Experiences in Rhodesia with various recovery techniques available for the laboratory diagnosis of infections with Dipetalonema perstans and Wuchereria bancrofti are discussed. A diagnostic laboratory regimen for routine filarial investigations is suggested. Included are preliminary observations on the use of mebendazole (Vermox) for the treatment of D. perstans infections.

The authors compare the serological results obtained before and after a single dose with diethylcarbamazine (6 mg./kg.) in a homogeneous and strongly infected with Wuchereria bancrofti var, pacifica population. They point out a statistically significant decrease of the hemagglutining antibody titres of filarian patients.

Brugia malayi and B. Pahangi microfilariae from gerbil intraperitoneal infections were inoculated into the thorax of male and female Toxorhynchites amboinesis and developed into third-stage larvae as early as 11 days. In a comparative study with Aedes togoi fed on microfilaremic gerbils, third-stage larvae were found at 10 days. Some third-stage larvae of B. malayi inoculated into gerbils developed to advanced stages. Third-stage larvae of Wuchereria bancrofti were recovered in low numbers from Tx. amboinesis and Tx. aurifluus inoculated with microfilariae recovered from human blood by membran…

In a transversal study on a sample of 386 children and adolescents from an outpatient clinic for filariasis in Recife, Northeast Brazil, the frequency of anti-Toxocara antibodies and its relation to age, gender, number of peripheral eosinophils, Wuchereria bancrofti microfilariae and intestinal helminths was determined. The total anti-Toxocara IgG antibody frequency was 39.4%, by ELISA technique. The difference in frequency between males (40.1%) and females (37.6%) was not statistically significant. The 6 to 10-year-old subset presented the highest frequency of anti-Toxocara antibodies (60%), …

The nematodes Wuchereria bancrofti and Brugia malayi; the principal etiologic agents of lymphatic filariasis are mosquito dependant in the biological transmission. Dirofilariasis is essentially a disease of canines which can also be transmitted to humans by culicine mosquitoes. The present work studied the histological and PCR xenomonitoring filarial infestation in culicine mosquitoes in Sohag Governorate. One hundred female mosquitoes of the 5 culicine spesies present in the selected localities, were dissected and histological sections of thoracic muscles were examined for filarial larvae. Al…

This paper describes a case of filarial pleural effusion, the fifth such to be reported. Microfilariae of Wuchereria bancrofti were detected in the pleural fluid on cytological examination. There was a prompt and complete response to treatment with diethylcarbamazine. There were, however, no symptoms and signs of tropical pulmonary eosinophilia nor any peripheral eosinophilia.

Eight patients from Uganda, Sudan, Nigeria, and Zaire presented with swelling of the eyelids, proptosis, or conjunctival granulomas. In 5 patients the cause was Mansonella perstans; in 1, it was a Wuchereria bancrofti-like worm; and in 2, an unidentifiable worm. The morphologic features and histopathologic changes in the conjunctiva and periorbital fat are described and illustrated.

A survey for lymphatic filariasis was conducted among tea garden workers of central Assam. Of the 656 night blood samples examined, 31 were found positive for Wuchereria bancrofti parasite (microfilaria rate 4.7%). Microfilaria rate was higher in male (7.3%) than females (2.1%). Culex quinquefasciatus was incriminated as vector mosquito.

A 71-year-old Australian-born man with previous extended travel to the Philippines presented with bilateral lymphoedema, fevers and rigors. Examination of a nocturnal blood film revealed microfilariae of Wuchereria bancrofti, confirming a diagnosis of Bancroftian filariasis. This case highlights the challenges of diagnosing and managing lymphatic filariasis in non-endemic regions.

The observation that children born to mothers that are infected with Wuchereria bancrofti ore more susceptible to filarial infection than those born to uninfected mothers, raises many questions, particularly regarding immune mechanisms. In this article, Allen Hightower, Patrick Lommie and Mark Eberhard discuss these issues and their implications for the epidemiology of filarial infection.

Macrofilariae have been recognized for many millennia. Microfilariae were, however, not demonstrable until microscopy attained an advanced degree of perfection. Demonstration of the mode of transmission of the various filariases (Wuchereria bancrofti, Onchocerca volvulus, and Loa loa), dominated by Manson's work on lymphatic filariasis, constitutes one of the most exciting phases inhuman parasitology.

The utility of the IgG fraction of human filarial serum immunoglobulin in detecting circulating antigen by sandwich enzyme linked immunosorbent assay (ELISA) was studied. 27 of 33 sera from persons with microfilaraemia, 19 of 30 sera from clinical cases of filariasis, 4 of 30 sera from normal persons from a region endemic for filariasis showed the presence of circulating filarial antigen. All the 20 normal sera from the area where filariasis was not endemic gave negative reaction for filarial antigen. Those sera from persons with microfilaraemia that showed the presence of circulating antigen

Indirect fluorescent antibody tests (IFAT) using Wuchereria bancrofti infective larvae as antigen had the highest positivity rates in detecting Malayan and Bancroftian filariasis as compared to IFAT using antigens prepared from 5 other animal filarial species, Brugia pahangi, Dirofilaria immitis, Dipetalonema viteae, Litomosoides carinii and Onchocerca gutturosa. This study also recommends the use of human filarioids as the source of antigen in serological tests. However, before B. malayi and especially W. bancrofti can be easily available from the common laboratory animals. B. pahangi seems t…

We demonstrate by an ELISA the presence of antibodies in human filarial sera that react with diethylcarbamazine (DEC); they appear to be primarily filarial antibodies cross-reacting with DEC skeleton, since affinity-purified DEC antibodies strongly react with Wuchereria bancrofti microfilariae. These observations indicate a possible antigenic mimicry between the drug and some parasite component.

BACKGROUND: Filariasis and leishmaniasis are two neglected tropical diseases in Mali. Due to distribution and associated clinical features, both diseases are of concern to public health. The goal of this study was to determine the prevalence of co-infection with filarial (Wuchereria bancrofti and Mansonella perstans) and Leishmania major parasites in two ecologically distinct areas of Mali, the Kolokani district (villages of Tieneguebougou and Bougoudiana) in North Sudan Savanna area, and the district of Kolondieba (village of Boundioba) in the South Sudan Savanna area.
METHODS: The prevalenc…

Lymphatic filariasis, caused by Wuchereria bancrofti, remains a global health challenge. The enzyme Wuchereria bancrofti dihydrofolate reductase (WbDHFR) is a potential therapeutic target due to DHFR's critical role in folate metabolism and DNA synthesis. In this study, we employed a virtual screening workflow to repurpose antifolate compounds as WbDHFR inhibitors. Using structural data from the Protein Data Bank, we constructed a library of 194 antifolates and docked them to the WbDHFR folate binding site. Compounds methotrexate, TSD001, TSD10, and TSD25, with docking scores ranging from -9 t…

Lymphatic filariasis is a neglected parasitic disease that affects millions in tropical and subtropical countries and is caused by Wuchereria and Brugia species. Specific and sensitive detection methods are essential in mapping infected areas where rapid tests are needed to cover underdeveloped and remote regions, which facilitates eliminating the disease as a public health problem. A few commercialized rapid tests based on antigen or antibody detection are available, but the former only detects infection by Wuchereria species and cross-reacts with nonlymphatic filaria, whereas antibody detect…
