Kinetic study on the inhibition of xanthine oxidase by extracts from two selected Algerian plants traditionally used for the treatment of inflammatory diseases.

In order to further understand and assess the validity of herbal medicine, we investigated the potential inhibitory effect of various extracts from Fraxinus angustifolia and Pistacia lentiscus, two plants used traditionally in Algeria against several inflammatory diseases such as rheumatism, arthritis, and gout, on purified bovine milk xanthine oxidase (XO) activity. The total phenolic contents of the leaves and bark of F. angustifolia and the leaves and seeds of P. lentiscus were estimated. P. lentiscus aqueous fractions from hexane and chloroform extractions and F. angustifolia aqueous fract…

KMUP-1, a xanthine derivative, induces relaxation of guinea-pig isolated trachea: the role of the epithelium, cyclic nucleotides and K+ channels.

7-[2-[4-(2-chlorophenyl)piperazinyl]ethyl]-1,3-dimethylxanthine (KMUP-1) produces tracheal relaxation, intracellular accumulation of cyclic nucleotides, inhibition of phosphodiesterases (PDEs) and activation of K+ channels. KMUP-1 (0.01-100 microm) induced concentration-dependent relaxation responses in guinea-pig epithelium-intact trachea precontracted with carbachol. Relaxation responses were also elicited by the PDE inhibitors theophylline, 3-isobutyl-1-methylxanthine (IBMX), milrinone, rolipram and zaprinast (100 microm), and a KATP channel opener, levcromakalim. Tracheal relaxation induce…

Allopurinol, an inhibitor of xanthine oxidase, reduces uric acid levels and modifies the signs associated with copper deficiency in rats fed fructose.

This study was designed to focus on the potential stress that xanthine oxidase could produce in copper-deficient rats fed fructose. Fructose consumption results in an excess production of uric acid due to an increased degradation of nucleotides. The enzyme xanthine oxidase catalyzes the oxidation of both hypoxanthine and xanthine. During the oxidation process free radicals are generated, which in turn, induce lipid peroxidation and premature death. Allopurinol -- a competitive inhibitor of xanthine oxidase -- could alleviate the combined effects of fructose feeding and copper deficiency. Twent…

Hereditary xanthinuria in 2 Pakistani sisters: asymptomatic in one with beta-thalassemia but causing xanthine stone, obstructive uropathy and hypertension in the other.

We describe a 3-year-old Pakistani girl who presented with recurrent urinary infections. She had a nonfunctioning hydronephrotic right kidney and hypertension. At operation a calculus was impacted in the right ureter with dilatation of the pelviocaliceal system. Nephrectomy was performed. Histology revealed end stage pyelonephritis. The calculus consisted of pure xanthine. Further investigations demonstrated low serum uric acid and absent urinary uric acid with increased excretion of xanthine. Eight months after nephrectomy blood pressure had decreased to normal. Her 5-year-old sister, who has…

Methylxanthine and non-xanthine phosphodiesterase inhibitors. Their effects on adenosine uptake and the low Km cyclic AMP phosphodiesterase in intact rat adipocyte.

The effects of methylxanthines and non-xanthine phosphodiesterase-inhibitors on the low Km cyclic AMP phosphodiesterase of intact rat adipocytes were studied. Methylxanthines and papaverine stimulated rather than inhibited the enzyme when intact adipocytes were incubated in their presence. The effect of papaverine was not abolished by adenosine deaminase and was enhanced by adenosine. On the other hand, the effect of xanthine inhibitors and adenosine do not enhance each other. The difference in behaviour of these inhibitors could not be explained by their effects on adenosine uptake at the con…

Plasma hypoxanthine and xanthine levels in the early newborn period in problem-free preterm babies and those with idiopathic respiratory distress syndrome.

The use of hypoxanthine measurements for quantitative monitoring of intrauterine asphyxia is generally accepted. A high level in blood or in CSF is a consequence of tissue hypoxia. Hypoxanthine and xanthine were measured by selective high pressure liquid chromatography in mature newborns, in healthy, symptom-free preterm babies, and in preterm babies affected by idiopathic respiratory distress syndrome. The measurements were carried out from peripheral venous blood within three hours after birth and at the age of 48-72 hours. In mature newborns the mean hypoxanthine level was 11.10 mumol/l in …

Purine Metabolism Profiling Reveals Distinct Associations of Uric Acid, Xanthine, and Hypoxanthine with Neurological Severity and Functional Outcome After Acute Ischemic Stroke.

