate vital functions in the living body. There are different methods to evaluate indirectly and directly autonomic, sympathetic and parasympathetic, neural functions of human body. Among various methods, microneurography is a technique to evaluate directly sympathetic neural functions in humans. Using this technique sympathetic neural traffic leading to skeletal muscles (muscle sympathetic nerve activity; MSNA) can be recorded from human peripheral nerves in situ. MSNA plays essentially important roles to maintain blood pressure homeostasis again

Echocardiographic measurements of astronaut cardiac function have documented an initial increase, followed by a progressive reduction in both left ventricular end-diastolic volume index and stroke volume with entry into microgravity (micro-G). The investigators hypothesize that the observed reduction in cardiac filling may, in part, be due to the absence of a gravitational acceleration dependent, intraventricular hydrostatic pressure difference in micro-G that exists in the ventricle in normal gravity (1-G) due to its size and anatomic orientation. This acceleration-dependent pressure differen

Main osteoporosis definitions and some results of bone tissue research in Russian astronauts, patients, and healthy subjects, using modern osteodensitometry, are presented. Bone mineral density (BMD) was regularly decreased at lower segments of skeleton. In the skull bone and some other sites of upper part of skeleton, a tendency was revealed for an increase of the bone mineral content (BMC). The mean value of bone loss was within the normal range and not correlated with duration of space flight; it revealed a high individual variability and in some cases was clinically qualified as local oste