Purine metabolism and oxidative stress are increasingly recognized as important contributors to ischemic brain injury following acute ischemic stroke (AIS). Although uric acid has been extensively investigated because of its dual antioxidant and pro-oxidant properties, comparatively limited data exist regarding the prognostic relevance of upstream purine metabolites such as xanthine and hypoxanthine. The present study aimed to evaluate the association between serum uric acid, xanthine, and hypoxanthine concentrations and neurological severity and functional outcome following AIS. We performed …

Carbothioamide functionalized paroxetine analogues as antiparasitic agents targeting the hypoxanthine-guanine-xanthine phosphoribosyltransferase enzyme: Synthesis, characterization, biochemical, biophysical, and in silico studies.

Hypoxanthine guanine xanthine phosphoribosyl transferase (HGXPRT) is one of the central enzymes of purine salvage pathway of protozoan parasites, including Plasmodium. These parasites are purine auxotrophs, and rely on purine salvage pathway for nucleic acid synthesis. Inhibiting the enzymatic activity can effectively reduce the parasite- induced infections, including malaria. In this context, a library of new carbothioamide derivatives 1-17 of paroxetine was synthesized via single-step chemical transformation strategy with a series of substituted phenyl isothiocyanates. All compounds were pur…

Hypoxanthine, xanthine, and uric acid concentrations in plasma, cerebrospinal fluid, vitreous humor, and urine in piglets subjected to intermittent versus continuous hypoxemia.

Infants with sudden infant death syndrome have higher hypoxanthine (Hx) concentrations in their vitreous humor than infants with respiratory distress syndrome and other infant control populations. However, previous research on piglets and pigs applying continuous hypoxemia has not been able to reproduce the concentrations observed in infants with sudden infant death syndrome. To test whether intermittent hypoxemia could, in part, explain this observed difference, Hx, xanthine (X), and uric acid were measured in vitreous humor, urine, plasma, and cerebrospinal fluid in newborn piglets during in…

Xanthine Oxidase Inhibitory Peptides from Sesame 11S Globulin Target the FAD Domain: Kinetics, Molecular Dynamics, and Validation in a Zebrafish Hyperuricemia Model.

Food-derived bioactive peptides are promising alternatives to conventional xanthine oxidase (XO) inhibitors, yet individual XO inhibitory peptides from sesame protein remain uncharacterized. Here, sesame and peanut protein isolates were screened with four proteases; the trypsin hydrolyzate of sesame (TSH, 120 min) showed the highest XO inhibition (47.16%). Eight 11S globulin peptides were identified by nanoUHPLC-ESI-Q-TOF MS/MS. QGDIVAIPSGAAHW (IC50 = 467.2 μM) and AFYLAGGVPR (IC50 = 536.1 μM) exhibited mixed-type inhibition (α > 1). Molecular docking against two XO crystal structures (3NVY, 3…

Design, Synthesis, and Biological Evaluation of Triazole-Linked Lignan-Monoterpenoid-Based Hybrid Molecules as Xanthine Oxidase Inhibitors with Potent In Vivo Efficacy.

A novel series of triazole-tethered monoterpenoid-lignan hybrid molecules has been designed to target xanthine oxidase (XO), the enzyme responsible for hyperuricemia when it is up-regulated, resulting in gout and other metabolic disorders. Designed molecules were synthesized and initially evaluated for their XO inhibitory potential, and MT7 was most active (XO: IC50 = 0.263 ± 0.06 μM) with radical scavenging efficacy. MT7 showed higher cytotoxic potential against XO harboring cancer cells (MBDA-MB-231 breast cancer cells) than non-XO-harboring cells (A547 skin cancer cells), confirming intrace…

Similar gout flare incidence rates when using once- or twice-daily 0.5 mg colchicine prophylaxis after the start of xanthine oxidase inhibitors.

OBJECTIVE: To investigate the potential superiority of colchicine prophylaxis twice daily over once daily in preventing gout flares in the first 6 months after initiating xanthine oxidase inhibitors (XOIs). METHOD: A retrospective cohort study was conducted in three rheumatology centres in the Netherlands. Gout flare incidence densities were calculated for patients using colchicine once or twice daily in the first 6 months after starting XOIs. The incidence rate ratios (IRRs) between groups were analysed using Poisson regression, adjusted for confounders. The proportion of patients reaching a…

A self-designed Chinese herbal formula alleviates GAstV infection-induced gout in goslings by inhibiting xanthine oxidase (XOD) and regulating intestinal microbiota.

Goose gout, a metabolic disorder characterized by systemic urate deposition in visceral organs (e.g., liver, kidneys) and joints of goslings, is strongly associated with goose astrovirus (GAstV) infection and environmental stressors such as high humidity. This disease severely compromises growth performance and survival rates, yet no licensed prophylactic or therapeuti interventions are currently available. Here, we investigated the efficacy of SIA, a Chinese herbal formula comprising Saposhnikovia divaricata, Isatis indigotica and Astragalus membranaceus against GAstV-induced gout in goslings

Xanthine oxidoreductase-mediated injury is amplified by upregulated AMP deaminase in type 2 diabetic rat hearts under the condition of pressure overload.

BACKGROUND: We previously reported that upregulated AMP deaminase (AMPD) contributes to diastolic ventricular dysfunction via depletion of the adenine nucleotide pool in a rat model of type 2 diabetes (T2DM), Otsuka Long-Evans-Tokushima Fatty rats (OLETF). Meanwhile, AMPD promotes the formation of substrates of xanthine oxidoreductase (XOR), which produces ROS as a byproduct. Here, we tested the hypothesis that a functional link between upregulated AMPD and XOR is involved in ventricular dysfunction in T2DM rats. METHODS AND RESULTS: Pressure-volume loop analysis revealed that pressure overlo…

Pharmacophore modeling, molecular docking and molecular dynamics studies on natural products database to discover novel skeleton as non-purine xanthine oxidase inhibitors.

Gout is a common inflammatory arthritis caused by the deposition of urate crystals within joints. It is increasingly in prevalence during the past few decades as shown by the epidemiological survey results. Xanthine oxidase (XO) is a key enzyme to transfer hypoxanthine and xanthine to uric acid, whose overproduction leads to gout. Therefore, inhibiting the activity of xanthine oxidase is an important way to reduce the production of urate. In the study, in order to identify the potential natural products targeting XO, pharmacophore modeling was employed to filter databases. Here, two methods, p…

The methanol extract of Euonymus laxiflorus, Rubia lanceolata and Gardenia jasminoides inhibits xanthine oxidase and reduce serum uric acid level in rats.

Chinese herbal medicinal plants, Euonymus laxiflorus (EL), Rubia lanceolata (RL) and Gardenia jasminoides (GJ), have been used wildly to treat arthritis and gout in Taiwan for decades. To understand the beneficial effects of these three plants, their xanthine oxidase (XO) inhibitory activity in vitro and hypouricaemic activity in vivo were investigated. Our results suggested that methanol extracts were better than water extracts for inhibition of XO activity and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, except the water extract of GJ, which exhibited the strongest radic…

[6,8-Dimethyl-2-piperidinomethyl-2,3-dihydrothiazolo[2,3-f]xanthine: a new inductor of the monooxygenase system inhibits experimental intrahepatic cholestasis in rats].

6,8-Dimethyl-2-piperidinomethyl-2,3-dihydrothiazolo[2,3-f]xanthine, a new inductor of the monooxygenase system, inhibited the development of experimental intrahepatic cholestasis induced by alpha-naphthylisothiocyanate in rats. The drug stimulated the detoxicating function of liver, increased the survivability of rats, restored the level of microsomal protein and cytochrome P-450, decreased the cell-average erythrocyte fragility, and reduced the activity of cholestasis markers and the amount of TBA-active products in hepatocytes.
